目的 探究二氢杨梅素(DMY)对脂多糖(LPS)诱导的小鼠神经炎症的影响.方法 60只C57BL/6J雄性小鼠随机分成正常组,模型组,低、中、高剂量组.低、中、高剂量组按照10、20、40 mg/kg腹腔注射DMY,正常组和模型组腹腔注射等体积的生理盐水,1次/d,共7 d.最后一次给药30 min后,模型组和低、中、高剂量组采用侧脑室注射LPS建立小鼠神经炎症模型,正常组侧脑室注射等体积的生理盐水.动物模型建立24 h后,RT-qPCR法检测5组小鼠大脑中CD206和CD14 mRNA的相对表达量情况,Western blot检测5组小鼠大脑中的S6/HIF 1信号通路蛋白的表达水平.结果 各组CD206 mRNA相对表达量比较,模型组低于正常组(P<0.05),低剂量组与模型组比较无明显变化(P>0.05),中、高剂量组高于模型组(P<0.05或0.01);各组CD14 mRNA相对表达量比较,模型组高于正常组(P<0.01),低、中、高剂量组低于模型组(P均<0.01);各组P-S6蛋白表达水平比较,模型组高于正常组(P<0.05),低、中、高剂量组低于模型组(P<0.05或0.01);各组P-S6K1蛋白表达水平比较,模型组高于正常组(P<0.01),低、中、高剂量组低于模型组(P<0.05或0.01);各组HIF1蛋白表达水平比较,模型组高于正常组(P<0.05),低、中、高剂量组低于模型组(P<0.05或0.01).结论 DMY能够改善LPS诱导的小鼠急性神经炎症,这可能与DMY抑制炎症因子的表达及S6k1 HIF1信号通路有关.
目的 探讨齐墩果酸(OA)对人结肠癌细胞(HT-29)增殖与凋亡的影响及其分子机制.方法 体外培养HT-29细胞,用40、80、120μmol·L-1 OA进行干预后,用白介素-6(IL-6)刺激,MTT法和集落形成实验检测细胞增殖情况,倒置显微镜观察细胞密度变化,流式细胞术检测细胞周期,DAPI染色和Annexin V/PI流式细胞仪检测细胞凋亡,Caspase-3活力测定试剂盒测定Caspase-3活力,反转录-聚合酶链反应和Western blot实验检测IL-6/STAT3信号通路活化及其通路相关基因和蛋白的表达.结果 OA可显著抑制HT-29的增殖能力(P<0.01),并诱导其凋亡(P<0.01),显著抑制IL-6/STAT3信号通路的活化.结论 OA通过抑制IL-6/STAT3信号通路的活化,抑制HT-29增殖和促进其凋亡.
目的 采用脂多糖(LPS)刺激小胶质细胞(BV2)以建立体外炎症模型,初步探讨二氢杨梅素(DMY)对小胶质细胞表型转化的影响. 方法 将BV2细胞分为空白组、模型组、DMY低、中、高剂量组(10、20、40 μmol/L),DMY干预细胞半小时后,给予LPS造模.采用MTT法检测DMY对BV2细胞活性的影响,WesternBlot检测CD14、iNOS、Arg1的蛋白表达情况. 结果 MTT结果显示DMY在10~50 μmol/L对BV2细胞活性无明显影响(P>0.05);Western Blot结果显示DMY能抑制LPS受体CD14及M1型小胶质细胞标志物蛋白iNOS的表达,同时能促进M2型小胶质细胞标志物蛋白Arg1的表达. 结论 DMY可抑制活化的小胶质细胞向M1型即促炎表型极化,并促进其向M2型即抗炎表型转化,以达到抑制神经炎症,保护神经元的作用.
OBJECTIVE To observe the effects of the radix pseudostellariae health wine on contraction of mice intestinal smooth muscles in vitro and explore the possible mechanisms.METHODS The mice isolated intestines were used to observe the effect on normal mice intestinal contraction and the intestinal stimulated antagonism to ace-tylcholie and barium chloride by the radix pseudostellariae health wine at different concentrations.RESULTS There are obvious effects on intestinal smooth muscle in normal intestine and incitant intestine caused by Ach and Atr.Conclusion Radix Pseudostellariae Health Wine can excite the intestinal constringency.
OBJECTIVE To observe the effects of Radix Pseudostellariae Health Wine the digestion func-tion.METHODS The mice were randomly divided into treatment group and control group,and the treatment group was fed with high dosage,middle dosage,low dosage.The secretion of gastric juice and gastric acid,pepsin,pepsin activity ,the distance of ink moving were used to evaluate the effect.RESULTS Radix Pseudostellariae Health Wine increase the rate of ink promoting, which was all dosage group to that in the Difenoxin model control group ( P<0.05),Radix Pseudostellariae Health Wine also could promote the secretion amount of gastric aicid,which was supe-rior to that in the control group ( P<0.05).Radix Pseudostellariae Health Wine also could promote the secretion a-mount of pepsin, which was middle dosage and low dosage group to that in the control group ( P <0.05 ).CONCLUSION Radix Pseudostellariae Health Wine has the function of promoting the digestion function.
ABSTRACT:OBJECTIVE To investigate effect of Xinhuang tablets and its Chinese medicine components on high uric acid Mice Caused by hypoxanthine , and discusse its mechanism .METHODS The mice models of high uric acid caused by injecting hypoxanthine and the normal mice were adopted .After giving different doses of intragastric administration with extracts from Xinhuang tablets and its Chinese medicine components on the mice by gavage ,the uric and xanthine oxidase level of each group were determined .RESULTS After 1h of intraperitoneal injection of hypoxanthine ,positive control group and Xinhuang tablets and its Chinese medicine components dosage ( 23.75、47.5、95 mg? kg -1 ) could against high uric mice induced by hypoxanthine and reduced the activity of xanthine oxi -dase in hepar ( P<0.01 ) .CONCLUSION Xinhuang tablets and its Chinese medicine components could decrese the blood uric acid levels by inhibiting the activity of xanthine oxidase .
目的:观察颈康片对退变颈椎间盘组织超微结构的影响并探讨其作用机制.方法:28只新西兰兔按随机数字表法分成模型组和正常对照组,模型组又分为颈康片组、颈复康组、生理盐水组,分别喂服颈康片、颈复康、生理盐水,正常对照组喂服生理盐水,4周后取颈椎间盘髓核进行电镜观察.结果:电镜观察见髓核软骨细胞及胶原纤维均发生退变,颈康片组退变程度最轻.结论:颈康片能有效减轻髓核软骨细胞及胶原纤维退变,这可能是其作用机制之一.
新癀片由肿节风、三七、人工牛黄、猪胆汁膏、肖梵天花和吲哚美辛等组成,具有清热解毒、活血化瘀、消肿止痛功效,主治热毒瘀血所致的咽喉肿痛,牙痛,痹痛,无名肿毒等症.新癀片中药组分是在新癀片的基础上去吲哚美辛而制成.近年来,新癀片在临床上也用于治疗痛风性关节炎[1-2],但未见新癀片对痛风性关节炎疗效的药效学报道.我们采用氧嗪酸钾引发小鼠出现高尿酸,观察新癀片及新癀片中药组分对小鼠血尿酸的影响.

目的 探讨血府逐瘀汤对人脐静脉内皮细胞(HUVEC)分泌血管内皮生长因子(VEGF)、血小板源性生长因子(PDGF)的影响及其与促进HUVEC增殖的关系.方法 采用血府逐瘀汤含药血清体外干预HUVEC培养,通过MTT法和酶联免疫吸附分析(ELISA)法观察血府逐瘀汤对HUVEC的体外增殖和分泌VEGF、PDGF的影响作用.结果 MTT检测结果显示血府逐瘀汤含药血清浓度为10%时对HUVEC具有显著增殖作用;ELISA检测结果显示血府逐瘀汤具有促进HUVEC分泌VEGF的作用,而对PDGF的分泌无明显的作用.结论 血府逐瘀汤在一定剂量范围内能促进HUVEC增殖和促进HUVEC分泌VEGF,而对PDGF的分泌无明显促进作用;提示血府逐瘀汤可能通过促进HUVEC分泌VEGF而促进细胞增殖,从而起到祛瘀生新的作用.
目的:研究芎芍胶囊对人脐静脉内皮细胞(HUVEC)增殖的影响.方法:采用血清药理学和体外培养HUVEC的方法,通过MTT法及流式细胞术(FCM)检测芎芍胶囊含药血清对细胞增殖能力的影响.结果:MTT法检测HUVEC在芎芍胶囊浓度5%时产生最大增殖效应,增殖率为54.98%;FCM法检测发现芎芍胶囊浓度为5%时S期比例最大,为42.65%:当浓度达10%及以上时,其促HUVEC增殖的作用逐渐减弱.结论:芎芍胶囊能促进HuVEC的增殖,在一定浓度内,其促进细胞增殖的作用与浓度有一定的关系;提示芎芍胶囊的促血管新生的作用可能是通过促进血管内皮细胞的增殖.
Objective To investigate the effect of xiongshao capsule on secretion of angiogenesis factor of human umbilical vein endothelial cells(HUVEC).Methods The serological pharmacological method and in vitro intervention were used to incubate HUVEC.MTT method and enzyme-linked immunosorbent assay method were adopted to observe the impacts of xiongshao capsule at low,medium and high doses on in vitro proliferation of HUVEC,the secretions of Ang,Ang-2,VEGF,bFGF,EGF and TGF-β1.Results Xiongshao capsule of each dose promoted HUVEC proliferation.That of low dose promoted the secretions of Ang,Ang-2,EGF and VEGF of HUVEC,but did not impact the secretions of bFGF and TGF-β1 apparently.That of moderate dose promoted the secretions of Ang,Ang-2,EGF and bFGF of HUVEC,inhibited the secretion of TGF-β1,but had not apparent impact on the secretion of VEGF.That of high dose increased secretions of Ang,EGF and bFGF of HUVEC and inhibited the secretions of VEGF and TGF-β1,but had no significant impact on Ang-2 secretion.Conclusion Xiongshao capsule promotes the proliferation of HUVEC and increases the secretion of angiogenesis factor and inhibits the expression of vascular growth factor,which is related to the dose of drug to a certain extent.
从原文和临床应用出发,反思桂枝汤的现代药理研究,提出当前方剂的药理研究应该要从流到源,紧密结合方剂原义和临床应用,回到其原来的科学内涵上。并从全面认识方剂、重视方剂对应的脉象和证候、根据方剂的原来内涵研究其物质基础和药理效应三方面进行论述。
Objective To study the protective effect of Hypericum japonicum against acute liver injury in mice and its mechanism.Methods After acate liver injury model was induced by carbon tetrachloride(CCl4),mice were administrated intragastrically with Hypericum japonicum.The serum ALT,AST,SOD,NO,TNF-α,IL-6 were determined and pathological change of liver was observed.Results Hypericum japonicum could significantly decrease the elevation of serum ALT,AST,NO,TNF-α,IL-6 induced by CCl4,and increase SOD activity,ameliorate the pathological injury of liver.Conclusion Hypericum japonicum shows protective effect against experimental liver injury in mice.Decrease the serum ALT and AST,increaseing anti-peroxide,restraining produce of the serum NO,TNF-α and IL-6,may be one of its mechanisms.
目的:观察并比较固表解毒合剂等对干酵母所致发热大鼠体温的影响。方法:大鼠皮下注射15%干酵母悬液10mL/kg后立即以固表解毒合剂灌胃给药,同时以不同中药及阿司匹林为对照,于给药后1、2、4、6、8h测体温,求出各测定时刻体温相对于正常体温的变化值。结果:固表解毒合剂在给药后的1h体温升高不明显,且在4、8h可显著降低体温的升高(P<0.05-P<0.01)。结论:固表解毒合剂具有一定的解热作用,可能与扶正固本的作用有关。
To study the effect of Gubiao Jiedu decoction on the function of immune systerm in mice depressed by cyclophosphamide.The contents of IgG,IgM,IFN-γ and IL-2 in serum were determined with ELISA.The rusults showed that the contents of IgG,IgM,IFN-γ and IL-2 were increased by low dosage of the decoction.It is suggested that Gubiao Jiedu decoction may enhance the function of immune systerm of mice depressed by cyclophosphamide.
To study the best preparation of kangmeifu burn ointment (KBO) by emulsifying process. According to the description of the ointment and the result of centrifugal test,the optimum proportion of matrix of KBO was determined by orthogonal experiment.The results showed the optimum proportion was stearic acid-white vaselin-liquid paraffin-glyceryl monostearate (3:2.5:2.5:4),emulsifier dosage is 6% of prescription amounts,HLB 11,and emulsifying temperature 80℃.Conclusion:The method is convenient and performed.
目的:研究固表解毒合剂体外抗病毒作用。方法:采用细胞培养法和鸡胚法,观察固表解毒合剂对甲型流感病毒的抑制作用,并与其他处方为对照。结果:固表解毒合剂对MDCK细胞和鸡胚的毒性小;当药物浓度为0.045g/mL时可保护50%细胞不产生病变,药物浓度为0.056g/mL时可保护50%鸡胚不受感染。结论:固表解毒合剂对甲型流感病毒具有抑制作用,其抑制作用比其他处方强。
