Objective: To investigate the effects of microRNA-381-3p (miR-381-3p) and microRNA-193a (miR-193a) on the growth and apoptosis of human papillary thyroid carcinoma B-CPAP cells. Method: Sixty-two samples of thyroid carcinoma tissues and sixty-two samples of corresponding adjacent tissues were removed and collected from the selected patients with thyroid cancer receiving surgical resection. The expression levels of miR-381-3p and miR-193a in carcinoma tissues and adjacent tissues were detected using quantitative real-time reverse transcription polymerase chain reaction. Expression vectors of miR-381-3p and miR-193a were established and transfected by human papillary thyroid carcinoma B-CPAP cells, followed by cell culture. Cell proliferation, invasion and apoptosis were detected by cell counting kit-8 assay, transwell assay, and flow cytometric analysis. Results: The expression levels of both miR-381-3p and miR-193a in the thyroid carcinoma tissues were significantly lower than those in the adjacent tissues (both P<0.001). MiR-381-3p mimic group and miR-193a mimic group were both significantly lower than blank group or NC group in cell proliferation activity at 48 h and 72 h as well as in number of invaded cells (all P<0.001). The apoptosis rates of miR-381-3p mimic group and miR-193a mimic group were both significantly higher than those of NC group or blank group (all P<0.001). Conclusion: MiR-381-3p and miR-193a present low expression levels in thyroid carcinoma tissues. Overexpression of miR-381-3p and miR-193a inhibited proliferation and invasion of human papillary thyroid carcinoma B-CPAP cells and promote apoptosis of the cells, suggesting that miR-381-3p and miR-193a may be used as diagnostic markers and therapeutic targets for thyroid cancer.
目的 探讨高迁移率族蛋白1(HMGB1)和缺氧诱导因子1a(HIF1a)在乳头状甲状腺癌(PTC)中的表达及与临床病理特征的关系.方法 采用免疫组织化学法检测120对PTC(肿瘤组)及其相应癌旁组织(非癌组织组)中HMGB1与HIF1a的表达,比较两组HMGB1与HIF1a蛋白表达的差异,并分析其表达水平与PTC患者临床病理特征的关系.结果 PTC组织中HMGB1阳性表达率高于癌旁组织(P<0.05);PTC组织中HIF1a阳性表达率高于癌旁组织(P<0.05).PTC组织中HMGB1与HIF1a的表达呈正相关(P<0.05).HIF1a和HMGB1在不同肿瘤直径、肿瘤数量、淋巴结转移情况及临床分期比较,差异有统计学意义(P<0.05)结论 HMGB1、HIF1a与PTC增殖和转移相关,可能是PTC发生、发展的重要分子,进一步研究两者的关系可能为抑制肿瘤增殖、转移提供潜在的分子靶点.
Recent studies have confirmed that aberrant expression of protein phosphatase 1H (PPM1H) plays multiple roles in various types of cancers, but a potential role in papillary thyroid cancer (PTC) has not been studied. This study aimed to explore the clinical characteristics of PPM1H in PTC and its roles in the proliferation and invasion of PTC cells. Down-regulation of PPM1H expression was frequently detected in primary PTC tumor tissues compared with those in non-tumor tissues, and it was significantly associated with tumor size (P = 0.048), lymph node metastasis (P = 0.016), and clinical stage (P = 0.033). Knockdown of PPM1H in SW579 cells, a human PTC cell line, decreased E-cadherin level, while increasing expression of Vimentin. After PPM1H inhibition, SW579 cells exhibited morphological change from an epithelial cobblestone phenotype to an elongated fibroblastic phenotype. Wound healing and transwell assays were used to analyze the migratory and invasive ability of cells, and it was found that SW579 cells had increased migratory and invasive ability after PPM1H knockdown. Furthermore, MTT and colony formation assays showed that down-regulation of PPM1H promoted cell proliferation (P<0.05). These results suggest that low expression of PPM1H is associated with increased cancer cell proliferation and invasion and that PPM1H may act as a tumor suppressor in PTC.
Objective To discuss role of the rich sequence specific binding protein 1( special at rich sequence binding protein 1, SATB1)in initiating PI3K/Akt signaling pathway on the occurrence of thyroid cancer cells as well as invasion and metastasis. Methods The cul-tured thyroid cancer cells were divided into 4 groups:untreated group:in addition to DMEM without any treatment;LY294002 inhibited group:Cultured in DMEM with 10 μmol/L inhibitor LY294002;SATB1 inducer group:DMEM medium plus SATB1 antibody 200 μg/L;TSATB1 in-duced inhibition of group:DMEM medium plus SATB1L antibody 200μg/L and 10 μmol/L inhibitor LY294002. After 24 hours of culture acqui-sition cultivating base in thyroid carcinoma cells in serum by elase method to detect the expression of MMP9,IL-1 and IL-6 expression,on the cleavage of the thyroid cells using Western blotting were used to detect the expression of PI3K,Akt,p-Akt and PIP3,immunohistochemical de-tection cell SATB1 expression. Results The expression of SATB1 in different experimental groups was detected by immunohistochemical method. The expression of SATB1 was mainly in the cytoplasm of yellow,which was not expressed in cytoplasm. SATB1 expression was different in different treatment groups and the difference was statistically significant( P ﹤0. 05). The SATB1 induced agent histone SATB1 expression highest expres-sion of the lowest LY294002 inhibited group and the difference was statistically significant( P ﹤0. 05). Western blot results showed that different groups of PI3K,Akt,p-Akt and PIP3 expression was varied and the difference has statistical significance( P ﹤0. 05). The SATB1 induced a-gent group and the protein PI3K,Akt,p-Akt and PIP3 expression was the highest,the lowest was LY294002 inhibited group and the difference was statistically significant( P ﹤0. 05). Elase assays confirmed that different groups of MMP9,IL-1 and IL-6 expression was varied and the difference has statistical significance( P ﹤0. 05). SATB1 induced agent group and the protein PMMP9,IL-1 and IL-6 expression was the highest,the lowest was LY294002 inhibited group and the difference was statistically significant( P ﹤0. 05). Conclusion SATB1 is associated with invasion and metastasis of thyroid carcinoma;SATB1 has a role in the K PI3 / Akt signaling pathway,and SATB1 may be involved in the ex-pression of tumor microenvironment and inflammatory factors.
OBJECTIVES: The study explored the association between rs10069690C/T and rs2736100G/T of human telomerase reverse transcriptase (hTERT) gene, and the prognosis of thyroid cancer.METHODS: The study had 452 thyroid cancer patients recruited as case group who hospitalized in Jingzhou Central Hospital from January 2001 to June 2004 and 452 healthy people recruited as control group at the same area. The hTERT gene polymorphisms at rs10069690 C/T and rs2736100 G/T were tested by polymerase chain reaction-restriction fragment length polymorphism (PCR-RFLP) method. The association between patients' life quality and hTERT gene polymorphisms six months after surgery was evaluated based on the Cancer patients' quality of life index rating scale.RESULTS: There were statistical differences in genotype and allele frequencies of rs10069690 C/T between the case group and control group (both P < 0.05). An association between rs10069690C/T polymorphism and an increased risk of thyroid cancer was shown by logistic regression analysis (CT vs. CC, OR = 1.333, 95%CI = 1.006-1.766, P = 0.045; TT vs. CC, OR = 1.910, 95%CI = 1.084-3.367, P = 0.023; CT + TT vs. CC, OR = 2.246, 95%CI = 1.078-1.840, P = 0.006; T vs. C, OR = 1.376, 95%CI = 1.104-1.715, P = 0.004). Genotype frequency of rs2736100G/T between the two groups had no statistical differences (P > 0.05). After stratification according to age, T stage, tumor size and tumor node metastasis (TNM) stage, the distribution frequencies of CC genotype and CT + TT genotype of rs10069690C/T showed significant difference (P < 0.05). The life quality of patients with CC genotype was better than that of patients with CT TT genotype. The results of Cox regression model multifactor analysis showed that age, T stage, tumor size and rs10069690C/T were independent risk factors of thyroid cancer prognosis.CONCLUSIONS: hTERT gene polymorphism at rs10069690C/T is associated with the risk and prognosis of thyroid cancer, but hTERT gene polymorphism at rs2736100G/T is not.
目的 探讨腔内激光(endovenous laser treatment,EVLT)治疗下肢静脉曲张术后并发症的发生原因及其防治对策.方法 回顾性分析华中科技大学同济医学院附属荆州医院EVLT治疗下肢静脉曲张142例(195条肢体)的临床资料.结果 142例患者住院时间7 ~15(中位数11)d,一次治愈率97.4% (190/195),余3例(5条下肢)术后残留少量曲张静脉,二期在局部麻醉下行再次激光治疗或泡沫硬化疗法.98条下肢术后发生并发症,发生率50.3% (98/195).其中皮下淤血、血肿42条(21.5%);明显静脉炎反应34条(17.4%);隐神经损伤导致皮肤感觉异常26条(13.3%);皮肤灼伤9条(4.6%);下肢深静脉血栓形成1条(0.5%).经相应治疗后均痊愈.142例随访3 ~ 24(中位数10)个月,未见复发.结论 大隐静脉曲张术中应适度使用腔内激光治疗,制定个体化的治疗方案、联合泡沫硬化疗法及术后早期活动等措施可预防或减少术后并发症的发生.
Objective To observe special AT-rich sequence binding protein-1 (SATB1) expression in thyroid cancer and the effect of SATB1 on invasion of thyroid cancer cells.Methods The expression of SATB1 in 85 cases of thyroid cancer was detected by using immunohistochemistry.The correlation between SATB1 and clinicopathological features was analvzed by Chi-Squared test.The effect of SATB1 on invasion of SW579 cells was analyzed by using invasion assay.Results The positive expression of SATB1 in thyroid cancer was 68.2% (58/85).SATB1 was mainly expressed in nucleus of thyroid cancer cells.SATB1 expression was associated with lymph node metastasis and pathological stage (P < 0.05),but not associated with sex,age,tumor sizes and number of tumor nodules (P > 0.05).Invasion assay showed that the migrating number of empty vector transfected group and pEGFP1-SATB1 transfected group was respectively (220±10) and (416±15) (P<0.05).Conclusion The up-regulation of SATB1 in thyroid cancer has a close relationship to the invasion of thyroid cancer cells.
Objective To explore the pattern of lymph node metastasis of differentiated thyroid carcinoma and its clinical significanced.Methods Clinical and pathological data of 41 differentiated thyroid cancer patients who underwent radical thyroidectomy and modified radical neck dissection were collected and retrospectively analyze.Results There were 38 cases positive cervical lymph node metastasis in the 41 cases.Cervical lymph node metastasis was commonly seen in region Ⅳ(56.1%) and regions Ⅱ,Ⅲ,Ⅵ(46.3%),there was no significant difference among the four groups.There were few metastatic lymph nodes in region Ⅰ and Ⅴ(9.7% and 12.2%,respectively),which had significantly different compared with regions Ⅱ,Ⅲ,Ⅳ,Ⅵ.The sensitivity of ultrasonograph that identified cervical lymph node metastases was high(97.4%).Conclusion When positive cervical lymph node metastasis are considered in differentiated thyroid carcinoma,the patients deserve formal Ⅱ,Ⅲ,Ⅳ,Ⅵ regions cervical lymph node dissection.
【Objective】 To explore the function and clinical significance of Menin and cyclin-D1 in parathyroid adenoma.【Methods】 The expression of the Menin protein and cyclin-D1 were determined by immunohistochemistry in 4 cases of normal parathyroid tissue,6 cases of parathyroid proliferation tissue and 17 cases of parathyroid adenoma tissue.【Results】 The expression of Menin protein and cyclin-D1was significantly different between normal parathyroid and parathyroid adenoma(P 0.05),and was not significantly different between normal parathyroid and parathyroid proliferation(P 0.05).【Conclusion】 Firstly,the inactivation of Menin protein would be play an important role in the development of parathyroid tumor;Secondly,over-expression of cyclin-D1 is related to the occurrence of parathyroid tumor.
目的探讨根治性远端胃大部切除术后胃瘫发生的危险因素。方法对7年间收治的508例行根治性远端胃大部切除术患者的临床资料进行回顾性分析。以手术后是否发生胃瘫为因变量,进行单因素及多因素非条件Logistic逐步回归分析。结果 25例术后发生胃瘫,单因素分析显示16个因素中有9个与术后胃瘫发生有关。非条件Logistic逐步回归分析模型共筛出10个术后胃瘫的相关因素。结论术前幽门梗阻、毕Ⅱ式胃肠吻合方式、淋巴结清扫范围、术后血糖水平、术后腹腔感染、术后吻合口瘘、术后开始行肠内营养时间、术前情绪紧张为术后胃瘫的危险因素;而术前血红蛋白水平及术后总蛋白水平为术后胃瘫的保护性因素。
目的 探讨原发性甲状旁腺机能亢进症(primary lhyperparathyroidism,PHPT)的早期诊断与合理治疗.方法 对近10年来两院收治的23例PHPT患者的临床资料进行回顾性分析.结果 女15例,男8例;年龄16~64岁,患者临床表现多样,病程迁延,术前均采用颈部彩超+颈部增强CT或MRI检查+血钙+血PTH检查的综合分析诊断模式.21例行单侧甲状旁腺探查术,2例行双侧甲状旁腺探查术,22例术后确诊为PHPT并治愈,1例永久性甲状旁腺机能减退.结论 对可疑PHPT患者可采用颈部彩超+颈部增强CT或MRI检查+血钙+血PTH检查的综合分析诊断模式可帮助早期诊断.单侧甲状旁腺探查术是PHPT有效的治疗方法.
我院近4年来收治的419例甲状腺功能亢进患者中26例术前针吸细胞学检查或/和术中快速冰冻切片提示、术后病理检查证实为合并甲状腺微小癌,现总结报告如下.

本实验通过免疫组化SP法研究凋亡抑制因子livin在肝癌组织中的表达情况,探讨其与原发性肝细胞癌(PLC)TNM分期、复发之间的关系.
多发性内分泌肿瘤1型(the multiple endocrine neoplasia type 1,MEN1)是一种具有染色体显性遗传倾向的肿瘤综合征.基于对MEN1患者肿瘤的杂合性丢失的研究和对患者家族进行基因连锁分析确定了MEN1基因位于染色体11q13[1],它是一种肿瘤抑制基因,编码蛋白质Menin.目前,MEN1基因的突变被认为是导致甲状旁腺肿瘤发生的几个主要因素之一.在动物实验中,使小鼠细胞Menin杂合性失活,结果这些小鼠多数发生了甲状旁腺肿瘤,在后期还发生了与多发性内分泌肿瘤1型类似的表现[2,3].尽管一些事实已表明Menin蛋白与基因的转录调控,细胞周期的调控和一些蛋白协同分子的作用有关,但是Menin的失活导致甲状旁腺肿瘤发生的具体机制以及Menin蛋白的生理功能还需要不断的探讨.
Objective To explore the role and interaction of Menin protein and TGF-β in parathyroid adnomas.Methods The expression of Menin protein and TGF-β was detected by immunohistochemistry in 7 cases of normal parathyroid tissue,12 cases of parathyroid proliferation tissue and 43 cases of parathyroid adnoma tissue.Results There was significant difference in the expression of Menin protein between normal parathyroid and parathyroid adnoma(P0.05),but there was no significant difference between normal parathyroid and parathyroid proliferation(P0.05).There was no significant difference in the expression of TGF-β among the three groups(P0.05).But the expression of Menin was significantly related to that of TGF-β in the adnoma group(P0.05).Conclusion The inactivation of Menin protein may play an important role in the development of parathyroid tumor;Menin may be a crucial protein in TGF-β pathway.
