Summary That the earliest believers in Jesus as the Messiah and Savior were entirely composed of people from a Jewish background is not questioned. Even when this “Christian” message went out to the Gentiles, for many years, the majority of these believers in Jesus were Jewish. The New Testament and other historians trace this early Jewish movement in a fairly detailed way until the phenomenon of “Jewish Christianity” began to fade in the 4th and 5th centuries ce. What were some of the distinct features of this movement and how was it shaped both by internal factors, as well as external circumstances? An important question is how the traditional Jewish rabbinic leaders responded to these Jewish Christians. Oftentimes, the dynamic relationship is described by the expression “The Parting of Ways.” The traditional answer to this question is that this final separation took place by the events surrounding the First Jewish War of 66 to 73 ce. A variation to this date for a “parting” extends the history and 21st centuries a significant movement of scholars have argued that there was no real “parting of the ways” until the Constantinian revolution in the mid-4th century. The existence of such sects as the Nazarenes and the Ebionites, as well as documents like the Pseudo-Clementine Recognitions, provides evidence of a continuing dynamic relationship between “Jewish Christians” and the “Jewish leadership.” While these features are undeniable, the idea of no significant “parting of the way” seems to contradict some very strong evidences from the first two centuries. The relationship between these two “Jewish movements” simply was irreconcilable due to features of Jewish Christianity that could not be accepted by the Jewish leadership that survived the Jewish Wars in the initial century of the Jewish Christian movement.
We present a simple and efficient way to add cis dialdehydes, phosphoryl groups, or cis diols to the 3 prime end of oligonucleotides using periodate oxidized nucleotides (oNTPs) and terminal deoxynucleotidyl transferase (TdT). The 3 prime end cis dialdehyde-modified oligos are generated by incubating TdT with an oligo for several minutes followed by addition of a oNTP and incubated at 30 degrees C for 30 minutes to an hour. After allowing the addition of the cis dialdehydes, heating the reaction mixture at 90 to 95 degrees C for 10 minutes yields oligonucleotides with 3 prime phosphoryl groups. The 3 prime cis diol modified oligos are synthesized by starting with 3 prime cis diol nucleotides (HO-NTPs). The cis dialdehyde modified oligonucleotides and cis diols may then be used for a variety of investigations such as studying the interaction of proteins with the 3 prime end of DNA, and possibly RNA. As an example, we demonstrate the efficacy of using an oligonucleotide modified with oGMP at the 3 prime end as an affinity label for TdT and identified a peptide fragment that has been shown to contain two of three aspartate residues found to be in the TdT active site.
The Islamic Centre, also known as the Institutional Mosque among members of Higher Learning Institutions, serves the same role as a mosque—primarily as a place to perform prayers. However, beyond this, the Islamic Centre should also serve as a hub for the dissemination and delivery of dakwah (Islamic outreach) among the campus community and surrounding society. The study was conducted to identify the challenges and solutions closely related to ensuring that the dissemination of da’wah can be effectively implemented in a university. This study is to ensure that the role of Islamic Centers in the dissemination of dakwah is appropriate according to current conditions. Every program or activity organized should be tailored to suit the target audience or mad’u, so that the objectives of the dakwah can have a positive and meaningful impact. Furthermore, the method of delivering dakwah should be carefully considered to ensure that the knowledge shared resonates effectively with the public.. The use of modern technology in Islamic Centres can enhance the image of religious institutions so they are not left behind in the era of modernity digitalization and the current Fourth Industrial Revolution (IR 4.0). In addition, social media channels that are widely used and popular should be studied and utilized so that the Islamic message can reach its intended audience. Dakwah practitioners must learn and understand the use of modern technologies and social media platforms to align with current trends in continuing their dakwah efforts for ongoing development. The role of Islamic Centre administration is also crucial in ensuring that the goals and objectives of making the Islamic Centre the heartbeat of Islamic dakwah dissemination can be successfully realized. Therefore, such a study is suitable to be conducted using a qualitative method
