Recent work using knockout mice has shown that Sepp1 transports selenium from the liver to cells throughout the body where it is taken up for its selenium by apoER2, a member of the low-density lipoprotein receptor family. Shortened forms of Sepp1 are filtered by the renal glomeruli and taken up from the urinary filtrate via the receptor megalin into the proximal convoluted tubule cells. Several projects are active in the lab, including 1) the characterization of Sepp1 (including metabolized forms of it) and elucidation of its function using Sepp1 conditional knockout mice and 2) the metabolism and function of extracellular glutathione and glutathione peroxidase (Gpx3) using Gpx3 knockout mice. The lab also carries out clinical studies of selenium metabolism.
