INTRODUCTION:There is a need for safe and potent antibiofilm agents for root canal irrigation. The aim of this study was to investigate the antibiofilm activity of enzymatic irrigants (Bromelain and Papain) and their cytocompatibility to mammalian cells. METHODS:Biofilms of Enterococcus faecalis were developed on dentin discs and treated with 0.05% bromelain, 4% papain, 2.5% sodium hypochlorite (NaOCl) or saline for 15 min. The antibiofilm effectiveness was characterized by quantifying the colony forming units (CFU) and confocal microscopic examination of the bacterial cells (live/dead staining) and extracellular matrix polysaccharide using a selective fluorophore binding assay. Cytotoxicity of 0.05% bromelain, 4% papain and 2.5% NaOCl to human periodontal ligament fibroblasts was investigated using a metabolic activity assay. Data were analyzed using analysis of variance and nonparametric tests, with significance set at P < .05. RESULTS:Papain (4%) and Bromelain (0.05%) demonstrated 5 log CFU reduction, which was comparable to 2.5% NaOCl (P > .05). Confocal microscopy further confirmed that bromelain, sodium hypochlorite, and papain caused significantly greater bacterial cell death compared to saline (P < .05). Papain significantly reduced the extracellular matrix polysaccharide content within the biofilm compared to all other groups (P < .05). CONCLUSION:This study demonstrated that bromelain and papain showed comparable antibiofilm activity as 2.5% NaOCl and were more biocompatible than NaOCl, highlighting their potential to be developed as root canal irrigants.
The aim of this study was to conduct a comparative assessment of the length (L), width (W), height (H), and sagittal angle (SA) of the occipital condyle (OC) with respect to age and sex estimation via cone beam computed tomography among the population of Dakshina Kannada district. A retrospective analysis was conducted on full-field-of-view (FOV) cone beam computed tomography (CBCT) images obtained for diagnostic and treatment purposes from department archives. The mean values of OC L, W, H, and SA on both the right and left sides were measured. The intraclass correlation coefficient was used to determine the inter- and intraobserver reliability. All the recorded variables were tested for differences with respect to the different age groups (20–35 years, 36–50 years, 51–65 years, and 66–80 years) via the F test and Welch’s ANOVA (W test). Post hoc analysis was conducted to compare the mean variables within the different age groups. The variables were then compared on the basis of sex via independent t tests. Stepwise discriminant function analysis was used to discriminate the sex of the participants. A p value < 0.05 was considered statistically significant. A total of 200 scans of which 99 were from males and 101 were from females were included. Furthermore, 50 scans each from four age groups were included. With respect to sex, comparison revealed that the R OCL and L OCL were significantly greater in males, with a p value of < 0.001. The R OCW was significantly greater among males, with a p value of 0.005. A comparison of R OCH and L OCH between the two groups revealed that both variables were greater in males, with a p value of < 0.001. No significant difference was found among the parameters related to age. The discriminant functional analysis revealed an overall accuracy of 95.50
Calcium hydroxide has been widely used in endodontics as an intracanal medicament due to its effective antimicrobial activity. Unfortunately, it does not destroy microbiota present in the dentinal tubules. Nanoparticle-loaded Ca(OH)2 has proven to enter the tubules and exhibit excellent antibacterial efficacy against various bacterial pathogens. To compare the antimicrobial efficacy of Ca(OH)2, loaded with Graphene(GNP) and Quercetin nanoparticles (QNP) as intracanal medicaments, using two different vehicles—propylene glycol(PG) and distilled water (DW)—against E. faecalis, using Confocal Laser Scanning Microscopy. Sixty decoronated single-rooted teeth were instrumented using 2.5
