猪细胞巨化病毒病又称猪包涵体鼻炎,是由猪细胞巨化病毒(PCMV)引起的仔猪的一种常见传染病[1].在易感猪群中可引起胚胎和仔猪死亡,表现生长发育不良、仔猪鼻炎、肺炎及增重差等临床症状.PCMV主要是一种"机会性感染"病毒,多呈隐性感染,潜伏感染状态的猪在长途运输、寒冷、分娩等应激因素作用下,病毒可被激活并重新排毒.常规条件下饲养的大多数猪群均存在PCMV感染,PCMV抗体阳性率较高,但大多数为亚临床型,临床发病则很少见[2,3].
Porcine cytomegalovirus is a β-herpesvirus.GX strain was isolated from the pig with the clinical symptom in December,2007 in Guangxi province.The gB gene of porcine cytomegalovirus GX strain was amplified through PCR,and the amplification product consisted of 1736bp.After being cloned and sequenced,the homology of the nucleotide sequence was 97.8%-98.9% and 32.0%-43.6%,respectively,compared with the published porcine and human cytomegalovirus gB gene sequence GenBank.The results indicated that gB gene of GX strain was conservative in the same genus.Phylogenetic tree analysis showed that GX strain had high homology to other porcine cytomegalovirus strain,and belonged to the same branch,but the homology was very different from human cytomegalovirus.
According to the complete genome of foot-and-mouth disease virus (FMDV) type O, a pair of special primers was designed to amplify VP1 gene. The VP1 gene was amplified by RT-PCR and subsequently inserted into the expression vector pGEX-6p-1 and induced by IPTG. Then SDS-PAGE showed the expressed protein was 51 kD in molecular weight. Then the product was purified by GSTrap FF columns. The product was detected through Western-blot that showed the protein has antigenicity. It provided fundamental data and materials for further investigation on diagnosis method of FMDV.
为了解广西猪伪狂犬病毒的感染状况,应用PCR技术对来自广西46个种猪场的1940份公猪精液进行了猪伪狂犬病毒(PRV)的检测,阳性率为0;同时对广西25个已使用猪伪狂犬病病毒gE基因缺失疫苗猪场的1455份送检血清,采用了猪伪狂犬病基因缺失鉴别ELISA方法进行了血清学抗体监测,结果检出猪伪狂犬gE抗体阳性11份,阳性率为0.76%.结果表明,广西猪群中存在猪伪狂犬病野毒感染,但感染率较低.
