为探究山东省滨州市畜禽粪污资源的空间分布现状及近年来畜禽粪污土地承载力变化情况,优化畜牧业发展格局,推动畜牧养殖业绿色发展,参照《土地承载力测算技术指南》中的相关计算方法,测算了2018年滨州市各县区的畜禽粪污养分供应量、农作物粪肥养分需求量以及畜禽粪污土地承载力,分析了滨州市近6年畜禽粪污土地承载力变化.结果表明,滨州市的种植业与畜牧业发展规模在空间上存在一定的错位,种植业发达的地区主要为惠民县、邹平市、沾化区,而畜牧业发达的地区则分布在阳信县、无棣县;2018年滨州市农作物粪肥氮、磷养分需求量分别为3.47万t和0.74万t,畜禽粪肥氮、磷的供给量分别为2.98万t和0.56万t,可见,该区域农作物粪肥养分需求量均高于畜禽粪污养分供给量.从时间上来看,近6年滨州市以氮、磷为基准的畜禽粪污土地承载力指数均呈持续下降趋势.结果表明,滨州市全市畜禽养殖发展空间较大,可适度增加畜禽养殖规模,但在农牧业生产布局和区域间协同上仍需重点关注.

A new type of cephodexD microcarrier was used to culture ST cells for the classical swine fever virus(CSFV) reproduction,and the results were compared with those of conventional monolayer culture.At 72 h,up to 18.9 × 105 cells/mL in microcarrier suspension culture,the cell density was two times than a static monolayer cultivation process.Compared with the monolayer culture,the virus titer was up to 7.5× 10s RID/mL in microcarrier suspension culture,increased by 50%.In one production process,more than two batch of qualification CSFV product could be harvested compared with traditional culture method.The CSFV produced by the new microcarrier suspension culture technology could stimulate the specific neutralizing antibody in pigs,and the protective rate was 100%.Therefore,it possessed a satisfied immunogenicity.The new microcarrier suspension culture process is superior to the monolayer culture method with an important application value in the field of CSFV large-scale production.

An HPLC-MS method has been developed and validated for the quantitative determination of milbemycin oxime (MBO) in dog plasma. The developed method has been then applied in in vivo clinical studies to obtain pharmacokinetics of MBO in blood after its oral administration. Samples were extracted using solid-phase extraction (SPE). Sample proteins were precipitated with acetonitrile (ACN) and sodium chloride (NaCl) and then diluted with methanol and water. Calibration standards were prepared by using plasma matrix and following the same SPE procedure. Chromatographic separation was performed on a Waters C18 packed column (3.5 μm particles diameter; 3 × 100 mm) with a C18 guard column (3.5 μm particles diameter; 3 × 20 mm). The mobile phase was an 85:15 (v/v) mixed solution of ACN and 5 mM ammonium acetate. The calibration curve was linear over a concentration range of 2.0-500 ng/mL with a limit of quantitation of 2.0 ng/mL. The oral administration of a pellet of 2.5 mg MBO produced blood concentrations ranging from 6.10 ± 0.92 to 78.81 ± 4.38 ng/mL within 6 h, with a terminal half-time of 11.66 ± 0.93 h. This study determined the suitability of the herein proposed method to investigate the pharmacokinetics of MBO after oral administration.
Ethnopharmacological relevance: Flos populi (male inflorescence of Populus tomentosa Carriere) has been traditionally used in East Asian countries for the treatment of various inflammatory diseases, strengthening the spleen and stomach, anti-rheumatic, anti-tumor and anti-diarrhoeal.Aim of the study: To evaluate the in vivo or in vitro anti-diarrhoeal and anti-microbial activity of Flos populi aqueous extract.Materials and methods: Acute toxicity of Flos populi aqueous extract (FPAE) was investigated. Castor oil-induced diarrhoea method was used to evaluate the anti-diarrhoeal activity, inhibition of defecation and diarrhoea were determined in mice, effects on castor oil-induced enteropooling, intestinal transit and intestinal fluid secretion in rats or mice. Minimum inhibitory concentration (MIC) and minimum bactericidal concentration (MBC) of FPAE against strains of three clinical bacterial isolates and one reference strain were used to test the anti-microbial activity.Results: The FPAE reduced the frequency of diarrhoea episodes and decreased the propulsion of charcoal meal through the gastrointestinal tract in a dose dependent manner. FPAE (100-500 mg/kg, p.o.) produced dose-dependent and significant (P < 0.01) protection of mice against castor oil-induced diarrhoea. FPAE, dose-dependently and significantly (P < 0.01) delayed the onset of castor-oil induced diarrhoea, decreased the frequency of defecation, and reduced the severity of diarrhoea. Compared with control animals, FPAE, dose-dependently and significantly (P < 0.01) decreased the volume of castor oil-induced intestinal fluid secretion, and reduced the number, weight and wetness of faecal droppings. There was no deaths or abnormalities in behaviour seen in the acute toxicity test. The aqueous extract displayed anti-microbial effects to three species of bacteria in anti-microbial test.Conclusion: The findings of this study indicate that FPAE possesses anti-diarrhoeal property in rats and mice and confirm the ethnomedicinal use of Flos Populi as a valuable natural remedy for the treatment, management and/or control of diarrhoea. Calpain system played an important role in muscle protein hydrolysis and muscle tenderization after animal slaughter. Fatty acid binding proteins affected muscle tenderness,flavor and juiciness,and had high relevance to backfat thickness,marbling, tenderness and water capacity of meat traits through taking part in the transport and metabolism of fatty acids. Myogenic regulatory factors controled the fusion of muscle cell and formation of muscle fiber during the muscle differentiation process. Myostatin,a negative regulator for muscle growth,could affect lean meat percentage of animal and better meat quality traits. Several meat quality candidate genes in cattle including calpain system family,fatty acid binding protein gene,myogenic regulatory factor,and myostatin were summarized in order to find the way to improve the quantity and quality of meat and further improve the meat quality traits through marker-assisted selection.
