The focus of Prof Falasca’s research in the past 25 year involved the investigation of signalling pathways regulating intracellular physiological and pathological processes. Prof Falasca work is mostly focused on intracellular signals regulated by specific lipids that act as “second messengers” inside a cell to control a plethora of cellular functions, including cell growth, proliferation and metabolism. Attention is specifically focused on lipids known as “phosphoinositides” that can themselves act as or be converted into messengers, ultimately regulating several cellular functions. His early work identified for the first time the biological activity and signalling properties of the lysophospholipid lysophosphatidylinositol (LPI). Subsequently, he focused his attention on the role of phosphoinositide 3-kinase (PI3K) isoforms and phospholipase Cgamma-1 (PLCg1). Deregulation of these pathways is associated with several diseases including cancer and diabetes. He was among the very first to demonstrate the ability of pleckstrin homology domains to bind phosphoinositides. He also identified a novel signalling role for the phosphoinositide phosphatidylinositol-3-monophosphate (PI3P). In parallel he identified class II PI3K as the enzymes responsible for the production of PI3P unravelling a novel functional role for this class of enzymes. Prof Falasca’s research group is currently focusing primarily on LPI and PI3K. Major achievements have been the identification of a novel signalling pathway important for insulin action in muscles and adipocytes and the identification of the critical role of the enzyme PLCg1 in metastasis development and progression.
