Small and medium technology-based agricultural enterprises have become an internal power of developing modern agriculture in China,while they face different problems and crises that can not be neglected.This article analyzes their financial management problems and proposes relevant strategies of improving their financial management.
Nicotiana tabacum inoculated with tomato spotted wilt virus(TSWV) and Gerbera jamesonii infected with tobacco rattle virus(TRV) and tobacco mosaic virus(TMV) were used as test materials,and a multiplex RT-PCR method was established to synchronously detect the above 3 kinds of viruses.The test results indicated that the DNA bands of all the viruses could be amplified just once by the established method,and the mixed viruses could be detected from 1 mg RNA-containing leaf with viruses.The sensitivity analysis of the RT-PCR method showed that the TSWV,TRV and TMV could be detected in a virus-infected material containing RNA of at least 1 mg,10 μg and 100 ng respectively.The established multiplex RT-PCR method had high specificity and sensitivity and could greatly reduce the detecting time.
利用感染了番茄黑环病毒、烟草花叶病毒和黄瓜花叶病毒的非洲菊叶片,建立同步检测非洲菊3种病毒的多重RT-PCR和实时荧光RT-PCR,多重RT-PCR可一次性扩增出3种病毒的DNA条带,最低可检测到相当于1μg的带毒叶片;实时荧光RT-PCR可分别检测相当于1ng、1ng和100pg的带毒叶片。同时采用茎尖、叶片和花托组织培养脱毒并结合培养基添加病毒A的脱毒方法,对接种3种病毒的非洲菊进行外植体脱毒试验,获得非洲菊组培苗脱毒方法。用建立的多重RT-PCR和实时荧光RT-PCR检测脱毒效果,茎尖脱毒率分别为75.0%和66.7%,花托脱毒率分别为52.0%和54.5%。
The multiplex RT-PCR approach was developed for simultaneous detections of Arabis mosaic virus(ArMV),Strawberry latent ringspot virus(SLRSV)and Lily symptomless virus(LSV)from the imported lily bulbs.The results indicated that good specificity and sensitivity for simultaneous detection were obtained.The ultimate of RNA detection with three viruses mixture was 305 pg.The ultimate of RNA detection with ArMV,SLRSV and LSV were 156.0,13.4 pg and 1.12 ng respectively.This approach has potential to be used in quarantine of imports and exports.
