Brassica rapa L. ssp. chinensis (L.) Hanelt is an important vegetable in eastern Asia. Its leaf area is one of main factor of influencing on the yield of individual plant. To reveal the mechanism of the leaf expansion, the joint segregation analysis of multiple generations (P1, P2, F1, B1, B2 and F2) was used to analyze the genetics of leaf expansion trait in combination SW-13×L-118 of Brassica rapa L. ssp. chinensis (L.) Hanelt. In this paper, the results showed that the leaf expansion trait was controlled by one pair of additive-dominant major genes plus additive-dominant-epistatic polygene (D model). The additive effect of the major gene was -2.3061, the dominant effect was -1.4525. The heritability of major gene of leaf expansion was 2.56% in B1 generation, 1.55% in B2 generation and 12.12% in F2 generation. The heritabilites of the polygene were 54.04% in B1 generation, 25.11% in B2 generation, and 62.60% in F2 generation, respectively. It appeared that the polygene effects should be given prior to the improvement of leaf expansion trait of combination SW-13×L-118, and the selection was effective in later generation, and the negative additive effect and negative dominant effect of major gene were noticed.
Objective:To prepare monoclonal antibody of Leghorn chicken FOXL2 protein.Methods: First,bioinformatics methods were used to predict its most possible epitopes,then polypeptides were synthesised,purity were analysised by liquid chromatography,coupled with KLH.Later,the mice were immunized by the polypeptides which coupled with KLH.After fused between the splenic cell of immunized mice and NS0 myeloma cell,the indirect ELISA and Western blot were used to detect whether the hybridoma cell can secrete antibody against polypeptide and FOXL2 protein.Last,the general nature,binding curve and IC50 of monoclonal antibody were measured.Results: It was found that the purity of artificial polypeptides were all > 95%.After cell fusion,its fusion rate was 75%,and one cell lines were screened by indirect ELISA and Westen blot;cell line supernatant titer was 1/128,the ascites titer was 1/256,IgG1 type,had 102 cell chromosome,ascitic fluid protein content 1.576 g/L;its affinity constant was 2.12×106 L/mol,IC50 value was 0.082 47 μg/L.Conclusion: We successfully prepared the FOXL2 protein monoclonal antibody,this work should helped for a deep research to FOXL2.
A sequence-independent PCR amplification method was used to identify viral nucleicacids in the small intestine and liver specimens of 21 chickens presenting with symptoms of acute viral infection which were reared together with ducks. One of all PCR products of length of 1596 bp was detected, and was cloned into pGEM-T vector for sequencing and analysis. Results showed that the identity of the nucleotide sequence of obtained gene between ZZS10 isolate and other DEV reference strains were from 99% - 100%, the identity of the deduced amino acid sequence of obtained gene of ZZS10 isolate was the same as anatid herpesvirus. The present study indicated that ZZSIO isolate might be a member of Alphaherpesvirinae.
The gene FOXL2 is a transcription factor that can effect vertebrate ovaries differentiation at the early time,which through Aromatase and SOX9 to make an indirect influence at the vertebrate sex determination.In this review,the structure and biological function of the FOXL2 gene and its relationship to sex determination of vertebrate were summarized.

Objective:To obtain more information about FOXL2 protein from Leghorn chicken.Method:Bioinformatics approach were applied to analyze FOXL2 protein's structures,composition,functions,physical and chemical properties,multiple sequence alignment with its homologous sequences,B-cell epitopes.Result:It was found that the highest content of amino acid was Pro,the molecular mass is 33504.8,the half-life is 30h,the theoreticaI isoelectric is 9.04,maybe play a role in the cell nucleus,the most likely function of this protein was transcription and transcription regulation,has the highest homology with Gallus gallus,the most likely epitope is 37-49,82-91,133-147.Conclusion:We obtained more biological information about FOXL2 protein by bioinformatic analysis,which is very useful for further research.
Uniform experimental design was applied to study the effects of using high pressure and the treatment time on water holding capacity of pork homogenate gelation.The results showed that soluble protein concentration of homogenate and water holding capacity rate of gelation were increased while the pressure and the treatment time decreased.The trend of the dehydration rate of pork homogenate gelation was varied by different conditions.With longer high pressure treatment,the dehydration rate was increased along with the increasing of the pressure;but at the shorter treatment time,the result was inversed.If the pressure is fixed,the dehydration rate was increased first and then decreased along with treatment time.
TbpA is a highly conserved transmembrane protein that may serve as a channel for transport of iron across the outer membrane, which is required for transferrin utilization and is responsible for removing the iron from transferrin and for transporting iron across the outer membrane in a TonB-dependent manner. In the present study, a 3D homology modeling of TbpA from Actinobacillus pleuropneumoniae (App) L20 strain, based upon the Crystal structure of the hemehemoglobin outer membrane transporter ShuA from Shigella dysenteriae (PDB code: 3fhh) was performed using bioinformatics tools, as no experimental 3D structures. The program VERIFY 3D assessed the quality of the predicted structure of TbpA with acceptable scores. All the results converged to the fact that the predicted 3-Dimenrsional structure of TbpA is of good quality with acceptable scores.
Quadratic general rotary design was adopted to study effects of pH,sodium chloride concentration and heating temperature on heat-induced gelation properties of pork homogenate from hind leg.The results showed that protein concentration of homogenate increased with the increasing of pH and sodium chloride concentration within a certain range.There were interactions between pH and sodium chloride concentration on purge loss,water holding capacity and gelation strength.Purge loss declined with the increasing of pH and sodium chloride concentration while it increased with the increase of heating temperature.To some extent,the correlation of water holding capacity of gelation with pH,sodium chloride concentration and heating temperature were positively correlated.At different heating temperatures,the effect of pH and sodium chloride concentration on gelation strength were similar.The optimization calculation showed that the optimum condition of the formation of gelation was pH6.33,sodium chloride concentration 0.67mol/L,heating temperature of 75℃.
The FoxO3 belongs to the O subclass of the forkhead family. FoxO3 is a tumour suppressor, a master regulator and suppressor of primordial follicle activation. We predicted FoxO3 cDNA sequence of Gallus gallus by in silico cloning, and analyzed its coding protein by bioinformatics methods. Results showed that the FoxO3 cDNA encoded a protein with 486 amino acid residues. The predicted FoxO3 protein with a Forkhead domain was not a secretory protein, and was located in the nucleus. The FoxO3 protein had highly homology with that of other species. This study might provide a basis for future studies on the FoxO3 gene of Gallus gallus and functions of its encoded protein.

To investigate B cell epitopes on the VP3 of infectious bursal disease virus (IBDV), the VP3 gene of IBDV were subcloned into the pET28a vector to construct expression vector of pETVP3. The recombinant VP3 (rVP3) was expressed in E. coli BL21(DE3) by inducing with IPTG. The expressed VP3 protein was recognized specifically by anti-IBDV antisera in western blot assays. According to antigenic structure of IBDV VP3, overlapping peptides which cover the full sequence of VP3 were synthesized and Conjugated to carrier Protein BSA to prepare conjugated antigens. Their reactivities with anti-IBDV monoclonal antibodies (MAb) or polyclonal antibody were detected by peptide-ELISA and Dot-ELISA The results showed that there were 2 linear epitopes recognized by neutralizing Mabs on the VP3, 728PRDWDRLPYLNL739 and 982PKPKPKPNAPTQ993, which can also react with IBDV antiserum. Dot-ELISA confirmed that there were still 4 linear epitopes recognized by polyclonal antibody on the VP3, 818LANAPQAGSKSQRA831, 851QREKDTRISKK METMGIYFATP872, 876ALNGHRGPSPGQLKYWQN TREI897 and 961QMKDLLLTAMEMK973. It is helpful for the developmentof IBD epitope vaccine.

In this paper, the comparative genomics overview of this family was described.
Pure pineapple juice was adopted to explore its quality characteristic during frozen storage.The results showed that: during frozen storage,the value of pH,L,a and sugar content decreased gradually significantly;and the value of b increased significantly;the sensory score of colour and fragrance decreased significantly,and the sensory score of flavor increased significantly,the total sensory score had no distinct trend.Frozen processing and storage was an appropriate manufacturing method for pure pineapple juice.
Recently, largescale, high-density single-nucleotide polymorphism (SNP) marker information has become available. However, the simple relation was not enough for describing the relation between markers and genotype value, and the genetic diversity should be carefully monitored as genomic selection for quantitative traits as a routine technology for animal genetic improvement. In this paper, back-propagation neural network is used to simulate and predict the genotype values, and the different gene effects were used to discuss the influences on estimating the polygenic genotype value. The results showed that after phenotype value being normalized, optimization network could be established for predicting the phenotype value without fearing that the gene effect is too large. If the number of hidden neurons is large enough, the stability of back-propagation artificial neural network established for predicting phenotype value is very well, the gene effect could not affected the precise of optimum neural network for estimating the animal phenotype, the optimum neural network could be selected for predicting the phenotype values of quantitative traits controlled by genes with small or large effects.
Uniform design was adopted, and two factors, i.e., frozen at different time postmortem with six levels, thawing at different thawing rate with three levels, were considered. Water holding capacity under pressure (WHC), thawing loss (TL) and production rate (PR) were measured to study the different effects of the two factors considered. The results showed: All the two factors considered had significant effects on WHC, TL and PR, and the effects of the two factors on them were nonlinear, there were interactions between the two factors considered. The optimization calculation showed that the later the frozen time postmortem and the smaller the thawing rate was, the higher the production rate of Chinese sausage was.

A Genomic island (GI) often encodes an integrase gene that specifies the island's position in the host genome, which is part of a genome that has evidence of horizontal origins. GI can code for many functions, which can be involved in symbiosis or pathogenesis, and may help an organism's adaptation. Many sub classes of GIs exist that are based on the function that they confer. GIs are frequently associated with microbial adaptations that are of medical and environmental interest, and they have had a substantial impact on bacterial evolution. Therefore, there is growing interest in efficiently identifying GIs in newly sequenced bacterial genomes. We compared and analyzed GIs of three Actinobacillus pleuropneumoniae strains by comparative genomic methods and highlighted the difference between them, which provided a reference for further research on the biological characteristics of Actinobacillus pleuropneumoniae.
NMR was used to investigate the chemical shift graphs of fresh pork at different time postmortem, and at the same time the ice crystals of meat freezing at different time postmortem were researched with SEM. The results showed there were significant (P<0.05)correlation between distribution and size of ice crytals freezing at different time postmortem and the chemical shift graphs of fresh pork at different time postmortem different. Using chemical shift graphs of meat could forecast whether the meat was suitable for frozen processing, and also could forecast the water holding capacity of it.
The PCR products of length of 414 bp and 450 bp were derived from air of new cell culture room by using random PCR,and were cloned into pGEM-T vector for sequencing and analysis.Results showed that the identity of the nucleotide sequence of obtained genes between HN09 and HN10 strains and other Cupriavidus metallidurans reference strains were from 79%-83% and 79%-84%,respectively,and the identity of the deduced amino acid sequences of obtained genes between HN09,HN10 strains and other Cupriavidus metallidurans reference strains were from 86.4%-89.1%.The present study indicated that these two strains may belong to different subtypes of Cupriavidus metallidurans.
To overcome the obstacle of the fascinating relation in predicting animal phenotype value, we have developed a neural network model to detect the complex non-linear relationships between the genotypes and phenotypes and the possible interactions that cannot be expressed with equations. In this paper, back-propagation neural network is used to discuss the influences of different allele frequencies on estimating the polygenic phenotype value. To ensure the precision of prediction, normalization was needed to train the prediction model. The results show that back-propagation artificial neural networks can be used to predict the phenotype value and perform very well in allele frequency from 0.2 to 0.8, when the allele frequency is very small (less than 0.2) or big (more than 0.8); however, the prediction model was not reliable and the predicted value should be carefully tested.
