In this study, through the construction of eukaryotic expressive vector, the recombinant Plasmids pEGFP-N3-vMIP-Ⅰ was transfected into Jurkat cells by electroporation. Then, by QRT-PCR technique, we detected the expression levels of anti-HIV genes: CCL5, APOBEC3F, MX1 in Jurkat cells which influenced by vMIP-Ⅰgene and explored the mechanisms of vMIP-Ⅰ against HIV infection. The results he recombinant plasmids of pEGFP-N3-vMIP-Ⅰwas successfully constructed and electroporation transfection efficiency reached about 40%. In comparison with non-transfected gourp, the transfected vMIP-Ⅰgroup can increase CCL5, A3G, A3F, and MX1 of Jurkat cells by 7.37, 1.58, 2.42 and 2.064 times seperately. The results suggest vMIP-Ⅰcan activate expressions of some relative anti-HIV genes in Jurkat cells, which probably is one of the mechanisms for anti-HIV infection.
Objective:To study the expression level of anti-HIV genes in KSHV (Kaposi’s sarcoma-associated herpesvirus) infected Jurkat cells.Methods: BCBL-1 cell was been stimulated by TPA (20ng/ml) 72 hours before havest KSHV virions solution.Jurkat cells were cultured with PRMI-1640 contain KSHV virions.24 hours after infection,The mRNA quantity of some anti-HIV genes in Jurkat cell infected with KSHV after 3 and 5 day’s were investigated by using Real-Time PCR.Results: After Jurkat cell infected with KSHV for 3 days,the mRNA production of RANTES,APOBEC3G,APOBEC3F,MX1,and MX2 increased 123,3.12,1.18,2.75,4.36 folds,respectively.In 5 days after infection,RANTES,MX1,and MX2 increased 11.91,2.72 and 2.22 times,respectively.Conclusions: the expression of anti-HIV genes(RANTES,APOBEC3G,APOBEC3F,MX1,MX2) up-regulated in KSHV infected Jurkat cells.
RNA interference is an evolutionarily conserved surveillance mechanism that responds to double-stranded RNA by sequence-specific silencing of homologous genes or interference homologous gene expression by DNA Methylation.Traditionally,RNA interference is also called "post-transcriptional gene silencing (PTGS)".However,recent studies have revealed that siRNA initiates the activation of methytransferases,which causes the methylation of the promotor and silencie gene expression at post-transcriptional level.RNA interference (RNAi) can regulate a variety of biological processes.Recent evidence supports the notion that both cellular and viral miRNA (vmiRNA) are able to modulate viral replication.There is also evidence that viruses like HIV-1 have evolved strategies to control the cell’s RNAi activity.So it is expected that RNA interference may effectively applied in HIV-1 infection prevention and AIDS treatment.This article reviews the current progress of studies on the mecha- nism and application of RNA interference in HIV-1infection.
