目前磷酸化肽段鉴定主要依赖于质谱技术,但磷酸化肽段的低丰度性以及来自非磷酸化肽段的干扰等因素,影响质谱的分析与鉴定.因此质谱分析前磷酸化肽段的富集,是深入研究磷酸化蛋白质组学的先决条件.该文介绍了磷酸化蛋白质组学中传统的以及新建立的一些磷酸化肽段分离富集方法的原理及优缺点,这些方法包括固相金属离子亲和色谱法(IMAC)、金属氧化亲和色谱法(MOAC)、强阳/阴离子交换色谱法(SCX/SAX)、亲水相互作用色谱法(HILIC)、静电排斥亲水相互作用色谱法(ERLIC)、化学衍生法、MALDI靶盘富集法以及多种富集方法相结合.
目的:建立气相色谱(GC)法同时测定炉甘石薄荷脑洗剂中薄荷脑和苯酚的含量,为炉甘石薄荷脑洗剂质量控制提供可靠的分析方法.方法:以樟脑为内标,95%乙醇为溶剂,Agilent DB-624毛细管柱(30 m×320tm,1.8 μm)为色谱柱,载气为氮气,柱温140 ℃,进样口温度为240 C,氢火焰离子化检测器(FID)温度为250℃,分流比为10∶1,进样量1μl.结果:薄荷脑、苯酚浓度分别在69.97~~ 129.95 μg/ml(r=0.999 1)和141.75~ 263.25 μg/ml(r=0.999 2)范围内与内标的峰面积呈良好线性关系.薄荷脑和苯酚的平均回收率分别为(103.35±1.14)%和(103.16±0.95)%(n=9).结论:该方法简便、可行、重复性好,可用于炉甘石薄荷脑洗剂的质量控制.
A label-free method for arginine(Arg) detection was developed based on the fluorescence enhancement of a new water-soluble conjugated polymer(P1) in aqueous solution.The linear ranges for Argdetermination were 6.0×10-8-3.06×10-6 mol/L and 3.06×10-6-7.11×10 5 mol/L with the detection limit as low as 2.0 × 10-8 mol/L.This method is simple,rapid,highly selective and sensitive,rendering its practical use for the sensing of arginine hydrochloride injection.
A novel conjugated polyelectrolyte containing meta-substituted pyridyl groups in the backbone (P1) and sulfonate groups in the side chain is reported as an optical pH-sensitive probe in aqueous solution.The ability of P1 to function as a colorimetric and fluorescent pH probe was demonstrated with dramatic color and fluorescence changes upon introduction of acid.These photophysical responses were attributed to the protonation of pyridyl units accompanied with an aggregation behavior of P1 in acidic solution,which is confirmed by dynamic light scattering.This work presents the feasibility of varying the mainchain structures to fine-tune the pH-dependent properties of the resulting conjugated polyelectrolytes (CPEs).
