Higher noise levels in computed tomography (CT) pictures make it hard to effectively identify lung disease because they mask important clinical indicators and sensitive anatomical details. This paper presents a self-supervised learning approach for precise reconstruction and objective quality assessment of noisy lung CT images, obviating the necessity for explicit ground-truth references. The suggested method effectively removes noise while keeping important diagnostic information by exploiting internal data consistency to directly recover intrinsic picture representations from corrupted data. A quantitative analysis of the input and processed CT images indicates significant enhancements in statistical characteristics and visual quality. The histogram spread goes from 172 to 200, which means that the contrast is better spread out. The entropy goes from 6.12 to 6.57, which means that the information is richer. The average brightness went up from 63.27 to 78.27, which means that the light was better. A PSNR of 21.81 dB and an SSIM of 0.759 show that the anatomical parts have been kept, which shows that the reconstruction quality is constant. Heatmaps that show pixel-by-pixel absolute changes can help to find areas of improvement more clearly without causing structural problems. The suggested self-supervised architecture provides a scalable and dependable way to improve CT scans in clinical settings when it comes to diagnosing lung diseases, where data availability and annotation problems still exist. This technique makes it easier to make decisions and analyze radiological data, and it works well with automation.
Endodontically treated teeth are highly susceptible to fracture due to loss of structural integrity. Therefore, it is of interest to compared fracture resistance and failure patterns among cast metal, prefabricated metal and fiber post-and-core systems under standardized conditions. Thirty mandibular premolars were endodontically treated, restored and subjected to oblique compressive loading at 135°C until failure. Fiber posts showed the highest fracture resistance (615 ± 50 N) and predominantly favorable, repairable fractures (80%), whereas metal posts exhibited lower resistance and mainly unfavorable root fractures (p < 0.001). Thus, we show controlled biomechanical evidence supporting fiber posts as a superior restorative option for preserving root integrity.
Abstract Background: Type 2 diabetes mellitus (T2DM) is the most common underdiagnosed metabolic disease due to lack of recognizable symptoms in the early phase. T2DM often goes undetected for years, leading to high rates of complications and mortality. RNA signatures in saliva could serve as valid, noninvasive biomarker for the detection of T2DM with high sensitivity and specificity. Aim: To analyze the relative gene expression of salivary transcriptomic biomarkers SAT1 and PSMB2 in individuals with T2DM. Settings and Design: This case–control study included two groups: age- and sex-matched T2DM patients and healthy controls, with 50 participants in each group. Subjects and Methods: Unstimulated saliva samples were collected from all participants. Relative gene expression levels of salivary transcriptomic biomarkers, such as SAT1 and PSMB2 were analyzed using real-time polymerase chain reaction. The messenger RNA expression levels and fold changes were calculated using 2-ΔΔCT method. Statistical Analysis: Student’s t -test was applied to check the P value significance ( P < 0.05). Results: The SAT1 gene was found to be downregulated by 54%, with a fold change of 0.46 in T2DM compared to controls, while PSMB2 was downregulated in T2DM when compared to healthy controls. Among the two biomarkers, PSMB2 showed a statistically significant difference ( P < 0.05), whereas SAT1 did not attain statistical significance. Conclusion: Our study demonstrated that analysis of the genetic expression of salivary transcriptomic biomarkers SAT1 and PSMB2 genes in the saliva of T2DM patients might help in the development of a noninvasive early diagnostic aid to facilitate early intervention of T2DM.
The wound healing is an innate physiological process that begins in response to tissue injury. Natural healing is crucial in bacterial infections, however, poor oxygenation and disruption in cellular remodeling and proliferation result in prolonged inflammatory response. Our current approach is to prepare a natural antiseptic wound-dressing for accelerating the healing process. In this study, Ovalbumin and Bovine Serum Albumin based hydrogels with Ajuga bracteosa extract was prepared in three different ratios. Ultraviolet-visible spectroscopy, Fourier-transform infrared spectroscopy and scanning electron microscopy were performed for initial characterization of the prepared ovalbumin and bovine serum albumin-based hydrogels. The antibacterial assay was performed against Pseudomonas aeruginosa, Staphylococcus aureus, and Escherichia coli to inspect antiseptics. To investigate the growth of cell, cell viability (MTT) was assessed using HaCaT cells treated over 48 h with 50 µL of each hydrogel formulation. In-silico docking analysis was performed to infer the active binding of the ligands from the plant extract and the albumins to further extend the study using active binding sites. The absorption spectrum of both the hydrogels has indicated shifts in the peaks (230 nm) formed by the mixtures that imply the interactions between plant extract and both the albumins. OVA-hydrogel has significantly inhibited the growth of the selected pathogens. Ovalbumin-hydrogel was significantly effective against the three pathogens at the concentration of 1:1 of the hydrogel composite ((P. aeruginosa 12.2 ± 0.2 mm), (S. aureus 17.7 ± 1.6 mm), and (E. coli 18.3 ± 0.4 mm)) compared to BSA hydrogel (8.1 ± 0.2 mm, 15.4 ± 0.4 mm, and 17.03 ± 0.1 mm respectively). Cytotoxicity assay (MTT) results in HaCaT cells treated with Ovalbumin-based hydrogel indicated the cells viability and accelerated wound closure (P < 0.0001****) compared to Bovine Serum Albumin based-hydrogel (P = 0.01**) at 48 h. the growth rate was found significantly faster in OVAH-treated cells (P < 0.0001****) relative to BSAH (P = 0.0128). Molecular docking analysis revealed that Deacetylajugarin IV and Ergosterol exhibited high binding affinities with bovine serum albumin (− 7.8 and − 9.5 kcal/mol) and moderate binding affinities with ovalbumin (− 6.2 and − 6.8 kcal/mol), suggesting effective ligand availability and active release at the wound site. Swiss-ADME analysis indicated the Deacetylajugarin IV meets the drug-likeness criteria and demonstrate favorable pharmacokinetic properties. The ovalbumin-based hydrogel demonstrated significant antibacterial efficacy and enhanced wound healing potential by promoting keratinocyte proliferation and tissue regeneration indicating its promising natural and effective antiseptic wound dressing.
The objective of the current research was intended to explore the eruption of primary teeth amidst children in Chennai, Tamil Nadu and evaluate the involvement of birth weight and term on eruption pattern. The research was a cross-sectional oral health examination conducted from February 2023 to July 2023. The sampling was all primary health care centres in Chennai. Primary health care centres were arbitrarily chosen using systematic random sampling procedure. Children aged 3 - 36 months were enlisted by simple random sampling method from immunization units of primary health care centres in Chennai. 1700 children were assessed out of which 828 were boys and 853 were girls. The infant’s birth weight and gestational age were designated utilising the pertinent World Health Organization calibre. Oral examination was performed visually to determine the count of type of erupted teeth using a dental mirror under good illumination. A tooth was contemplated to be erupted whenever any portion of the crown had pierced the gingiva and furthermore noticeable intraorally. Statistical Package for Social Sciences software version 25 was used to analyse the data. Data normality was found to be non-normal using Kolmogorov - Smirnov test. Descriptive demographics was demonstrated using mean, standard deviation, frequency and percentage. Kruskal Wallis trial was utilized for inter group comparison between birth weight and eruption, and chi-square test for comparison between term of birth and eruption. Association of birth weight and eruption of teeth is significant in almost every tooth except maxillary lateral incisors, maxillary first molars and mandibular central incisors. Term of month, i.e. full term and pre term birth are significantly associated with all the erupted teeth. Term of birth and birth weight was significantly and negatively associated with eruption of teeth in primary dentition. This study also puts forward that a notable 2-3 months delayed eruption of almost all the primary dentition is seen in Chennai compared to standard chronology.
