Objective To observe the effect of Qianliening on IL-1β and its mRNA expression in rats with chronic bacterial prostatitis.Methods 40 rats were divided randomly into normal group,model group,Qianliening granule group and Qianlietong control group.The rat models of chronic bacterial prostatitis were established with injection of E.coli suspension.The pathological reactions of prostate tissue were observed with optical microscope,the content of IL-1β in serum was detected by radioimmunoassay,and the expression of IL-1β mRNA was detected by RT-PCR after 30 days.Results In Qianliening granule group,the pathological reactions of rats with chronic bacterial prostatitis were alleviated,the content of IL-1β in serum and its mRNA expression were decreased.Conclusion Qianliening has good effect on rats with chronic bacterial prostatitis,and the mechanism is possibly related to the decreased content of IL-1β and its mRNA expression level.
目的 探讨血府逐瘀汤对人脐静脉内皮细胞(HUVEC)分泌血管内皮生长因子(VEGF)、血小板源性生长因子(PDGF)的影响及其与促进HUVEC增殖的关系.方法 采用血府逐瘀汤含药血清体外干预HUVEC培养,通过MTT法和酶联免疫吸附分析(ELISA)法观察血府逐瘀汤对HUVEC的体外增殖和分泌VEGF、PDGF的影响作用.结果 MTT检测结果显示血府逐瘀汤含药血清浓度为10%时对HUVEC具有显著增殖作用;ELISA检测结果显示血府逐瘀汤具有促进HUVEC分泌VEGF的作用,而对PDGF的分泌无明显的作用.结论 血府逐瘀汤在一定剂量范围内能促进HUVEC增殖和促进HUVEC分泌VEGF,而对PDGF的分泌无明显促进作用;提示血府逐瘀汤可能通过促进HUVEC分泌VEGF而促进细胞增殖,从而起到祛瘀生新的作用.
Objective To investigate the effects of different extracts from Xuefu Zhuyu decoction(XFZYD) on the proliferation of human umbilical vein endothelial cells(HUVEC).Methods XFZYD water extracts were extracted respectively with n-butanol,petroleum ether,and chloroform,and drug-containing serum of rats were prepared by the different extracts and remainder.HUVEC were treated by the drug-containing serum with different concentrations and the VEGF level in cell culture fluid was measured by ELISA,and the concentration of iNOS and NO were measured by nitrate reductase method.Results The concentration of VEGF,iNOS and NO in cell culture fluid in chloroform group and remainder group increased obviously compared with that of other groups.Conclusion The chloroform extract and remainder from XFZYD can promote obviously the proliferation of HUVEC.
Objective To study the effect of Qianliening on rats with benign prostatic hyperplasia. Methods 120 SD rats were divided randomly into blank group, model group, Baoliezhi control group and Qianliening granule groups with different doses. The models were established with hypodermic injection of testosterone propionate at 4 mg/kg after orchiectomy, at the same time, these rats were gastric perfused with different drugs respectively for 30 days. The body weight, prostate weight, prostate index, morphological changes, the level of T and E2 were measured. Results The body weight in model group was significantly lower than that in control group (P0.05), the prostate weight and index were obviously higher than that in control group (P0.01). The prostate weight and index were decreased significantly in Qianliening granule groups with meddle and high doses (P0.01), the level of testosterone was increased significantly and E2 was decreased obviously in Qianliening granule group with high doses. Conclusion Qianliening can decrease the prostate index and wet weight, and adjust the rate of T/E2, it shows an obvious dose-effect relationship on rats with benign prostatic hyperplasia.
