This study aimed to compare the effects of bone morphogenetic protein BMP2/7 heterodimer and BMP homodimers on bone regeneration in bone defects model. Identical peri-implant bone defects model were created using proper controls on the frontal skull in 18 minipigs. Collagen sponges with low-dose (30 ng/mL) BMP2/7 heterodimer, BMP2 or BMP7 homodimer were filled in the defects. New bone formation and the expression of type I collagen (Col1), alkaline phosphatase (ALP) and osteocalcin (OCN) were evaluated after 2, 3, and 6 weeks of implantation. BMP2/7 resulted in significantly higher new bone areas percentage in the defect region than BMP2 and BMP7 (p<0.05). Immunohistochemical staining of Col1, ALP and OCN was stronger in BMP2/7 group than BMP2, BMP7 and control group (p<0.05). These results demonstrate that BMP2/7 heterodimer is a stronger inducer of osteoblastogenesis and could be applied at low dose to reduce the cost and side effects of BMP homodimers.
The aim of this study was to investigate the postnatal growth changes in the condyle and disc of the rabbit craniomandibular joint (CMJ). Forty-eight rabbits from newborn to an age of 120 days were divided into eight groups, and chondrocytic differentiation and function were evaluated within the CMJ by in situ hybridization of type II collagen and aggrecan mRNA. The morphology of the posterior band and the bilaminar zone were similar in the newborn group and were composed primarily of mesenchymal cells and capillaries. After weaning, mastication loading induced the differentiation of mesenchymal cells, which was accompanied by structural differentiation between the posterior band and the bilaminar zone. Aggrecan first appeared in the posterior band of the disc at 30 days postnatally, when the rabbits began to masticate solid food. Type II collagen emerged in the disc at the age of 45 days. Both genes coexpressed in the deeper half of the proliferative layer, the whole hypertrophic layer, and the mineralized layer of the condylar cartilage and staining intensity increased with age. The coexpression of aggrecan and Type II collagen indicates the maturation of chondrocyte differentiation in the disc and condyle, which contributes to the biomechanical characteristics of the CMJ that resist functional stimulation.
Knowledge of the protein interaction network is useful to assist molecular mechanism studies. Several major repositories have been established to collect and organize reported protein interactions. Many interactions have been reported in several model organisms, yet a very limited number of plant interactions can thus far be found in these major databases. Computational identification of potential plant interactions, therefore, is desired to facilitate relevant research. In this work, we constructed a support vector machine model to predict potential Arabidopsis (Arabidopsis thaliana) protein interactions based on a variety of indirect evidence. In a 100-iteration bootstrap evaluation, the confidence of our predicted interactions was estimated to be 48.67%, and these interactions were expected to cover 29.02% of the entire interactome. The sensitivity of our model was validated with an independent evaluation data set consisting of newly reported interactions that did not overlap with the examples used in model training and testing. Results showed that our model successfully recognized 28.91% of the new interactions, similar to its expected sensitivity (29.02%). Applying this model to all possible Arabidopsis protein pairs resulted in 224,206 potential interactions, which is the largest and most accurate set of predicted Arabidopsis interactions at present. In order to facilitate the use of our results, we present the Predicted Arabidopsis Interactome Resource, with detailed annotations and more specific per interaction confidence measurements.
