Objective:To research the expression and significance of MTA 1 in endometriosis.Methods:The expression levels of MAT1 in eutopic endometrium and ectopic endometrium of 30 cases with endometriosis and normal endometrium of 20 cases without endometriosis were detected by immunohistochemical method.Results:The positive rates of MTA1 in normal endometrium,ectopic endometrium,and eutopic endometrium were 30.0%(6/20),53.3%(16/30),and 73.3%(22/30),respectively,there was statistically significant difference in positive expression rate of MTA1 between eutopic endometrium and normal endometrium(P=0.002),there was no statistically significant difference in positive expression rate of MTA1 between normal endometrium and ectopic endometrium(P=0.103),there was no statistically significant difference in positive expression rate of MTA1 between eutopic endometrium and ectopic endometrium(P=0.108),there was no statistically significant difference in positive expression rates of MTA1 at proliferative phase and secretory phase among normal endometrium,ectopic endometrium,and eutopic endometrium(P>0.05).Conclusion:Over-expression of MTA1 in eutopic endometrium may play a ceratin role in occurrence and development of endometriosis,which provides a new clue for treatment and prognostic evaluation of endometriosis.
Objective:To investigate the reversal effect of CRM197 on drug-resistant ovarian cancer and its treatment for ovarian cancer by researching the inhibition of CRM197 for the xenografts of ovarian cancer in nude mice.Methods:The parental ovarian cancer cell A2780,cells resistant to paclitaxel A2780/Taxol and cisplatin-resistant cells A2780/DDP were used to establish the xenograft model in nude mice,the differentiation for the growth of xenograft tumors in the group which was injected with CRM197 and the negative control group was detected.Immunohistochemistry was used to test the expression of HB-EGF and P-gp in xenograft tumors of each team.Results:The expression of HB-EGF and P-gp were in different levels in each group.It was depressed obviously in the xenograft tumors which were injected CRM197.The difference was significant in statistics.Conclusion:The inhibition of CRM197 on the activation of the signal transduction pathway of heparin-binding epidermal growth factor inhibited the growth of the xenograft tumors of ovarian cancer and depressed the expression of HB-EGF and P-gp in xenograft tumors.
目的:通过检测人类软骨蛋白39(YKL-40)和核转录因子KappaB(NF—KB)在子宫内膜异位症组织中的表达,探讨二者与子宫内膜异位症的关系及二者的相关性。方法:用免疫组化二步法检测40例子宫内膜异位症(EMS)患者的异位内膜和在位内膜及40例子宫肌瘤患者子宫内膜(对照组)YKL-40和NF—KB的表达,并对检测结果进行统计学分析。结果:YKL-40、NF—KB在子宫内膜异位症患者的异位和在位内膜及对照组内膜中的表达率分别为67.5%、62.5%、35%及88.5%、57.5%、32.5%,差异有统计学意义(P〈0.01);在不同月经周期YKL-40表达无统计学意义;在位内膜和正常内膜NF-KB的表达分泌期高于增殖期,差异有统计学意义(P〈0.05);YKL-40和NF-KB在三种内膜中的表达具有正相关性,相关系数分别是0.305,0.267和0.457(P〈0.01)。结论:YKL-40和NF—KB在EMS发生发展中起重要的作用。
Objective: To investigate the expressions of Human cartilage glycoprotein-39(YKL-40) and Nuclear factor-kappaB in endometriosis,and to evaluate the relationship between the YKL-40 and NF-κ B expressions.Methods: Immunohistochemistry was used to examine the YKL-40 and NF-κ B protein expressions in eutopic(n=40) and ectopic(n=40) endometrium of Ems,as well as endometrium of hystromyoma(n=40).The results were analyzed statistically.Results: The positive rate of YKL-40 and NF-κ B expressions in ectopic,eutopic and normal endometrium respectively were 67.5%,62.5%,35% and 88.5%,57.5%,32.5%(P<0.01);NF-κ B expression was higher in secretory phase than that in proliferative phase in the eutopic and normal endometrium(P<0.05),but YKL-40 was not correlated with the phase of menstrual cycle;Furthemore,the expression of YKL-40 and NF-κ B protein was positively correlated in these three medometrial tissue(rs=0.305,0.247 and 0.467,P<0.01).Conclusion: The over expression of YKL-40 and NF-κ B may play an important role in the pathogenesis of endometriosis.
