ABSTRACT During pregnancy, the maternal liver undergoes crucial metabolic adaptations that remain incompletely understood, particularly under adverse dietary conditions. This study investigates whether a high‐cholesterol diet supplemented with Methionine during pregnancy acts as an aggravating factor for hepatic disorders and alterations in matrix metalloproteinases (MMP‐2 and MMP‐9) activities. Two successive pregnancies were performed in 29 rabbits divided into standard (SD) and cholesterol‐ methionine (CMD) diet groups. The females received their respective diet 15 days before mating and then during two successive pregnancies. Lipids and iron are evaluated in the liver. Phospholipids are analyzed using thin‐layer chromatography, and the metalloproteinase MMP‐2 and MMP‐9 activities are evaluated by zymography method. A topographic (Masson) and histochemical (Nile blue, and Perl's stain) staining are used for histological study. The accumulation of hepatic lipids concerned Total Cholesterol (TC) (324%) and phospholipids (50 times) with the involvement of Phosphatidyl ethanolamine (PE), Phosphatidyl serine (PS), Phosphatidylcholine, and cardiolipin, whereas Triglycerides (TG) decrease (20.47%). The iron content increased in the CMD female liver. The liver structure showed disorganization, fibrosis associated with MMP‐2 activity rise, and inflammatory cell infiltration. Our study shows the amplified effect of successive pregnancies on structural disorders (matrix remodeling) and metabolic disorders (iron overload, lipid accumulation) caused by a cholesterol‐methionine‐enriched diet.
Several studies have shown that hyperhomocysteinemia (Hhcy), a risk factor for cardiovascular diseases, is linked to other pathologies, including the aggravation of renal lesions in humans and some animal models. To our knowledge,studiesno have been conducted on the potential impact of Hhcy on renal fibrosis and tissue remodeling in sandratsPsammomys ( obesus). The aim of this study was to analyze renal histopathology as well as the immunoexpression of some fibrosis markers during Hhcy inrat.sand Hhcy was induced in this species by intraperitoneal administration of methionine (150 mg/kg body weight/day) for three Themonths. kidney removed at the end of the experiment was subjected to histomorphometric (Masson's trichrome staining, Sirius red) immunohistochemical (collagen I and III,alpha-SMA, MMP-2 and-9) studies. Our results showed that the kidneyofPsammomys Hhcy is the site of significant interstitial fibrosis, leukocytic infiltration, and disorganization of tubular and corpuscular structures. Immunohistochemistry revealed the involvement of type I and III fibrillar collagen in the development of fibrosis. Strongcytoplasmic immunoexpression of collagen I and III in inflammatory cells and alpha-SMA in the renal interstitium indicated the involvement of both leukocytes and myofibroblasts in the fibrosis process. Furthermore, we observed a decrease in MMP-2 and-9 immunoexpression tubular cells and an increase in interstitial tissue and inflammatory cells. In conclusion, the kidneysofPsammomys obesusexhibited fibrosis and tissue remodeling secondary to Hhcy.

However, the post-transplant condition and graft quality depend on several factors, including anemia, which is a major complication in transplant recipients. Objective. To evaluate the prevalence of anemia in patients transplanted from living donors and to analyze all factors associated with this complication. Population and Methods. This was a single-center, 3-year follow-up study including 90 patients transplanted from living donors. Urea, creatinine (Cr), uric acid (AU), albumin, 24-hour proteinuria (Prot24h), and hemoglobin (Hb) levels were measured using recommended methods. Glomerular filtration rate (GFR) and body mass index (BMI) were calculated. Patients were randomized into two groups based on Hb levels: a group with Hb ≥ 12 g/dL and a group with Hb < 12 g/dL. Furthermore, these same parameters were studied according to the severity of anemia, considering levels ≤ 10 g/dL as severe anemia. Results. The prevalence of anemia in our patients represented 34.61%, of which 10.25% suffered from severe anemia. The latter is significantly higher in women compared to men (48.28% vs 26.54% p=0.04), as well as, in patients who returned to chronic dialysis compared to those who were stable on transplantation (60% vs 40%, NS). A a significant association was noted between mortality, return to dialysis, and anemia. Conclusion. Given the strong association of anemia with graft function and mortality, transplant recipients must benefit from close monitoring and correction of anemia as soon as it develops. [Introduction. Studies have shown an association between homocysteine (Hey) and several pathological conditions, including cardiovascular disease. However, the relationship between Hey and testicular disorders is still unclear. The present study investigated the effects of Hcy on testicular structure. Thirteen adult Wistar albino rats were divided into two groups. The control group and the methionine group received an oral solution of L-methionine 200 mg/kg per day for six months. Blood samples and testes were collected for subsequent experiments. Serum levels of Hcy and testosterone were analyzed. We examined the histopathological and immunohistochemical localization of GATA-4, vimentin (VIM), connexin-43 (Cx-43), occludin (OCLN), androgen receptor (AR), estrogen receptor 1 (ESR1), estrogen receptor 2 (ESR2) in the rat testis. Results showed that rats receiving methionine showed a reduction in body and testicular weights. Histological and immunohistochemical examination of the testicular tissues of methionine group showed altered structure of seminiferous tubules with decreased immunoexpression of Cx-43, GATA-4, OCLN, VIM, AR, ESRI and ESR2 compared to control. The results suggest that Hey has a negative effect on the testes with a probable disruption of spermatogenesis.
Methionine is an essential amino acid involved in critical metabolic process, and regulation of methionine through metabolism is important to supply this amino acid for cell needs. Elevation in plasma methionine commonly occurs due mutations in methionine-metabolizing enzymes, such as methionine adenosyltransferase. This elevation could cause hyperhomocysteinemia (HHcy), an independent risk factor of cardiovascular diseases, including atherosclerosis. Several lines evidences suggest that HHcy due to methionine supplementation can affect the matrix production by involving cell types, such vascular smooth muscle cells and fibroblasts. To this end, aortic adventitial fibroblasts primary cultures were exposed to methionine. Methionine at a concentration of 20 mM produced definite effects on the in vitro grown fibroblasts after 72 hours. The mostdent evi were retraction of cell processes, vacuolation and fragmentation of the cytoplasm, production of irregular contour of ustheadnucle nucleolus, decrease of cytoplasmic basophilia and the presence of pycnosis.
Nos travaux antérieurs ont montré qu'un régime alimentaire riche en méthionine est à l'origine d'une hyperhomocystéinémie (Hhcy), facteur de risque des maladies cardiovasculaires, conduisant à de nombreuses altérations tissulaires. L'objectif de notre étude est d'analyser certains éléments de la balance MMP/TIMP au niveau myocardique chez le rat des sables, Psammomys obesus (P. obesus) rendu hyperhomocystéinémique. L'induction de l'Hhcy chez le rat des sables est obtenue grâce à une injection intrapéritonéale de méthionine à la dose de 300 mg/kg de poids corporel/jour pendant 1 mois. Nous avons utilisé une approche histo-morphométrique afin d'étudier les altérations matricielles du myocarde (colorations au trichrome de Masson et au rouge Sirius). L'immunohistochimie nous a permis d'analyser les MMP-2 et -9 (Métalloprotéases matricielles) et leurs inhibiteurs tissulaires, les TIMP-1 et -2 dans le myocarde des P. obesus Contrôle et Hhcy. La quantification de ces marqueurs est réalisée grâce à la méthode de l'intensité réciproque. L'augmentation d'environ 875 % (p < 0,0001) de l'Hcy plasmatique enregistrée chez les rats des sables soumis à la méthionine en fin d'expérimentation, confirme l'installation de l'Hhcy. Le myocarde de P. obesus Hhcy est le siège d'une modulation de la composition biochimique de la matrice extracellulaire ou remodelage matriciel aboutissant à l'installation d'une fibrose interstitielle et périvasculaire. Nos résultats immunohistochimiques montrent que cette fibrose est associée à une variabilité d'immuno-expression des MMP-2 et -9 et de leurs inhibiteurs tissulaires (TIMP-1 et -2). En effet, chez P. obesus Hhcy, une diminution de 14,15 % (p < 0,0001) de la MMP-2 et une augmentation de 18,83 % (p < 0,0001) de la MMP-9 sont observées au niveau cardiomyocytaire et matriciel respectivement. L'inhibiteur tissulaire TIMP-1 diminue chez P. obesus Hhcy de 34,88 % (p < 0,0001). Un fort immunomarquage du TIMP-2 est observé chez le Rat des sables Hhcy alors qu'aucun immunomarquage de cet inhibiteur tissulaire n'est observé dans les cardiomyocytes des animaux Contrôle. La fibrose, marqueur du remodelage matriciel, est associée à une augmentation du rapport MMP-9/TIMP-1 et une diminution du rapport MMP-2/TIMP-2 chez les rats Hhcy vs rats Contrôle. Ces résultats mettent en évidence, l'implication de la balance MMPs/TIMPs dans le remodelage de la matrice extracellulaire myocardique secondaire à une Hhcy chez P. obesus.
Artemisia herba-alba (AHA) is a traditionally used plant to treat various diseases, including diabetes and metabolic dysfunctions. Plant extracts are generally explored empirically without a deeper assessment of their mechanism of action. Here, we describe a combinatorial study of biochemical, molecular, and bioinformatic (metabolite-protein pharmacology network) analyses to elucidate the mechanism of action of AHA and shed light on its multilevel effects in the treatment of diabetes-related advanced glycation end-products (AGE)-induced liver damages. The extract’s polyphenols and flavonoids content were measured and then identified via LC-Q-TOF–MS/MS. Active compounds were used to generate a metabolite-target interaction network via Swiss Target Prediction and other databases. The extract was tested for its antiglycation and aggregation properties. Next, THLE-2 liver cells were challenged with AGEs, and the mechanistic markers were measured [TNF-α, IL-6, nitric oxide, total antioxidant capacity, lipid peroxidation (LPO), and caspase 3]. Metabolite and network screening showed the involvement of AHA in diabetes, glycation, liver diseases, aging, and apoptosis. Experimental confirmation showed that AHA inhibited protein modification and AGE formation. Additionally, AHA reduced inflammatory mediators (IL-6, TNFα), oxidative stress markers (NO, LPO), and apoptosis (Caspase 3). On the other hand, cellular total antioxidant capacity was restored to normal levels. The combinatorial study showed that AHA regulates AGE-induced liver damages through MAPK-AKT and AGE-RAGE signaling pathways. This report highlights the combination of experimental and network pharmacology for the exact elucidation of AHA mechanism of action as a multitarget option in the therapy of diabetes and AGEs-related diseases.
Kidney transplantation is the best treatment received by an uremic patient. One of the major advantages of transplantation is restoring a hormonal profile as before the chronic kidney disease. However, the posttransplant state depends on several factors including the quality of the graft. In this study, we assessed the donor age in living donors-recipients as well as the exploration of their hormonal profile. This is a single-center follow-up study over a period of 3 years, including 90 kidney recipients transplanted by living donors. We performed blood measurements of parathyroid hormone, prolactin, follicle stimulating hormone, luteinizing hormone, testosterone, estradiol, blood glucose, urea, creatinine, total cholesterol, triglycerides, high density lipoprotein-cholesterol, uric acid, albumin, and 24-h proteinuria. Glomerular filtration rate, body mass index, and low-density cholesterol were calculated. To analyze the donor age effect on recipients, the patients were divided into two groups according to donors age (<40 years and ≥40 years). The hormonal profile was normal for almost the majority of patients. In addition, we noted a return to dialysis associated with certain metabolic abnormalities and a donor age >40 years. Exploring the hormonal profile of the recipient is recommended. The donor age significantly influences the recipient fate but not their hormonal profile.
INTRODUCTIONCardiovascular diseases were defined as coronary artery, cerebrovascular, or peripheral arterial disease.Hyperhomocysteinemia (Hhcy) is an independent risk factor of cardiovascular diseases, including atherosclerosis. Our previous studies demonstrated the involvement of Hhcy in cardiovascular remodeling in the sand rat Psammomys obesus.MATERIAL AND METHODSAn experimental Hhcy was induced, in the sand rat Psammomys obesus, by a daily intraperitoneal injection of 70 mg/kg of methionine for a total duration of 6 months. The impact of Hhcy on the cellular and matrix structures of the heart, aorta and liver was analyzed using histological techniques. Additionally we treated primary cultures of aortic smooth muscle cells (SMCs) with high concentration of methionine to investigate the effects of methionine at the cellular level.RESULTSA moderate Hhcy induced a significant increase in the extracellular matrix components particularly collagens which accumulated in the interstitial and perivascular spaces in the studied organs indicating a developing fibrosis. A liver steatosis was also observed following methionine treatment. Further analysis of the aorta showed that Hhcy also induced vascular alterations including SMCs reorientation and proliferation associated with aneurysm formation.CONCLUSIONSOur results show for the first time that Hhcy can induce a cardiovascular and liver diseases phenotype in Psammomys obesus, a species previously shown to be a good model for the studies of diabetes and other metabolism-related pathologies.
INTRODUCTION The chronicity of advanced glycation end-products (AGEs) imparts various damages resulting in metabolic dysfunction and diseases involving inflammation and oxidative stress. The use of plant extracts is of high interest in complementary medicine. Yet, extracts are multicomponent mixtures, and difficult to pinpoint their exact mechanism. OBJECTIVES We hypothesise that network pharmacology and bioinformatics can help experimental findings depict the exact active components and mechanism of action by which they induce their effects. Additionally, the toxicity and variability can be lowered and standardised with proper encapsulation methods. METHODOLOGY Here, we propose the formulation of phytoniosomes encapsulating two Artemisia species (Artemisia dracunculus and Artemisia absinthium) to mitigate AGEs and their induced cell redox dysregulation in the liver. Extracts from different solvents were identified via liquid chromatography quadrupole time-of-flight mass spectrometry (LC-Q-TOF-MS/MS). Phytoniosomes were explored for their anti-glycating effect and modulation of AGE-induced damages in THLE-2 liver cells. Network pharmacology tools were used to identify possible targets and signalling pathways implicated. RESULTS Data demonstrated that A. absinthium phytoniosomes had a significant anti-AGE effect comparable to reference molecules and higher than A. dracunculus. They were able to restore cell dysfunction through the restoration of tumour necrosis alpha (TNF-α), interleukin 6 (IL-6), nitric oxide, and total antioxidant capacity. Phytoniosomes were able to protect cells from apoptosis by decreasing caspase 3 activity. Network pharmacology and bioinformatic analysis confirmed the induction of the effect via Akt-PI3K-MAPK and AGE-RAGE signalling pathways through quercetin and luteolin actions. CONCLUSION The current report highlights the potential of Artemisia phytoniosomes as strong contenders in AGE-related disease therapy.
Plusieurs études ont montré que les taux plasmatiques en homocystéine (Hcy) supérieurs à 14 μMol/L, indépendamment des facteurs de risques dits traditionnels, sont impliqués dans des désordres et dysfonctionnements du système vasculaire. C’est pourquoi développer une démarche thérapeutique visant à normaliser ou réduire l’Hcy est un enjeu de santé publique. Le but de ce travail est d’analyser l’impact d’une surcharge en méthionine sur certaines enzymes du cycle de la méthionine et du stress oxydatif et sa répercussion sur le remodelage du tissu cardiaque. Des rats Wistar mâles sont divisés en 05 groupes, lot 1(NaCl), Lot 2 (Huile d’olive), lot 3 (05 mg de lycopène/Kg de PC, lot 4 (500 mg de méthionine/Kg de P.C/J), et le lot 5 (05 mg de lycopène et 500 mg de méthionine/Kg de P.C/J). La méthionine et le lycopène sont administrés par voie orale tous les jours pendant trois mois. La pesée corporelle, le dosage de l’Hcy, de certains enzymes (CBS, SAHH, PON1, MDA et la MPO) sont réalisés. Au terme de l’expérimentation, nous avons procédé à une analyse histologique du tissu cardiaque avec une attention particulière à la matrice extracellulaire (MEC). L’administration chronique de la méthionine a révélé une augmentation significative de l’Hcy, du MDA et de la MPO et une diminution de la SAHH de la CBS et de la PON1. Le traitement préventif par le lycopène a restauré significativement l’équilibre biochimique des activités des enzymes étudiés. Au niveau tissulaire, nous avons noté la présence de diverses altérations touchant aussi bien les cellules que la matrice extracellulaire cardiaque. Cette étude suggère que les altérations biochimiques et tissulaires engendrées par l’installation d’un état d’hyperhomocystéinémie, sont réduites par la supplémentation en lycopène, lequel exerce son effet protecteur grâce à ses propriétés anti-oxydantes, anti-agrégantes plaquettaires, et modulateur du stress oxydatif.
L’Hyperhomocystéinémie (Hhcy), facteur de risque associé à différents états pathologiques en particulier vasculaires, impacte négativement sur la biochimie de la matrice extracellulaire (MEC). L’objectif de ce travail est l’étude de l’impact de Hhcy sur l’intégrité testiculaire par l’analyse biochimique de la MEC d’une part et de l’occludine d’autre part, chez le Rat des sables, Psammomys obesus (Po). L’étude est menée chez 19 Po mâles répartis en deux groupes : un groupe Témoin nourris à l’aide de leur régime naturel à base de plantes halophiles ; le groupe Expérimenté recevant le même régime alimentaire supplémenté en méthionine (200 mg/kg de poids corporel/jour) pendant 3 mois. L’homocystéinémie plasmatique est déterminée par la méthode FPIA. Les altérations tissulaires sont analysées via une étude histologique, histochimique (collagènes fibrillaires, des glycoprotéines et des protéoglycanes) complétée par une approche immunohistochimique de l’occludine. Nos résultats ont mis en évidence l’installation d’un état d’Hhcy associé à des altérations matricielles caractérisées essentiellement par une importante accumulation de collagènes conduisant à une fibrose et de faibles modifications des glycoprotéines et protéoglycanes. Le testicule est le siège d’altérations structurales (épaississement de la vitrée, l’albuginée, présence d’espaces blancs entre les cellules germinales…). Ces dernières laisseraient penser à un phénomène d’apoptose. Nous avons, également mis en évidence, une altération de l’expression de l’occludine (protéine des jonctions serrées) chez les rats hyperhomocystéinémiques. L’hyperhomocystéinémie expérimentale est à l’origine d’altérations structurales et matricielles. La déstabilisation de l’occludine laisserait supposer l’altération de l’intégrité de la barrière hémato-testiculaire.

Also, Hhcy induces apoptosis of some cardiomyocytes and cardiac fibroblasts by increasing of activated caspase 3 expression. These results highlight a remodeling of cardiac tissue in hyperhomocysteinemic Psammomys obesus.
Dans un précédent travail, nous avons montré que l’hyperhomocystéinémie (Hhcy), facteur indépendant de risque cardiométabolique est associée à des altérations structurales (matricielles, cellulaires et vasculaires) des vésicules séminales du rat des sables ou Psammomys obesus (Po). Notre travail a pour objectif d’analyser la relation Hhcy-régulation endocrinienne, par les œstrogènes, de la vésicule séminale de P. obesus en saison de reproduction. Pour ce faire, 19 Po, sont répartis en deux lots, un lot témoin constitué de 9 rats et un lot comportant 10 rats soumis à une administration de méthionine à raison de 200 mg/kg de poids corporel/jour pendant 3 mois. L’homocystéinémie est déterminée mensuellement, par des dosages plasmatiques par méthode FPIA. Par ailleurs, la régulation hormonale de la vésicule par les œstrogènes a été abordée par l’immunolocalisation de leurs récepteurs (ERα et ERβ) par méthode immunohistochimique indirecte. L’administration de la méthionine a engendré chez le rat des sables, un état d’hyperhomocystéinémie. L’étude immunohistochimique a montré chez le rat des sables témoin, que la vésicule séminale exprime les récepteurs ERα et ERβ au niveau des cellules épithéliales et cellules musculaires. Chez le rat des sables hyperhomocystéinémique, nous avons enregistré une immunolocalisation similaire des récepteurs ERα et ERβ. Cependant nous avons mis en évidence une différence d’expression. Chez P. obesus, l’hyperhomocystéinémie semble influencer l’expression des récepteurs aux œstrogènes. Nos résultats laissent à penser qu’il existerait une relation Hhcy-fertilité.
We study the effect of an enriched cholesterol-methionine diet administered to females on the cardiac tissue remodelling of the offspring during two successive pregnancies. Two groups are constituted, standard diet (SD) group fed a standard diet and CD group fed a combined diet (standard + cholesterol 1%-methionine 0.25%). The diet is administered during 80 days. The results show changes in serum and cardiac parameters of CD newborn, with the involvement of phospholipids (PLs) (phosphatidylethanolamine (PE) and phosphatidylcholine (PC), variations in malondialdehyde (MDA), conjugated diene (CD), and vitamin C [VIT-C] rates). Under the CD effect, serum matrix metalloproteinase (MMP)-2, pro-MMP-9, and MMP-9 activities change. As to cardiac MMP-2 activity, a rise is noticed in the second pregnancy. Histological analysis reveals constricted blood capillaries, collagen fibre deposits, and lipid accumulation in the CD newborn heart. Our study shows the amplified effect of the maternal cholesterol-methionine diet in the second pregnancy on newborn cardiac disorders (matrix remodelling, oxidative stress, and lipid accumulation).
