Objective:To investigate the reversal effect of CRM197 on drug-resistant ovarian cancer and its treatment for ovarian cancer by researching the inhibition of CRM197 for the xenografts of ovarian cancer in nude mice.Methods:The parental ovarian cancer cell A2780,cells resistant to paclitaxel A2780/Taxol and cisplatin-resistant cells A2780/DDP were used to establish the xenograft model in nude mice,the differentiation for the growth of xenograft tumors in the group which was injected with CRM197 and the negative control group was detected.Immunohistochemistry was used to test the expression of HB-EGF and P-gp in xenograft tumors of each team.Results:The expression of HB-EGF and P-gp were in different levels in each group.It was depressed obviously in the xenograft tumors which were injected CRM197.The difference was significant in statistics.Conclusion:The inhibition of CRM197 on the activation of the signal transduction pathway of heparin-binding epidermal growth factor inhibited the growth of the xenograft tumors of ovarian cancer and depressed the expression of HB-EGF and P-gp in xenograft tumors.
YKL-40 is expressed and secreted by several types of cancer cells and inflammatory cells,and the exact biological function of YKL-40 is unknown,while studies indicate that it may play an important role in cancer cell proliferation,differentiation,protection against apoptosis,stimulation of angiogenesis and regulation of extracellular tissue remodeling.In recent years,studies have showed that YKL-40 is closely related to various gynecologic diseases such as ovarian cancer,cervical cancer,endometrial carcinoma and endometriosis.Here is to make a review on the structure characteristics,tissue source,biological functions of YKL-40 and the related research with gynecologic neoplasms.
目的:通过检测人类软骨蛋白39(YKL-40)和核转录因子KappaB(NF—KB)在子宫内膜异位症组织中的表达,探讨二者与子宫内膜异位症的关系及二者的相关性。方法:用免疫组化二步法检测40例子宫内膜异位症(EMS)患者的异位内膜和在位内膜及40例子宫肌瘤患者子宫内膜(对照组)YKL-40和NF—KB的表达,并对检测结果进行统计学分析。结果:YKL-40、NF—KB在子宫内膜异位症患者的异位和在位内膜及对照组内膜中的表达率分别为67.5%、62.5%、35%及88.5%、57.5%、32.5%,差异有统计学意义(P〈0.01);在不同月经周期YKL-40表达无统计学意义;在位内膜和正常内膜NF-KB的表达分泌期高于增殖期,差异有统计学意义(P〈0.05);YKL-40和NF-KB在三种内膜中的表达具有正相关性,相关系数分别是0.305,0.267和0.457(P〈0.01)。结论:YKL-40和NF—KB在EMS发生发展中起重要的作用。
Objective: To investigate the expressions of Human cartilage glycoprotein-39(YKL-40) and Nuclear factor-kappaB in endometriosis,and to evaluate the relationship between the YKL-40 and NF-κ B expressions.Methods: Immunohistochemistry was used to examine the YKL-40 and NF-κ B protein expressions in eutopic(n=40) and ectopic(n=40) endometrium of Ems,as well as endometrium of hystromyoma(n=40).The results were analyzed statistically.Results: The positive rate of YKL-40 and NF-κ B expressions in ectopic,eutopic and normal endometrium respectively were 67.5%,62.5%,35% and 88.5%,57.5%,32.5%(P<0.01);NF-κ B expression was higher in secretory phase than that in proliferative phase in the eutopic and normal endometrium(P<0.05),but YKL-40 was not correlated with the phase of menstrual cycle;Furthemore,the expression of YKL-40 and NF-κ B protein was positively correlated in these three medometrial tissue(rs=0.305,0.247 and 0.467,P<0.01).Conclusion: The over expression of YKL-40 and NF-κ B may play an important role in the pathogenesis of endometriosis.
目的解除宫颈癌放疗后放射性膀胱炎大出血致血块阻塞尿路。方法经尿道膀胱镜下,利用24F电切镜置入膀胱镜内,配合采用球式灌注器,反复冲洗,挤压,吸出淤积于膀胱内的血块,使尿路通畅。结果8例患者,其中7例一次成功清除膀胱内积血,1例二次完成。术后配合膀胱冲洗、抗炎、止血及支持疗法,挽救了生命,达到了治愈的目的。结论该方法治疗放射性膀胱炎急性大出血致血块十分有效。
Objective To summarize the investigation of the related genes of osteoporosis and study the relationship between the related genes and osteoporosis. Methods Consulting the domestic and overseas literature to summarize. Results & Conclusion Osteoporosis is a disease of many genes. The polymorphism of some genes and some cell factors with the change of bone mass are obviously correlated. But the mechanism of action has not been researched clearly and we need to extensively explore.
Objective To investigate the association of estrogen receptor-α(ER-α) and vitamin D receptor(VDR) gene polymorphisms with bone mineral density in Harbin postmenopausal women.Methods The ER-αPvuⅡand XbaⅠgenotypes and VDR BSMⅠgenotypes were determined by PCR-RFLP in 81 unrelated healthy women aged(51~70) years of Han nationality in Harbin.Bone mineral density(BMD) was measured by dual-energy X-ray absorptiometry.Results Frequencies of ER-αPvuⅡgenotype PP,Pp and pp were 13.6%,49.4% and 37.0% respectively.Frequencies of ER-αXbaⅠgenotype XX,Xx and xx were 4.9%,40.7% and 54.4% respectively.Frequencies of VDR BSMⅠgenotype BB,Bb and bb were 0%,16% and 84% respectively.T analysis indicated that a significant association was found between ER genotype and BMD at L_(2-4) in postmenopausal women and there were significant relationship in bone mineral density between the VDR BSMⅠgenotype at the trochiter and thighbone neck.PvuⅡgenotype and BSMⅠgenotype together have significant influence over BMD.Conclusion Hardy-Weinberg equilibrium was evident for both ER-α and VDR gene polymorphisms.Estrogen receptor-αand vitamin D receptor are associated with BMD,especially there were close relation between genes and BMD.
Objective To investigate the relationship between the vitamin D receptor gene polymorphism and bone mineral density (BMD) of osteoporotic fracture in Han nationality in Harbin. Methods The 98 subjects in Harbin district were divided into fracture and control groups. The patterns of VDR gene of 98 subjects were identified by polymerase chain reaction chain reaction-restriction fragment length polymorphism and bone mineral density (BMD) of all of them were determined by dual-energy X-ray absorptionetry (include lumbar 2~4, thighbone neck, wards triangle, troch, radius). Results There have significant difference of BMD at any sites in fracture and control groups (P0.01),The frequency distribution of genotypes bb, Bb, BB was 83.7%,16.3%,0%, respectively. The b allelic frequencies reached as high as 91.8%. There was no difference in two groups. X2 analysis indicated that there was no connection between osteoporotic fracture and VDR BsmⅠ. Through the comparison between control groups and osteoporotic fracture in lumbar, thighbone neck and radius, it showed that there was not significant difference in Bb and bb genotypes. Conclusion The majority VDR gene of 98 subjects in Harbin are Bb and bb and there was no connection between osteoporotic fracture and VDR BsmⅠ.
目的 将重组骨形态发生蛋白7质粒(pcDNA3.1-BMP7)转染至体外培养的兔骨髓间充质干细胞中,测定骨形态发生蛋白7基因在该细胞中的表达.方法 骨形态发生蛋白7基因序列测定、拼接后与报道的骨形态发生蛋白7基因序列对照;脂质体介导pcDNA3.1-BMP7转染骨髓间充质干细胞,G418筛选14 d,Western blot、PCR、原位杂交检测骨形态发生蛋白7基因的表达.结果 序列测定证实与GenBank报道的骨形态发生蛋白7全长基因序列一致;转染pcDNA3.1-BMP7质粒的骨髓间充质干细胞有大量骨形态发生蛋白7蛋白及骨态发生蛋白7 mRNA表达.结论 脂质体介导下,重组真核表达载体pcDNA3.1-BMP7能被转入骨髓间充质干细胞,且表达了骨形态发生蛋白7及骨形态发生蛋白7 mRNA.
