[Objective] To isolate and identify a spoilage bacteria in the ultra-high pressure preserved Lentinula edodes foods.[Method] Spoilage microorganisms in ultra-high pressure treated Lentinula edodes food were isolated by diluted plate and streak cultivation methods.And the morphological,physiological,biochemical properties,16S rDNA sequence and phylogenetic tree of the isolated strains were analyzed.[Result] A strain of Bacillus sp.(No.MW1001) was isolated from the treated shiitake mushroom samples.Based on the results of 16S rDNA sequencing,and physiological and biochemical properties,the strain showed high genetic relation to Bacillus methylotrophicus and Bacillus subtilis with the homology of 99.55% and 99.24% respectively.[Condusion]The research provided theoretical basis for further improving the ultra-high pressure preservation of Lentinula edodes.
A bacterial isolate,WM1002,obtained from packaged Pleurotus geesterani fruit bodies that had been processed using super high pressure technology,was identified as a strain of Bacillus based on 16S rRNA sequencing.The 16S rRNA sequence has been deposited in GenBank under the accession number,HQ730134.A phylogenetic tree consisting of 16S rRNA sequences from other Bacillus species and constructed using the neighbor-joining method revealed that isolate WM1002 exhibited a high genetic similarity(99.8%) with B.velezensis CR-502,B.vallismortis DSM11031,B.methylotrophicus CBMB205 and B.atrophaeus JCM9070.
The aim of this study is to identify the bacteria strain numbered as WM1003 isolated from the package of the Agrocybe cylindracea processed by super high pressure technology. The genomic DNA of the strain WM1003 was isolated and purified. 16S rRNA gene sequence was amplified using genomic DNA of the strain WM1003 as template and was sequenced. Phylogenetic position of the WM1003 was analyzed by using MEGA4.0 based on the 16S rRNA sequences. PCR product of the strain WM1003 was about 1600 bp. The 16S rRNA gene sequence of the strain WM1003 was accepted by GenBank as HM748831. Phylogenetic analysis based on the 16S rRNA sequences showed that the strain WM1003 held highest similarity with Paenibacillus jamilae CECT 5266 (99.5%). In the present study, the strain WM1003 was identified through 16S rRNA gene sequence and phylogenetic analysis. This laid the foundation for the application of super high pressurs technology in Agrocybe cylindracea.
In a construction project of reinforced concrete tube structure of prilling tower for compound fertilizer,used the slip form to raise the form-erecting-flat,embedded the embedded part in the concrete tower body,take the form-erecting-flat as erecting flat of high-altitude stiff beam and slab.Introducing the above mentioned construction technique,can help decreasing the difficulty of high-altitude construction,and saving labour and material cost,reducing the erection time.
