目的:比较同一促排周期来源的两次冻融胚胎移植,两次均为单胎妊娠分娩临床结局的差异.方法:回顾分析2011年至2018年于中国科学技术大学附属第一医院生殖医学中心接受助孕治疗,移植同一促排周期来源的冻胚后两次单胎妊娠分娩产妇101例.按胚胎冻存年限分为0~3年组和4~9年组,按孕妇年龄分为≤30岁、30~35岁、≥35岁组,分层比较孕妇的一般情况,包括平均年龄、孕囊数、治疗方案、受精方式.同时比较一胎和二胎孕期、胎儿平均体重、剖宫产率、低体重儿率、巨大儿率、正常体重儿率、出生婴儿性别.结果:在产妇年龄、冷冻胚胎保存年限的分层研究中,孕期、胎儿平均体重等评价指标差异无统计学意义(P>0.05).结论:同一促排周期获得胚胎冷冻保存,间隔数年的两次冻融胚胎移植,两次单胎临床结局无显著差异.
Objective:To evaluate the safety and effectiveness of The Stork OTC conception aid kit for the treatment of male infertility compared with natural sexual intercourse.Methods:A multi-center, prospective, open, self-controlled, randomly crossed trial was performed in 57 infertility couples who were unable to naturally conceive, and were selected in Shanghai General Hospital, Shanghai Jiao Tong University, Reproductive Medicine Center,Tongji Medical College, Huazhong University of Science and Technology and Anhui Provincial Hospital from July 2017 to September 2018. Patients were required to participate both the conception aid kit and natural sexual intercourse in two consecutive periods of ovulation. A random envelope method was used to determine the application sequences of two methods. Twenty-nine couples were treated with the conception aid kit in the first month of ovulation, and applied the natural sexual intercourse in the second month of ovulation as group A. Twenty-eight couples were applied the natural sexual intercourse in the first month of ovulation, and were treated with the conception aid kit in the second month of ovulation as group B. Test results collected from all couples who used the conception aid kit were categorized as the test group data. Meanwhile, test results out of those couples who completed natural sexual intercourse were collected as control group data. Compared with its self-controlled natural sexual intercourse two months before and after, the effectiveness and safety of the conception aid kit was verified by general vaginal examination, routine leucorrhea examination, motile sperm score from post coital test (PCT), pregnancy rate comparison and other methods.Results:The trial was conducted at three clinical trial centers. A total of 57 infertility couples were enrolled. A total of 54 couples were validated eventually including 26 couples in group A and 28 infertility couples in group B. Effectiveness was evaluated. The PCT results showed that the sperm score of test group was 6.11±3.00, and the score of the sperm score of control group was 1.22±0.79. The motile sperm score of test group was 5 times ( P<0.001) than that in control group. In the follow-up, 7 (13.0%) of the couples successfully achieved pregnancy with the conception aid kit, 6 (85.7%) of them had healthy offspring, 1 (14.3%) had spontaneous abortion. No couples were pregnant after natural sexual intercourse. Safety was evaluated. The general examination of the vagina and the routine tests of the vaginal leukorrhea showed no difference between test group and control group. All 54 effective couples who completed the test can successfully use the conception aid kid to place the seminal vesicles containing semen and remove them with the attached pulling thread on time without obvious discomfort. Conclusion:The Stork OTC conception aid kits can effectively treat infertility, significantly increase the number of forward-moving sperm entering the cervix, and significantly increase the pregnancy rate. It is safe, non-invasive, simple, can be easily operate by patients themselves, and meets the needs of clinical applications.
目的 研究辅助生殖技术后胚胎停育患者绒毛染色体同年龄、胚停史及自身免疫性抗体之间的关系.方法 回顾性分析2016年2月~2018年8月进行体外受精-胚胎移植(in vitro fertilization and embyro transplantation,IVF-ET)助孕术后孕早期胚胎停育行绒毛染色体检查的患者113例.同时检测患者抗核抗体(anti-nuclear antibody,ANA)、抗心磷脂抗体(anti-cardiolipin antibody,ACA)抗甲状腺过氧化物酶抗体(anti-thyroperoxidase,ATA)、抗甲状腺球蛋白抗体(anti-thyroglobulin,ATG).结果 ①113例患者染色体正常64例,染色体异常者49例;②35岁以上患者染色体异常率显著高于35岁以下患者(67.57%VS31.58%,P<0.0001);③既往有无胚停史、胚停次数与绒毛染色体异常之间并无显著联系;④染色体正常组中测出ACA、ANA、ATA/ATG阳性率虽高于染色体异常组,但差异不显著.结论 随着年龄增加,染色体异常比例显著增高;既往有无胚停史与染色体是否异常之间并无显著联系.
Objective: To explore the influence of P450 aromatase inhibitor letrozole on endogenous androgen synthesis, testicular interstitial cell proliferation and spermatogenic function. Methods: Forty SPF-grade male Kunming mice were randomly divided into control group and experimental group. Clysis of 0.01 mg/kg letrozole was conducted on all rats for three days in the experimental group and eyeball vein blood collection was conducted 24 h, 4 W and 8 W after withdrawal. ELISA method was adopted to test the levels of testosterone (T) and estradiol (E2) and then testicular histological change was observed through light microscope and electron microscope; epididymal development was observed and testis coefficient and epididymis coefficient (testicular or epididymal weight on both sides/body weight) of mice were calculated; sperm density was calculated and apoptosis of testicular interstitial cells was tested with the method of flow cytometry. Proliferation capacity of testicular interstitial cells was tested through CCK8. Results: Compared with the control group, the levels of testosterone (T) and estradiol (E2) in serum of mice was obviously reduced by letrozole 8 W upon medication (both P<0.05) and there was no obvious difference between 24 h and 4 W upon medication (both P> 0.05). Compared with the control group, testicular and epididymal coefficients were both obviously reduced by letrozole 4 W and 8 W upon medication (both P< 0.05) and there was damage to testicular histomorphology to different degrees below electron microscope in the experimental group; sperm density of testicular tissue was significantly reduced by letrozole 4 W and 8 W upon medication (both P< 0.05). Compared with the control group, obvious decrease of sperm density and significant increase of cell apoptosis in testicular tissue were observed by applying letrozole for 4 W and 8 W of medication (all P< 0.05). Conclusion: Letrozole could inhibit proliferation of testicular interstitial cells, reduce testis coefficient and epididymis coefficient, decrease sperm density and testicular interstitial cells apoptosis through reducing synthesis of endogenous estrogen in the mouse and making testosterone and estradiol level lower.
Puerarin, a well-known isoflavone-C-glucosideisolated from Pueraria radix, has been demonstrated the anticancer activity in animal models as well as proliferation inhibition and apoptosis induction in a variety of cancer cell lines in vitro. However, the effect and mechanisms of puerarin on ovarian cancer (OC) have not been well understood. This study aimed to test whether puerarin could inhibit growth of ovarian cancer cells and reveal its underlying molecular mechanism. Cells viability was evaluated using cell counting Kit-8 in SKOV-3 and OVCAR-3 cells. Apoptosis of ovarian cancer cells was analyzed by flow cytometry. Bcl-2 levels were analyzed by Western blot and microRNAs levels were determined by real-time RT-PCR. Our study showed that puerarin inhibited SKOV-3 and OVCAR-3 cells growth in a concentration-dependent manner. Puerarin induced apoptosis by promoting expression of miR-125b, and then inhibiting expression of Bcl-2, a known target for miR-125b. Moreover, knockdown of miR-125b could reverse the reduction of cell viability induced by puerarin in SKOV-3 and OVCAR-3 cells. Taken together, our study unraveled the ability of puerarin to suppress ovarian cancer growth and elucidated the participation of miR-125b in the anti-cancer action of puerarin. Our findings suggest that puerarin can be considered to be a promising anticancer agent for ovarian cancer.
Objective To explore the relationship between calcium concentration and sperm activity.Methods Calcium imaging system was used to observe the calcium signal of sperm in the experimental group and control mice. Results The results showed that the concentration of calcium in the sperms of rats decreased because of smoking, leading to a reduced sperm motility.Conclusion Somking has a detrimental affect on sperm activity and may lead to a harmful reproductive injury.
Objective To provide reliable experimental data for molecular mechanism research on male reproductive impairment by establishing the mice model of smoking to detect the expression of specific cation channel CatSper mRNA and protein in sperm, and analyze its relation with sperm motility. Methods Mice model of smoking was established, sperm samples were separated by Percoll discontinuous density gradients and sperm motility was detected, mice sperm CatSper mRNA was determined by RT-PCR, the distribution of CatSper protein was observed by immunocytochemical technique , CatSper protein level was detected by western blotting. Results In normal control group, mild smoking group, moderate smoking group, and severe smoking group, the proportion of sperm motility (a + b) grade were (87.6 ± 16.2)%vs. (72.9 ± 13.0)%vs. (68.1 ± 16.0)%vs. (44.6 ± 15.0)%(P<0.05), the relative contents of CatSper mRNA were 0.915 ± 0.202 vs. 0.712 ± 0.121 vs. 0.522 ± 0.126 vs. 0.258 ± 0.177 (P<0.05), the relative contents of CatSper protein were 0.882 ± 0.208 vs. 0.693 ± 0.202 vs. 0.471 ± 0.251 vs. 0.263 ± 0.198 (P < 0.05). CatSper protein expression was depressed by smoking in testis testified by immunocytochemical technique, and the results indicated significant negative correlation between CatSper mRNA/protein and the proportion of sperm motility (a+ b) grade. Conclusions The expression of cation channel CatSper was significant inhibited by smoking with a positive correlation with sperm motility , which suggest that the lower expression of CatSper may be involved in the molecular mechanism of male reproductions impairment by smoking.
Objective Investigate the relation between severe oligospermia and chromosome translocation,chromosome polymorphism.Methods In 612 patient with severe oligospermia,peripheral blood lymphocyte culture were set up for G-banding and karyotype analysis.30-60 metaphase were counted for each patient,karyotype were analysied in 3-6 metaphase according to ISCN1995 standard.Results In all 612 cases,13 cases showed autosomal translocation,and chromosomal polymorphism in 8 cases.Conclusion Autosomal abnormalities could result in oligospermia.
