对食用药用真菌灰树花发酵进行建模,获得使目标发酵产物达到最大产量的培养条件.运用支持向量机(support vector machine,SVM)方法进行非线性拟合,并采用遗传算法预测优化培养基成分,结果表明其能够较好预测灰树花发酵过程.运用此方法可在灰树花发酵生产过程中根据所需产物控制发酵条件与时间,具有较高指导意义.
To study the protective effects of intracellular polysaccharide from Morchella esculenta(MEP)on acute liver injury caused by CCl4 in mice,the mice were given i.g.MEP daily for 18 days before the models were made by i.p.injection of CCl4.The content of ALT,AST activity were measured in the serum,SOD,MDA were measured in the liver and 1iver index were also detected.MEP could significantly decrease the content of ALT,AST,MDA and liver index,and also improve SOD activity in liver.The lesions in liver lobule were ameliorated obviously.The results showed that MEP had protective effects on mice acute hepatic injury.
Objective: To study the antitumor effects of extracellular polysaccharides from Morchella esculenta (EPM) and the immunological function on S180 tumor-bearing mice. Method: EPM was intragastrically given to S180-bearing mice at the dose of 200 mg/kg and 400 mg/kg. Normal sodium and cyclophosphamide were taken as blank control and positive control respectively. The tumor weight, immunologic organ weights and life span of S180-bearing mice were measured. S180 tumor cells were incubated with various concentrations of EPM for 24 h, and antiproliferative activities against tumor cells were determined by MTT assay. Result: EPM significantly increases the spleen index, percent of T-lymphocyte, macrophage phagocytic index and prolongs survival time of S180-bearing mice.The inhibiting tumor rates at the low and high does are 49.04% and 64.42% respectively. EPM can kill S180 tumor cells directly in vitro, with the highest inhibiting rate 46.3% at the dose of 50μg/well.Conclusion: EPM has significant antitumor effects and improves immunological function.
A polysaccharide(MEP-Ⅱ)could be extracted from the mycelia of Morchella esculenta,after purified by DEAE-Sepharose F.F chromatography and Superdex 200 chromatography.MEP-Ⅱwas identified as a homogeneous polysaccharide-protein complex by agarose gel electrophoresis and HPLC.Its average M_w was estimated to be 2.8×10~4 u by gel filtration.The content of total saccharide was 87.5 %.The content of total protein was 8.4%.The content of acid polysaccharide was 23.2%.β-glucosidic bond was determined by IR.Typical absorption of polysaccharide was shown in its IR spectrum.Rha,Gal,Glc,and Man were iden- tified in MEP-Ⅱwith the molar ratio of 2.1 :11.3:35.6:1.0 by GC.β-elimination reaction demonstrated that MEP-Ⅱwas linked to peptides chain through O-glycosidic linkage in it.
Purpose To extract and identify the polysaccharide of Semiaguilegia adoxoides(DC) Makino(SAP).Methods The polysaccharide was extracted with water and the content of total polysaccharide was determined with phenol-sulfuric acid.And acidic polysaccharide was determined by carbazole method.Protein was determined by biuret method.Molecular weight of SAP was determined by high performance gel permeation chromatography.The monosaccharide component was determined by GC.In addition,SAP was analyzed by IR spectroscopy.Results The content of total sugar and protein was 80.4% and 16.6% respectively and acidic polysaccharide was 6.8%.The molecular weight of SAP was 6.3×104.The component analysis indicated that SAP was composed of Glc,Gal,Man,Ara and Fuc with molar ratio of 6.23∶2.04∶3.15∶1.03∶1.The IR spectrum revealed that SAP was a furanose.Conclusion The SPA is a mixture of 5 monosaccharides.
To study the protective effects of Morchella esculenta on acute hepatic injury induced by CCl_4 in miee.Pretreated with the extract from Morchella esculenta for 16 days,mice modles of acute hepatic injury were estabhshed by the treatment of carbon tetrachloride(CCl_4) .After 24 hours,the protective effects of the extract from Morchella esctdenta in acute hepatic injury were observed through determing the activities of alanine aminotransferase(ALT),(SOD),Maleie Dialdehyde(MDA),and the contents of proteins in serum. The extract from Morchella eseulenta significantly reduce the level of serum ALT,MDA and the hepatic index,and increasing SOD activity,level of serum protein.It has showed that Morehella esctdenta has proactive effects on mice acute hepatic injury.
