隐匿性丙型肝炎(OCI)是指在血清/血浆中检不出HCV RNA,而存在于肝细胞和外周血单个核细胞(PB-MC)中.OCI存在于普通人群和无偿献血者中及其传染性和经输血传播风险已得到证实.采供血机构的血液筛查无论是酶免法还是核酸检测,检测标本均为血浆/血清,无法检出PBMC中的HCV RNA,OCI已成为输血安全的潜在威胁,需要研发行之有效的检测和传播阻断技术.
In order to explore the effect of Gegenqinliantang and puerarin on lipopolysaccharide (LPS) leading mice intestines microvascular endothelial cells (MIMVECs) to secret NO and endothelin (ET-1),the Gegenqinliantang and puerarin accompanying with LPS were added into cell monolayers,respectively,to reduce the pathologic impact to MIMVECs.The secreting NO and ET-1 from the treated MIMVECs were detected by nitrate reduction method and ELISA.The results showed that the concentration of NO between the experimental group of Gegenqinliantang and control group of LPS were significant different (p<0.05) at 3hrs and 12hrs,the concentration of ET-1 between experimental group and control group had significant difference (p<0.05) during 3hrs and 12hrs,the concentration of ET-1 between the experimental group of puerarin and the control group of LPS were also significant different.The results showed that both Gegenqinliantang and puerarin were able to reduce the damage of MIMVECs induced by LPS.These data provided a base for further exploring the mechanism of Gegenqinliantang and its active ingredient hurt MIMVECs.
目的:评价多种临床量表在急性肺栓塞症(APE)诊断中的作用.方法:对2008年7月-2016年2月352例可疑PTE患者进行前瞻性调查,Wells、Geneva和修正Geneva等3种临床量表评估PTE发病的可能性.结果:3个量表对PTE预测的ROC曲线下面积分别为:Wells 0.733(95% CI 0.651~0.815),Geneva 0.724(95 %CI 0.639~0.809),修正Geneva 0.719(95%CI 0.633~0.805),差异无统计学意义(P>0.05).Wells量表一致性系数(κ=0.45)明显低于Geneva量表(κ=0.77)和修正Geneva量表(κ=0.82),差异有统计学意义(P<0.01).结论:修正Geneva量表简便、实用、准确,有助于急性肺栓塞的诊断,节约检查费用.
目的:探讨前脑钠肽(pro-BNP)在急性肺栓塞(APE)病情监测中的作用。方法根据心脏超声结果连续入选伴或不伴右室功能不全的APE确诊患者各20例,ELISA法测定pro-BNP,比较其与右心功能不全、严重不良事件发生率之间的关联。结果右心室功能不全患者的pro-BNP水平(187.4pg/ml±107.5pg/ml)明显高于无右心室功能不全组(51.5±45.4pg/ml)(F=27.1,P<0.01)。pro-BNP水平与低氧血症、低血压、心肺复苏和住院死亡等发生率密切相关。结论 pro-BNP在急性肺栓塞的病情评估、预后判断和疗效评估具有一定价值。
Objective To get the gene encoding extracellular domains of gB protein of B virus and analyze its expression in the eukaryocyte cell.Methods synthesizing gene fragment encoding extracellular domains of gB protein of B virus was by using synthesis gene, then digested with the restriction endonucleases BamHⅠand NotⅠand inserted into eukaryotic expressing vector pEGFP-N3.pEGFP-N3-GB合 was transfected into 293 cells.After protein extraction, the expression of gene was detcted by western blotting, and the cellular localization of the gene was analyzed by immunofluorescence and laser scanning confocal microscopy.Results pEGFP-N3-GB合were expressed in 293 cells and on the cell membrane.Conclusion eukaryotic expressing system can produce specific antigen recombination protein of B virus gB protein and express on the cell membrane.
目的:就慢性阻塞性肺疾病急性加重期下呼吸道病原菌感染及药敏进行分析。方法选取2008年1月~2013年1月我院收治的128例慢性阻塞性肺疾病急性加重期患者,本组全部患者都行痰标本采集,采集前应先用双氧水或清水漱口数次,然后再在痰培养标本盒内吐出无菌的痰液吐,送细菌室检验。结果本组患者共培养获得的病原菌有116株。26例(20.31%)是属于2种以上病原菌混合感染。其中铜绿假单胞菌位居首位。革兰氏阴性杆菌对亚胺培南最敏感,对氨苄西林耐药性最高;革兰氏阳性球菌对万古霉素最敏感对青霉素有较高耐药性。结论临床上应该加强药敏监测,开展药敏试验,结合不同地区的具体细菌耐药情况,进行联合用药,轮换使用,以此来大幅度减少耐药菌株。
AIM:To observe the curative effect and safety of Budesonide inhalation suspension(BIS)in maintenance therapy of patients with severe persistent asthma.METHODS:24patients with severe asthma were screened,the patients enrolled were treated with BIS 2 mg twice a day and salbutamol aerosol as need for 8weeks.The curative effect outcomes after 4treating weeks and 8treating weeks,including pulmonary function test(FEV1),asthma symptom scores,salbutamol aerosol use,as well as the side effects index at the end of study,such as urocortisol level,were collected and compared before study(baseline).RESULTS:21patients were successfully enrolled,20of which completed the treatment.The curative effect outcomes were improved compared with the baseline(P0.05),FEV1was improved by 12.7% and13.7%,daytime symptom scores were improved by 31.3% and 29.8%,night symptom scores were improved by 32.8% and 36.8%,times of Ventolin daily use were reduced by 39.1% and41.2% after 4 weeks treatment and 8 weeks treatment respectively.After 8 weeks treatment,urocortisol in all patients was normal range and no severe side effect occurred.CONCLUSION:BIS has good curative effect and safety in treating patients with severe persistent asthma as maintenance therapy.
Objective To assess the clinical effects of leukotriene receptor antagonist(LTA)-montelukast on acute exacerbation of chronic obstructive pulmonary disease(AECOPD) and its influence on pulmonary hypertension.Methods 80 patients with AECOPD were randomly divided into group A and group B.40 patients in group A were treated with the routine therapy for ten days.40 patients in group B were treated with the intervention of montelukast based on the routine therapy in the same time.The score of health-related quality of life,Pulmonary function,pulmonary hypertension were measured before and after treatment.Results Before the treatment,there were no significant diference between group A and group B(P0.05).After treatment for 10 days,The score of health-related quality of life and Pulmonary function were significantly improved,and the pulmonary hypertension were obviously decreased in both group A and group B,but all the indexes in group A were better than those in group B(P0.05).No obvious adverse efects were found in the period of treatment.Conclusion Montelukast Can relieve the acute exacerbation with COPD and improve the Pulmonary function of patients,and it can decline pulmonary artery pressure(PAP) through its direct and indirect effects.It plays an important role in the prevention and treatment of chronic cor pulmonale.
This study tries to product rat gloma cells hypox anthine-guaninep hosphoribosyl transferase deficiency lines by combined effects of N-methyl-N′-nitro-nitrosoguanidine,8-azaguanine,6-thioguanine and ultraviolet irradiation in order to establish rat pituitary gonadotropic hormone hybridoma cell lines in future,but wo are not able to gain the expectant glioma HGPRT-cell lines which can be sieved by hypoxan thine-aminop terinthymidine selective medium.The experiment indicate that the HGPRT gene of C6 cells is too stabile to be induced deficient on this experiment conditions.
Objective To investigate the relation of proteasome active center LMP7 subuint with the severity of oxidative stress.Methods First establish alcoholic fatty liver group,by means of alcohol intragastric treatment.Then divided them into two groups,one group continued alcohol intragastric administration,which caused alcoholic hepatitis.The other group was given equal NS.The both groups were killed after 4 weeks.MDA content in the plasma and liver tissue were determined by spectrophotometer.By HE staining,histological pathology was analized under light microscope.The expression of proteasome LMP7 subuint mRNA were examined by reverse transcription and real-time PCR.The content of LMP7 subunit protein were determined by Western Blot.Results The level of MDA content of serum and liver tissue homogenate in control group were 0.64±0.16 vs 5.43±0.14,the alcoholic fatty liver group level was 1.32±0.19 vs 12.18±0.78,the hepatitis group level was 1.56±0.1 vs 14.0±0.5,the NS group level was 1.23±0.26 vs 10.4±0.6.Compared with the control group,the LMP7 mRNA level of alcoholic fatty liver group was 36% of the control group level,The NS group level rised to 51%,and the hepatitis group level was the lowest,which level was only 26% of the control level.The difference coincided with the severity of the liver pathology.Western Blot result showed the control group level was 0.50±0.01;the alcoholic fatty group level was 0.39±0.02,the hepatitis group level was 0.30±0.04,and the NS group level was 0.38±0.02.Conclusions The proteasome LMP7 mRNA expression and protein content decreased in alcoholic liver group,and the descending degree was associated with the severity of the oxidative stress.It indicated that the proteasome LMP7 subunit was one of the protection factor which postponed the occurrence of alcoholic liver diseases.
