First study to characterize the proteomic alterations induced by β-Hydroxy-β-Methylbutyrate in combination with Dexamethasone. Evaluation of β-Hydroxy-β-Methylbutyrate in a severe glucocorticoid-induced muscle atrophy model. Analysis focused on white (fast-twitch) skeletal muscle, which is preferentially affected by glucocorticoid-induced atrophy. Although some effects of glucocorticoids are necessary for metabolic responses during stress, chronic exposure to high levels of these hormones can cause side effects such as hyperglycemia, insulin resistance, and muscle atrophy. Elevated glucocorticoid levels are observed in several clinical conditions, and their exogenous administration, such as high-dose dexamethasone treatments, is included in many clinical protocols. In this context, the supplement β-Hydroxy-β-methylbutyrate (HMB) emerges as a promising approach to counteract side effects its anticatabolic action. Therefore, the aim of this study was to evaluate the protein profile in the muscles and liver of rats treated with HMB in response to a model of severe muscle atrophy induced by dexamethasone. A total of 24 male Wistar rats, 60 days old, were used and distributed into the following groups: (1) Placebo Experimental Group (PEG), n = 8, treated only with saline solution; (2) Dexamethasone Experimental Group (DEG), n = 8, treated with intraperitoneal Dexamethasone injection (1 mg/kg/day); and (3) Dexamethasone + HMB Experimental Group (DEHG), n = 8, treated with intraperitoneal Dexamethasone injection (1 mg/kg/day) and gavaged with HMB (0.3 g/kg/day). After 10 days of treatment, the animals were euthanized for collection of the Soleus, Extensor Digitorum Longus (EDL) muscles, and liver, followed by protein processing and identification. In the Soleus muscle, when comparing DEG vs. PEG, proteins such as Myosin heavy chain 2, Myosin-4, Myosin-6, and Myosin-7 showed decreased expression in DEG compared to PEG. Comparing PEG vs. DEHG also showed a decrease in myosin expression in DEHG, but only Myosin-6 and Myosin-7 were reduced among the identified isoforms. In the EDL muscle, both DEG and DEHG showed reduced levels of all identified myosin subtypes. However, when comparing DEG and DEHG, the only myosin isoform preserved in DEHG compared to DEG was Myosin heavy chain 2. In the liver, dexamethasone treatment impaired glucose uptake, altered the Cytochrome P450 system, and reduced mitochondrial function, as reflected by reduced ATP levels. In conclusion, dexamethasone induced the loss of contractile proteins (myosins) in both muscles. The HMB dosage used in this study partially prevented myosin loss in the Soleus muscle but did not inhibit myosin loss in the EDL muscle. In the liver, dexamethasone induced changes in several proteins related to glucose metabolism; however, HMB was unable to attenuate dexamethasone-induced hepatic glycogenolysis.
The purpose of this study was to analyze the occurrence of Toxoplasma gondii in the samples of ice cream made with açai (Euterpe oleracea) pulp. Fifty samples of ice cream made with açaí pulp, purchased from 11 municipalities in the state of São Paulo, were analyzed. Conventional polymerase chain reaction (PCR) was performed with the primers TOX4/TOX5, and nested PCR (nPCR) was carried out with the primers TgNP1/TgNP2. Of the 50 samples evaluated, 1 (2%) was positive according to nPCR. The molecular detection of T. gondii in the samples of ice cream made with açai pulp in this study serves as an alert of the need for continuing epidemiological vigilance to detect this zoonosis, whose occurrence is increasing due to the widespread and expanding consumption of food products made from açai pulp.
A crescente demanda por ambientes hospitalares que promovam o bem-estar e favoreçam a recuperação dos pacientes evidencia a necessidade de abordagens projetuais mais humanizadas. Nesse contexto, a biofilia destaca-se por integrar elementos naturais ao ambiente construído, mostrando-se especialmente relevante para pacientes oncológicos submetidos a tratamentos prolongados e a elevados níveis de estresse e desgaste emocional. O objetivo deste artigo é analisar, sob uma perspectiva teórica, a aplicação dos princípios biofílicos na arquitetura hospitalar e suas contribuições para a recuperação desses pacientes. A metodologia adotada consiste em um estudo teórico, fundamentado em uma revisão bibliográfica de produções científicas nacionais e internacionais que abordam a relação entre biofilia, arquitetura hospitalar e bem-estar em contextos terapêuticos. Os resultados apontam que a incorporação de elementos como iluminação natural, vegetação, ventilação adequada, vistas para áreas verdes e o uso de materiais naturais contribui para a redução da sensação de isolamento, promove conforto psicológico e favorece a estabilidade emocional dos pacientes, refletindo positivamente no processo de recuperação. Conclui-se que a biofilia constitui uma estratégia projetual relevante para a qualificação dos ambientes hospitalares, contribuindo para o bem-estar de pacientes oncológicos e para o desenvolvimento de espaços terapêuticos mais humanos e eficazes.
Este artigo fundamenta-se em um estudo de iniciação científica desenvolvido no curso de Arquitetura e Urbanismo do UNISAGRADO, com o objetivo de investigar a influência do Metaverso na arquitetura e a evolução tecnológica da representação gráfica. De caráter exploratório e descritivo, a pesquisa analisou como os meios de representação, historicamente responsáveis pela expressão das intenções projetuais, foram ampliados pelas ferramentas digitais, possibilitando visualizações mais precisas, validação espacial e simulações detalhadas. A partir do surgimento do Metaverso, concebido por Neal Stephenson em 1992 como um ambiente imersivo e interativo, observa-se uma redefinição dos processos projetuais por meio da integração entre realidade física e virtual. Nesse cenário, a realidade aumentada e a inteligência artificial ampliam as possibilidades de experimentação, exigindo, contudo, um uso crítico e equilibrado. Conclui-se que tais tecnologias enriquecem a prática arquitetônica, desde que não substituam a criatividade, a experiência e a visão artística do arquiteto, elementos centrais no desenvolvimento do projeto.
