近年来,肠道微生物与畜禽生产性状相关性研究成为动物养殖领域的研究热点.本研究探讨了不同性别河流型水牛的肠道微生物种群结构的异同,研究结果表明,河流型水牛肠道微生物主要以Firmicutes、Eur-yarchaeota、Spirochaetes和Proteobacteria为主要菌门,且不同性别的水牛肠道微生物在门水平的种群结构相似,但是各菌门在两组样本中的分布存在一定差异,公水牛样本中Verrucomicrobia丰度显著高于母水牛组的.在属水平,母水牛组样本中Peptostreptococcaceae noname和Lysinibacillus的丰度显著高于公水牛组的,而Streptococcus和Klebsiella的丰度显著低于公水牛组的.两组样品在种水平上母水牛组与公水牛组中Pepto-streptococcaceae noname unclassified、Lysinibacillus unclassified、Methanobrevibacter smithii、Streptococcus infan-tarius和Klebsiella pneumoniae等5种物种的相对丰度存在显著差异(P<0.05).本研究对不同性别河流型水牛的肠道微生物的种群结构进行初步的研究,为研究水牛肠道菌群结构与生产性状的相关性奠定了工作基础.

In a world of globally integrated financial markets and high capital mobility, how to collect effective taxation of cross-border income has become problematic under international tax practices. A feasible method to solve the puzzle is to collect withholding taxes at source. This paper compares Chinese withholding taxation with other countries in the world from the aspects of tax base, taxpayers and tax rates. With such comparison analysis, it provides some helpful suggestions to improve Chinese withholding tax system.
Objective To investigate the effect of TNF-α on the expression of Notch1 in Raji cell line. Methods The expression level of Notch1 mRNA and protein in Raji cells treated with TNF-α was detected by RT-PCR and flow cytometry respectively. Results The expression of Notch1 mRNA was up-regulated in a dose-dependent manner in Raji cells treated by TNF-α (P<0.01). Raji cells treated with TNF-a had a higher fluorescence intensity of Notch1 protein as compared with untreated cells (P<0.01). Conclusion Notch1 protein was expressed in Raji cells and the expression of Notch1 protein was increased after TNF-α treatment, suggesting that activated Notch1 signaling pathway plays an important role in pathogenesis of lymphoma and it might be a potential new target for treatment.

OBJECTIVE To investigate the effect of curcumin and tumour necrosis factor(TNF)-α on the expression of vascular endothelial growth factor in Raji cells and their effect on angiogenesis in human umbilical vein endothelial cells(HUVECs)-derived cell line(ECV304).METHODS VEGF secreted by Raji cell line was determined by ELISA.Angiogenesis was tested by network formation of endothelial cells on Matrigel.Levels of VEGF mRNA in Raji cells and Notch1 mRNA in ECV304 cells were measured by RT-PCR.RESULTS The content of VEGF secreted by Raji cells was increased by TNF-α and suppressed by curcumin(P0.01).The VEGF mRNA expression,mainly VEGF165 and VEGF121 in Raji cells(containing 165 and 121 amino acid residues,respectively) was detected in all fractions.TNF-α augmented the expression of VEGF165 and VEGF121 mRNA and curcumin reduced the expression(P0.01).No network and cord were formed in control and curcumin group.There was tube formation on matrigel in the supernatants of Raji culture group and the supernatants groups treated by VEGF and TNF-α in Raji cell.Notch1 mRNA was detected in ECV304 cell line but no difference in VEGF group compared with control(P0.05).CONCLUSION The expression of VEGF in Raji cells was increased by TNF-α and suppressed by curcumin.VEGF and TNF-α can induce angiogenesis and curcumin can inhibit angiogenesis in ECV304 cells.
AIM: To investigate the effect of triptolide on the expression of vascular endothelial growth factor (VEGF) in Raji cell line and on angiogenesis in cell line (ECV304). METHODS: The experiment was performed at the Molecular Biology Laboratory of Public Health Constitute, Tongji Medical College, Huazhong University of Science and Technology from August 2005 to February 2006. ①The antiproliferative effects of triptolide (3.125, 6.25, 12.5, 25 and 50 nmol/L) against Raji cells were determined by the MTT method at 12, 24, 36, 48, 60 and 72 hours. ②After 24 hours incubations treated by different concentrations of triptolide (12.5, 25, 50 nmol/L), the mRNA level of VEGF by Raji cells was measured by reverse transcription-polymerase chain reaction (RT-PCR). ③After 24 hours treatment with 25 nmol/L triptolide, VEGF concentrations in cell culture supernatants were determined by enzyme-labeled immunosorbent assay (ELISA). ④ Effect of triptolide on angiopoiesis of ECV304 cells in matrigel was detected by angiopoiesis test. RESULTS: ①The triptolide significantly inhibited the proliferation of Raji cells in time- and dose-dependent manner. IC50 value was 25 mmol/L. ②The mRNA expression of VEGF165 and VEGF121 (containing 165 and 121 amino acid residues, respectively) was detected in all groups. Triptolide reduced the mRNA expression of VEGF165 and VEGF121 in a dose-dependent manner. ③ Triptolide significantly suppressed VEGF expression in Raji cells (P < 0.01). ④There was blood vessel formation on matrigel in the supernatants from Raji culture group and VEGF group. No network and cord formed were seen in control and triptolide groups. CONCLUSION: Triptolide could inhibit the proliferation of Raji cells in time- and dose-dependent manners as well as the expressions of VEGF in Raji cells and angiogenesis in ECV304 cells.
The financial crisis in Asia, beginning with pressures on the Thai baht and spreading to other countries in the region, has intensified the interest in impulse-contagion mechanism of financial crisis. In this article, first we discuss speculative attack, which is the main form of external impact to financial crisis. Then we construct the time and spatial contagion frame of financial crisis. Finally we analyze the general contagion mechanism of financial crisis systematically.
OBJECTIVE To investigate the effect of trichostatin A,one kind of HDAC inhibitors,on expression of IκB-α induced by TNF-α and activation of NF-κB in U937 cells.METHODS U937 cells were treated with TSA,TNF-α respectively or with both,the protein expression and subcellular localization of NF-κB/p65 were observed by western blot and fluorescence microscopy respectively;the expression and degradation of IκB-α were investigated through by flow cytometry and western blot.RESULTS ①Under fluorescence microscopy,NF-κB/p65 was only localized in cytoplasm of empty control and group-treated with TSA for 45 min,p65 was localized in nucleus of U937 cells treated with TSA and TNF-α,while it was expressed in both cytoplasm and nucleus of cells treated with TNF-α.Western blot also showed the same result:TNF-α-treated group cells nuclears expressed much more p65 protein than those treated with TNF-α plus TSA for 45 min.②After being treated with TSA and TNF-α for 45 min,the protein expression of IκBα in U937 cells detected by flow cytometry was stronger than those only treated with TNF-α(P<0.05).Through Western blot,it was found that IκB-α expression was fewer in TNF-α-treated group compared to control;which was obviously enhanced in group treated with TSA plus TNF-α than that treated with TNF-α for 45 min,but it was no differences between them after being treated for 2 h.CONCLUSION TSA can partially inhibit degradation of IκB-α induced by TNF-α and NF-κB activation in U937 cells.TSA can work through NFκB signal pathway against tumor,which can be a target of TSA for cancer therapy.

The study was purposed to investigate the effect of deguelin on expression of nucleoporin 98 (nup98) in leukemia K562 cells. MTT assay was used to assess the effects of deguelin on cell proliferation. FCM and RT-PCR were used to analyze the changes of nup98 mRNA and protein in K562 cells after treating with deguelin. The results showed that deguelin inhibited the proliferation of K562 cells in a time- and dose-dependent manner; mean fluorescence intensity of nup98 in blank group (34.22 +/- 1.63) was significantly higher than that in control group (2.83 +/- 0.02, P < 0.01), 10 nmol/L deguelin could significantly inhibit the expression of nup98 protein; 10 nmol/L could not inhibit the expression of nup98 mRNA, 20, 40, 80, 160 nmol/L deguelin could significantly inhibit the expression of nup98 mRNA in a dose-dependant manner. It is concluded that deguelin inhibits the proliferation of K562 cell through inhibiting the expression of nup98, and may be considered as a new target for therapy of acute leukemia.
目的:探讨鱼藤素对DNA拓扑异构酶Topo Ⅰ表达的影响,进一步明确鱼藤素抑制肿瘤细胞增殖的作用机制.方法:流式细胞术及RT-PCR法分析鱼藤素作用K562细胞后Topo Ⅰ蛋白及mRNA含量的变化.结果:流式细胞术分析结果发现Topo Ⅰ蛋白的平均荧光强度随着鱼藤素作用浓度的增高而逐渐减少,与空白对照组比较有显著差异(P<0.05);RT-PCR半定量检测结果显示,鱼藤素作用K562细胞引起Topo Ⅰ mRNA含量减少,并与其浓度正相关.结论:鱼藤素可通过抑制肿瘤细胞内Topo Ⅰ而发挥作用.
Early warning systems help detect financial vulnerabilities and prevent financial crises. This paper establishes a new Early Warning System(EWS)for predicting financial crises based on P-S method and conducts empirical researches with the data of China and those countries that broke out financial crisis in 1997 like Thailand, Korea, Indonesia, Malaysia from 1992 to 1996.
