Objective:To investigate the correlation between expression of MMP-2 and TIMP-1 in the process of scald wound repairing.Methods:Backs of eight big rats were scalded into 20% TBSA deep partial thickness burn.Biopsies from wound skins of each rat harvested at 24PBHS and 4,7,14,20,29,35,62PBDS.The expressions of MMP-2 and TIMP-1 in the samples were detected immunohistochemically.MMP-2 and TIMP-1 were also detected in the skin of another 2 normal rats as normal contro1.Result:MMP-2 and TIMP-1 were hardly expressed in the normal skins of rats.After rats were scalded,MMP2 and TIMP-1 had different expressions in the process of scald wound repairing.MMP-2 and TIMP-1 began to express at 24 PBHS,and enhanced gradually,and peaked on 20PBDS,and continued for 15 days then reduced;but MMP-2 and TIMP-1 were still expressed up to wound healing.There were correlations between the expression of MMP-2 and the expression of TIMP-1.Conclusion:The expression levels of MMP-2 and TIMP-1 had certain laws during wound repairing,which were closely related to wound repairing and the process of scar tissue growing.
Objective:To analyze the CT and MRI features of liposarcoma of the extremities and trunk for improving the diagnosis of the disease.Methods:The CT and MRI findings of 12 cases of primary liposarcoma confirmed by operation and pathology were analyzed retrospectively.Seven cases were examined by CT,11 cases by MRI,and 6 cases by both CT and MRI.Results:Among the 12 cases,6 were well differentiated liposarcoma,2 sclerosing liposarcoma,3 myxoid iposarcoma,2 pleomorphic and 1 was dedifferentiated liposarcoma.Seven cases were irregularly margined and five cases regularly margined.The mean diameter of the lesions ranged from 6.1 to 19.9 cm.Well differentiated liposarcom contained a large amount of mature fat.CT and MRI showed fat tissue which was well defined,and homogeneous in density and signal intensity(SI).Myxoid liposarcoma consisted mainly of myxoid matrix,containing a few well differentiated proliferating lipoblasts;CT and MRI demonstrated no or little fat with irregular density and mildly inhomogeneous SI.Pleomorphic liposarcoma had no characteristic of fat tissue,and the adjacent structures were infiltrated and edematous with apparently inhomogeneous density and SI.Dedifferentiated liposarcoma had a pretty large volume with apparently inhomogeneous density and SI.The enhancement was highly intensified.Conclusions:CT and MRI can reflect the histological features of liposarcoma of the extremities and trunk,and demonstrate the exact boundaries of tumors,the actual area and surrounding tissues.The degree of histology differentiation and malignancy can be evaluated on the basis of CT and MRI findings of liposarcoma.
Objective To investigate the expression of survivin and its relationship with Caspase-3 and clinical factors in breast carcinoma.Methods Survivin and Caspase-3/CerbB-2/Ki67/p53/ER/PR expressions were detected by immuohistochemisitry in the normal breast tissues,14 cases),proliferative breast disease (epithelial hyperplasia,14 cases),breast preinvasive carcinoma (20 cases) and breast invasive carcinoma (52 cases).Results The positive rates of survivin expressin were 21.4%,35.7%,60.0% and 71.2% in the four groups,respectively(P0.05).Survivin expression was related to lymphnode metastasis(P0.05).The expressin of Caspase-3 was negatively correlated with the expression of survivin (P0.05).The Ki67/p53 indexes were significantly higher in survivin positive tumors (48.57±23.07)/(32.76±33.37) than those in the negative ones (21.30±20.50)/(15.43±24.44) (P0.05).Conclusion The overexpression of survivin may play an important role in the pathogenesis and progress of breast carcinoma by the way of inhibition of the active Caspase-3 and of the accelerating proliferation.Survivin may be served as a new target for apoptosis-inducing therapy for breast carcinoma.
Objective:To analyze the MRI features of liposarcoma of the extremities and to correlate with the degree of histopathologic differentiation,so that to improve its diagnostic accuracy.Methods:The MRI findings of 10 cases with primary liposarcoma proved by operation and pathology were analyzed retrospectively.The tumor size,morphology,margination,signal characteristic,and relationship of the tumor to adjacent structures were also assessed,and correlated them with surgical pathology.Results:This group of liposarcoma included five histological types(2 subtypes involved).There were 5 cases of well-differentiated liposarcoma,1 case of sclerosing liposarcoma,2 cases of myxoid liposarcoma,2 cases of pleomorphic,1 case of dedifferentiated liposarcoma.Well-differentiated liposarcom had a large amount of mature fat,relatively high signal lntensity(SI) on T1WI and T2WI,tumor was well defined homogeneous in SI,adjacent structures were not invaded readily.Myxoid liposarcoma consisted mainly of myxoid matrix,containing a few well-differentiated proliferating lipoblasts,demonstrating no or little signal on MRI characteristic for fat,mildly inhomogeneous.Plepmorpic liposarcoma was obviously hetergeneous,no signal characteristic for fat adjacent structures was infiltrated and edematous.Dedifferentiated liposarcoma had a pretty lager volume,SI had apparently inhomogeneous,which was highly enhanced.Conclusion:MR can reflect features of histology of liposarcoma of the extremities,demonstrate the exact boundaries of tumors,and assess the actual area and surrounding tissues.The degree of histology differentiation and malignancy can be evaluated on the basis of MRI findings of liposarcoma.
目的 探讨透明细胞肉瘤的临床病理特征、诊断要点及鉴别诊断.方法 对一例CCS进行临床病理分析,并进行组织学、免疫组织化学染色观察并结合文献回顾分析.结果 肿瘤由被纤维组织分割成巢或束状的胞质透亮的或淡伊红染的上皮样细胞和梭形细胞构成,核圆形或卵圆形,较空淡,可见明显核仁.免疫组化观察肿瘤表达Vimentin、S-100、HMB-45、melan-A.结论 CCS是好发于年轻人肢体末端的软组织肉瘤,预后不良,属于高度恶性.诊断应结合临床与病理形态,免疫组化在诊断和鉴别诊断中起重要作用.
Objective To investigate the composition and histogenesis of sclerosing hemangioma of the lung(SHL),in order to elevate the recognition of clinicopathological feathers of SHL.Method The clinicopathlogical and immunohistochemical feathers were reviewed in 15 cases of SHL.Results SHL has a characteristic architecture composed of round cell and surface cubic cell.It has a variety of tissular structure,mainly expression with four regions that contains solid regions,papillary regions,angiomatoid regions and sclerosing regions.The expression of round cells about EMA,TTF-1,vimentin is positive and the expression of round cells about NSE,SYN,ER,PR is positive partly.As far as EMA,TTF-1 and SP-B are concered,the expression of surface cubic cell is positive.Conclusion Both solid round cells and surface cubic cells are neoplastic.SHL is a tumor that probably originates from the epithelial cell of primary respiratory tract.
Objective: To investigate the expression of MMP-2 in the process of scald wound repair and the role of its synthesized inhibitor.Methods: Sixteen big mice were randomly divided into two groups and their backs were scalded into 20% TBSA deep partial thickness burn.The experimental group was given food which contained Captopril 0.5~1mg/kg/day.Biopsies from wound skin harvested at 24PBHS and 4,7,14,20,29,35,62PBDS.The expression of MMP-2 in the samples were detected immunohistochemically.MMP-2 was also detected in the skin of another 2 normal mice as normal control. Results: MMP-2 was not expressed in the normal skins of mouse.After mice were scalded,MMP-2 had different expression in the process of scald wound repair.MMP-2 began to express at 24 PBHS,and enhanced gradually,and peaked on 20PBDS,and continued for 15days then reduced;but MMP-2 was still expressed up to wound healing.The expression of MMP-2 in experimental group was apparently lower than that of control group,which was significantly lower than that in control group at 7,14,20,29PBDS(P0.05),and the healing time of wound in experimental group was earlier than that of control group. Conclusion: The expression level of MMP-2 had certain law during wound repair,the expression of MMP-2 was closely related to wound repair and the process of scar tissue growing;its synthesized inhibitor may inhibit its activity and accelerate wound healing.
目的 探讨乳腺原发性腺泡状软组织肉瘤(ASPS)的临床病理特征、诊断及鉴别诊断,提高对该肿瘤的认识.方法 对1例ASPS进行HE染色、光镜观察、免疫组化染色并结合文献回顾综合分析.结果 1例ASPS为13岁女性,病变位于右乳外上限胸大肌与胸小肌之间,大体上为部分有包膜境界较清楚的肿块.镜下肿瘤呈巢状或腺泡状排列,周围是富含血窦的间质.瘤细胞呈圆形或多边形,胞质丰富,内含丰富的嗜酸性颗粒及结晶体,核仁明显,核分裂像少见.PAS染色阳性,免疫组化:vim(+)、myoglobin(+)、NSE(-)、s-100(-)、HMB45(-)、SYN(-)、Des(-)、CK(-)、P53(-)、Actin(-)、EMA(-)、Ki67 15%(+).结论 ASPs为一种少见的软组织恶性肿瘤,其组织起源不明,确诊必须依靠病理检查,治疗以手术为主.
Objective:1.To investigate the feasibility to proceed in situ the detection of DNA after PCR in paraffin section;2.To investigate the origin and lineage of the H/R-S cells from HL.Methods:Detect the DNA of the H/R-S cells after in situ PCR in Hodgkin's Lymphoma from three samples of paraffin section by Bio-labeled heavy chain family-specific primers.Results:1.VH2,VH3 and FR3 gene rearrangements and VH1,VH3 gene rearrangements were respectively observed in two cases of nodular sclerosis HL(NSHL).2.VH2,VH3 and FR2a gene rearrangements were revealed in one case of mixed cellarity HL(MCHL).Positive signal was found in the nuclei of surrounding small lymphocyte.Conclusions:1.It was feasible and validable to proceed in situ the detection of DNA after PCR in paraffin section.2.The appearance of VH1,VH2,VH3,FR2a,FR3 gene rearrangements suggested that the H/R-S cells from classical HL(NSHL and MCHL)originated from B lineage cells at various stages of differentiation.FR1,FR2 and FR3 gene rearrangements were demonstrated in different H/R-S cells in the same section,which suggested that H/R-S cells of NSHL and MCHL might originate from different clone of B cell.The H/R-S cells and the surrounding background cells might originate from the same B cell clone.
