为了建立青蒿的 SRAP 最佳扩增体系,并筛选出 SRAP 多态性引物,本研究以青蒿叶片 DNA 为模板,采用正交试验设计,以 Mg 2 + 、 dNTP Mix 、 Taq DNA 聚合酶、引物和 DNA 模板 5 种因素 5 个水平,对青蒿 SRAP 反应体系进行研究。结果表明,青蒿 SRAP-PCR 最佳反应体系为:引物 0.6 μ mol / L 、 Mg 2 + 2.0 mmol / L 、模板 DNA 5.1 ng 、 Taq DNA 聚合酶 2.0 U 、 dNTPs 0.25 mmol / L ,总体积为 25 μ L 。各因素对扩增反应均有不同影响,其中引物浓度的影响最大, dNTPs 的影响最小。运用该体系对不同种质资源的青蒿进行验证,证明该体系稳定可靠,并在 30 个引物组合中筛选出了 25 对扩增条带清晰,多态性丰富的引物组合。这一结论为今后利用 SRAP 标记技术进行青蒿分子遗传学研究提供了科学依据。
旨在为树莓叶黄酮的开发应用提供数据参考,通过正交试验与响应曲面法设计改良乙醇提取工艺,并通过HPLC建立树莓叶黄酮的色谱分离条件为:Agilent ZorBax SB色谱柱,流动相为1 mg/mL乙酸-乙腈=72.5:27.5,等度分离,柱温30℃,检测波长:257 nm,完成树莓叶黄酮的初步分离,以及黄酮中芦丁含量的准确测定和验证.最佳提取工艺为:646 mL/mL乙醇,料液比1:12.5,41.9℃加热回流173.3 min,最佳工艺所得提取率1.127%,响应曲面法最佳工艺提取率略高于正交试验法,且其工艺条件均较优于正交试验所得工艺,可为后期研发树莓叶芦丁兽用中药制剂提供理论参考.
利用12C6+重离子束对黄花蒿干种子辐照后,利用发芽率、根长、下胚轴长和株高4个指标研究不同剂量12C6+离子束辐照对黄花蒿的生物学效应。结果表明,随着辐照剂量的增加,黄花蒿的存活率和根长都呈现逐渐降低的趋势,而下胚轴长先上升后下降,株高总体呈现下降的趋势,但在60和90 Gy同时存在矮化与增高的植株。利用SRAP (sequence related amplified polymorphism)技术对辐照后M1代的黄花蒿植株进行研究,结果表明对照组和处理组之间的差异体现为特异性条带的变化。综合分析表明,12C6+的重离子辐照黄花蒿干种子可以产生显著的当代损伤效应,同时引起植物DNA的多态性变化。
利用不同剂量的碳离子束辐照处理牛至精油微胶囊,研究对其缓释特性及抗鸡球虫病活性的影响.研究结果表明,通过碳离子束处理的牛至精油微胶囊,其主要活性成分百里香酚和香芹酚含量未发生变化,辐照处理后反应收率及产品的生物活性均提高,其中400 Gy剂量处理的牛至精油微囊的收率和生物活性均最高,收率达68.44%.通过对感染柔嫩艾美耳球虫的小鸡喂食后,其盲肠和十二指肠肿胀明显减轻,血液性内容物明显减少,血便指数在感染的第七天已降为0,抗球虫指数(ACI)达到179.39.
Using the Markov Chain Monte-Carlo method to estimate the parameters in the Logistic regression model,we constructed a non-periodic irreducible Markov Chain with the posterior distribution of the parameters as stationary distribution,and then used the sample points extracted from the stationary distribution to calculate the Monte-Carlo integral.The above theoretical method can solve the difficult problem of classical logistic regression modeling because of the existence and limitation of the experimental sample data and the multicollinearity.In the classical regression setup with a continuous response,the predicted values can range over all real numbers.Therefore,a different modelling technique is needed.In this work,the results describe in detail a previously unknown lethality trend following 12C6+ heavy-ion irradiation of Clostridium tyrobutyricum.By Markov Chain Monte-Carlo can calculate the model fit for a randomly selected subset of the chain and calculate the predictive envelope of the model.The grey areas in the plot correspond to 50%,90%,95%,and 99% posterior regions.More importantly,although this study focused on the use of the method in heavy-ion irradiation of microbial,its results are broadly applicable.

A software system used for the consoles of NMR spectrometers,designed using a multi-layer architecture,is presented.The software system is divided into five layers: Linux driver layer,driver interface layer,BZ logic layer,socket layer and protocol control layer.Each layer has a specific function and is maintained independently.It was shown that multi-layer architecture design not only reduces coupling in the system and simplifies the system structure,but also allows developing different layers simultaneously,thus improving the programming efficiency and reducing the development cycle time.The software system was debugged on the hardwares developed independently in the lab and passed rigorous testing.
Software testing has been widely used as a way to improve the quality of software.In this paper,a method for software testing in the development of a nuclear magnetic resonance(NMR) spectrometer was developed.Beginning with the introduction of the concepts for testing,the difficulties and solutions during the testing were analyzed according to the software structure of the console system of the spectrometer and the characteristics of the NMR experiments.The realization of software testing,which played a constructive role in improving the reliability and stability of the NMR spectrometer,was introduced in detail.
