目的 探讨lncRNA GM 10786靶向结合miR-142抑制糖尿病肾病(DN)单核巨噬细胞炎性因子分泌的作用及机制.方法 高通量转录芯片筛选DN患者与健康对照者外周血单核细胞差异表达lncRNA.实时荧光定量PCR(qRT-PCR)检测高糖刺激下THP-1细胞中GM10786的表达水平.在THP-1细胞中转染Lv-control、Lv-GM10786后,qRT-PCR测定高糖刺激下转染后细胞中pro-IL-1β和TNF-α的表达水平.采用双荧光素酶报告基因实验验证GM10786与miR-142的靶向结合及miR-142靶向调控Nrf2.THP-1细胞中转染空质粒(Control)、miR-142、以及共转染GM10786和miR-142后,qRT-PCR和Western blotting检测Nrf2和HO-1的表达水平,ELISA和Western blotting检测高糖刺激下转染细胞培养上清及细胞中IL-1β和TNF-α的表达水平.结果 与健康对照组相比,GM10786在DN患者外周血单核细胞中的表达显著下调(P<0.05).在高糖刺激下GM10786在THP-1中的表达相较正常培养基也显著下调(P<0.05).与HG+Lv-control组相比,HG+ Lv-GM 10786组中pro-IL-1β和TNF-α的表达水平均下调,差异有统计学意义(P<0.001).双荧光素酶报告基因证实GM10786能够与miR-142特异性结合,Nrf2是miR-142的靶基因.与转染miR-142组相比,共转染GM10786和miR-142组的THP-1细胞中Nrf2和HO-1表达水平显著增加(P<0.05),高糖刺激下IL-1β和TNF-α的表达水平均显著下降(P<0.05).结论 GM10786是DN进程中重要的负调控因子,GM10786通过海绵吸附作用靶向结合miR-142和调节Nrf2表达来抑制DN中单核巨噬细胞炎性因子的分泌,该研究将有助于DN的靶向治疗.
Nephrotoxicity is a common complication of cisplatin chemotherapy and thus limits the use of cisplatin in clinic. The purinergic 2X7 receptor (P2X7R) plays important roles in inflammation and apoptosis in some inflammatory diseases; however, its roles in cisplatin-induced nephrotoxicity remain unclear. In this study, we first assessed the expression of P2X7R in cisplatin-induced nephrotoxicity in C57BL/6 mice, and then we investigated the changes of renal function, histological injury, inflammatory response, and apoptosis in renal tissues after P2X7R blockade in vivo using an antagonist A-438079. Moreover, we measured the changes of nod-like receptor family, pyrin domain containing proteins (NLRP3) inflammasome components, oxidative stress, and proapoptotic genes in renal tissues in cisplatin-induced nephrotoxicity after treatment with A-438079. We found that the expression of P2X7R was significantly upregulated in the renal tubular epithelial cells in cisplatin-induced nephrotoxicity compared with that of the normal control group. Furthermore, pretreatment with A-438079 markedly attenuated the cisplatin-induced renal injury while lightening the histological damage, inflammatory response and apoptosis in renal tissue, and improved the renal function. These effects were associated with the significantly reduced levels of NLRP3 inflammasome components, oxidative stress, p53 and caspase-3 in renal tissues in cisplatin-induced nephrotoxicity. In conclusions, our studies suggest that the upregulated activity of P2X7R might play important roles in the development of cisplatin-induced nephrotoxicity, and P2X7R blockade might become an effective therapeutic strategy for this disease.
Activation and infiltration of T cells and macrophages are key features of renal tubulointerstitial injury. The costimulatory molecule V-set and immunoglobulin domain-containing protein-4 (VSIG4), which is exclusively expressed on macrophages, is capable of inhibiting the T cell response. However, it is unclear whether VSIG4 is involved in renal tubulointerstitial injury. This study was designed to investigate the role of VSIG4 in renal tubulointerstitial injury and the related T cell infiltration.
Objective To observe the effects of endoplasmic reticulum stress (ERS) on the activation of monocytes induced by high glucose and explore the underlying mechanism.Methods The monocyte cell line THP-1 was stimulated with high glucose,and then treated with molecular chaperone betaine.The levels of glucose regulation protein 78 (GRP78) and p-JNK,which were associated with ERS were detected by real-time PCR and Western blotting.The proliferation of the cell line was detected by MTT method.Transwell and immunofluorescence were applied to observe the chemotaxis and phenotype of cells respectively.Results The levels of GRP78 and p-JNK of THP-1 cells stimulated by high glucose were significantly increased compared with the normal control group (all P < 0.05).The proliferation and chemotactic were also enhanced (all P < 0.05).The number of cells in M1 phenotype was increased remarkably (P < 0.05).All the indexes above could be rescued by betaine.Conclusion The activation of THP-1 cells can be induced by high glucose through ERS,while molecular chaperone betaine can reverse the activation.
Objective To investigate the effects of p-cresol on human umbilical vein endothelial cells.Methods The effects of p-cresol on endothelial cell growth,cell cycle,cell morphological change and p21 protein were detected by the CCK-8 assay,flow cytometry assay,inverted microscope and Western blotting.Results P-cresol could inhibit the growth of human umbilical vein endothelial cells in dose-and time-dependent manners (all P < 0.05).The human umbilical vein endothelial cells treated with p-cresol became elongated processes,cloudy cytoplasm,and irregular shapes.The p-cresol stopped human umbilical vein endothelial cells at cell cycle G1 and had no effect on cell apoptosis.The p-cresol could increase protein expression of p21 in a dose dependent manner (P < 0.05).Conclusion P-cresol can increase protein expression of p21,induce cell cycle arrest at G1 stage and inhibit the proliferation of human umbilical vein endothelial cells.

慢性肾脏病(CKD)的发病率高达10%,已成为威胁人类健康的隐形杀手.慢性肾衰竭患者普遍伴随着钙磷代谢紊乱,长期高磷血症除了引发继发性甲状旁腺功能亢进、肾性骨病、维生素D代谢障碍,还能引起心脑血管病变和严重心脑血管事件,是终末期肾病患者心脑血管疾病发生率和死亡率增高的重要因素[1].如何有效控制慢性肾衰竭患者血磷水平,防治各种并发症,改善患者生活质量,日益受到临床医师的重视.现就近年高磷血症发生机制、危害及其治疗研究进展结合我们的工作谈如下体会.
Objective To investigate the change and clinic significance of serum Lkn-1 and TNF-α levels in patients with chronic glomerulonephritis.Methods Forty-three chronic glomerulonephritis,patients(include focal segmental glomerulonephritis,membranous nephropathy,IgA nephropathy,mesangial proliferative glomerulonephritis),14 chronic renal failure patients(8 patients received renal histology) and 11 normal controls were involved in this experiment.The serum levels of Lkn-1 and TNF-α were tested by means of sandwich ELISA technique.24 hours urine protein quantity and serum creatinine were also detected.The ratios of sclerosing glomeruli,crescents were calculated.Results The levels of Lkn-1 in focal segmental glomerulonephritis,IgA nephropathy were much higher than that in controls.There was no significant difference in the serum level of Lkn-1 among membranous nephropathy,mesangial proliferative glomerulonephritis and controls.The levels of TNF-α in IgA nephropathy were much higher than that in controls.No significant difference was found in the serum level of Lkn-1 among membranous nephropathy,mesangial proliferative glomerulonephritis and controls.The level of Lkn-1 and TNF-α in patients with chronic glomerulonephritis was correlated positively to the ratio of sclerosing glomeruli.The level of TNF-α was correlated positively to 24 hours urine protein quantity.Results showed that there was highly positive correlation of Lkn-1 and TNF-a level with serum creatinine in chronic renal failure patients.With the progress of glomeruli damage,the atrophy tubular and interstitial fibrosis,the levels of Lkn-1 and TNF-α were gradually increased.Conclusion Serum levels of Lkn-1 and TNF-α increased in patients with chronic glomerulonephritis,and they were positively related with the renal inflammation and fibrosis,which suggested that they may take part in chronic renal inflammation and fibrosis.Observeing the serum levels of these cytokines is helpful to estimate the renal fibrosis.

Immunomodulatory enzyme indoleamine 2, 3-dioxygenase (IDO) is one of the initial and rate-limiting enzymes involved in the catabolism of the essential amino acid tryptophan. Via catalysing tryptophan degradation, IDO suppresses adaptive T cell-mediated immunity and plays an important role in various forms of immune tolerance. Its role in T helper type 1 (Th1)-directed, cell-mediated crescentic glomerulonephritis (GN) is still unclear. Therefore, we investigated the activity and role of IDO in crescentic GN using a model of nephrotoxic serum nephritis (NTN), and IDO activity was inhibited by 1-methyl-tryptophan (1-MT) in vivo. Our results showed that activity of IDO, as determined by high performance liquid chromatography analysis of the kynurenine/tryptophan ratio, was increased markedly in the serum and renal tissue of NTN mice, and immunohistochemistry revealed that expression of IDO was up-regulated significantly in glomeruli and renal tubular epithelial cells during NTN. Treatment with 1-MT resulted in significantly exacerbated kidney disease with increased glomerular crescent formation, accumulation of CD4(+)T cells and macrophages in renal tissue, and augmented renal injury compared with phosphate-buffered saline-treated NTN mice, which was associated with enhanced Th1 responses and intrarenal cellular proliferation. These findings suggest that the development of NTN was regulated negatively by increased IDO activity, and IDO might play an important role in the pathogenesis of crescentic GN.
Objective To reach parameter on sterilization wrap of medical instrument. Methods The parameter of one hundred twenty sterilization wraps with size 30 cm×40 cm×60 cm and weight 20kg was tested by Belimed sterilizer. Results There are 155 masculine gender and 445 negative in 600 quick reading biological indicators of 120 sterilization wraps and 53 masculine in bacteria detection, 9 cases failure in 1243 indicator, 52 wet wraps. Conclusion The parameter was to take three times negative pressure pulse and five times pressure pulse, sterilization temperature 134℃, sterilization time 11min and dry time 11min, matching to sterilization request of Technical Standard For disinfection.
