目的 通过对献血者ABO血型正反定型不符标本的进一步检测,总结分析正反定型不符的原因,以保证临床输血的安全有效.方法 对87例无偿献血者ABO血型正反定型不符标本采用ABO血型正反定型、抗体筛选试验、吸收放散试验等血型血清学方法、并采用PCR-SSP和ABO基因测序方法辅助检测.结果 87例正反定型不符标本中抗体筛选阳性者36例(41.38%,36/87);血清抗体减弱21例(24.14%,21/87);ABO亚型13例(14.94%,13/87);冷凝集素17例(19.54%,17/87).结论 综合分析各项实验结果,当血型血清学方法无法确定时,应进行基因检测,确保每一位献血者血型鉴定的准确性,并根据不同的原因,采用合理的用血建议.
目的 探讨直接抗人球蛋白试验(DAT)阳性输血患者的疾病类型分布情况及DAT阳性对输血患者临床疗效的影响.方法 回顾性分析内蒙古自治区血液中心 2020 年 4 月至 2022 年 7 月收治的需行输注红细胞治疗的 116 例患者的临床资料,将其中 58 例DAT阴性患者分为A组,58例DAT阳性患者分为B组.记录DAT阳性输血患者疾病类型分布情况;比较两组患者的输血效果,输血前后血红蛋白(Hb)、红细胞计数(RBC)、红细胞压积(Hct)的变化,以及DAT阳性不同分型患者的输血治疗效果.结果 DAT阳性输血患者中分型为免疫球蛋白G阳性(IgG)最多,占比为 84.48%(49/58),疾病中血液病患者占比最高,占比为43.10%(25/58),其次是感染性疾病,占 25.86%(15/58);B组患者输血总有效率低于A组;输血前后B组患者Hb、RBC、Hct均低于A组,输血后B组Hb、RBC、Hct上升值低于A组(均P<0.05);单纯C3d阳性患者和IgG+C3d阳性患者的输血总有效率均为100.00%,高于单纯IgG阳性患者的 53.06%(P<0.05).结论 DAT阳性分型中,单独IgG最多,且需输血的患者主要集中于血液病,其次是感染性疾病,DAT阳性会影响患者的输血效率,3种分型患者的输血效率存在差异.
目的 在内蒙古呼和浩特地区无偿献血者人群中筛选稀有血型,并了解稀有血型的种类和频率.方法 随机对22 309名无偿献血者标本,采用尿素溶血试验筛选Jk(a-b-)表型;对1 766名O型蒙古族无偿献血者标本,用96孔微量板法和改良的抗球蛋白试验筛选Oh、i、Ge、Lub、GPA、Wrb、Rh等稀有血型,用试管法和经典的间接抗球蛋白试验方法进行确认.结果 在22 309名无偿献血者中共筛选到Jk(a-b-)血型1例,其分布频率为0.004 4%;在1 766名蒙古族献血人群中共筛出Lu(b-)2例,其分布频率为0.113%.结论 Jk(a-b-)分布频率低于广州(0.02%)和成都(0.022 1%),与上海(0.0041%)、长春(0.005%)接近,高于日本(0.002 2%);Lu(b-)分布频率存在地区和民族差异,与国内报道比较,低于广西侗族(0.363%)和广西壮族(1.259%),高于广东番禹地区汉族(0.048%)、新疆维吾尔族(0/3 335)和浙江丽水畲族(0/3 580).
Objective To understand the phenotype distribution of negative RhD blood donors in local region ,and to establish negative RhD phenotype database .Methods A total of 554 specimens screened firstly as negative RhD were confirmed by indirect antiglobulin test .Repeat donors were excluded ,the remaining specimens were 366 cases ,which were analyzed the phenotype of Rh blood .Results Distribution characteristics of negative RhD phenotype was ccdee (56 .28% ) > Ccdee (29 .51% ) > ccdEe (7 .38% )> CcdEe (3 .55% )> CCdee (3 .01% )> ccdEE (0 .27% ) ,there were no CcdEE ,CCdEe ,CCdEE detected .Conclusion The establishment of Negative RhD phenotype database is help for providing matching blood for persons with negative RhD blood or same antibody blood ,and meeting the emergency blood usage ,it′s important for scientific management and reasonable application of negative RhD blood ,and accumulate data for then negative RhD blood of the local area ,especially for Mongolian .
Rh血型系是红细胞血型系统中较为复杂的一类,以IgG型抗体为主。该血型在输血治疗和胎母引起的新生儿溶血病等方面有着很重要的临床意义。本中心曾在工作中遇到1例由Rh血型抗体伴抗-Jkb、抗-Fyb引起的配血困难,现报道如下。1病例简介患者,女,29岁,内蒙古包头人。13岁时查出患有再生障碍性贫血,14岁在包头输血2次。
Objective:To explore the function of CDK2,a stable expression of CDK2 SiRNA astrocytoma cell line was established.Method: The Eukaryotic expression vector PGenesil-1-CDK2,CKD2 specific RNA interference,was constructed based on the sequence from Genbank.The plasmid was sequenced and transfected to SHG44 cell line using oligofectamine.The stable transfectants were selected by G418.Results: The sequence of the construct was confirmed right.All stable transfectants had significant lower expression of CDK2 compared with the control clones.Conclusion: The PGenesil-1-CDK2-SHG44 stable transfectants were successfully established with the constructed PGenesil-1-CDK2vector.
Objective Eukaryotic expression vector of RNA interference specific for CyclinE was transfected to inhibit the expression of CyclinE in PC12 cell.It is critical to research the role that MPPs play in the nerve cells and provide valuable theoretical basis for the parkinsonism.Method 1.PC12 cell line was transfected with siRNA vectors above via oligofectamine,and green fluorescent protein contents was assayed by fluorescence microscopy.2.The auxodrome of transfection cell line was assayed.Result 1.eukaryotic expression vector of RNA interference specific for CyclinE was transfected into PC12 cells.2.Transfected PC 12cell line grows slow than others.Conclusion CyclinE siRNA can significantly inhibit proliferation of PC 12 cells.
