In order to compare the separation methods of bovine fibroblasts,the primary cells were isolated and cultured by tissue attachment method and tissue attachment + double enzyme digestion method(trypsin + collagenase Ⅱ)after the samples of bovine ear skin were collected and brought back to the laboratory at low temperature.The results showed that it took 6~7 days for the cells to migrate from the tissue blocks,spindle-shaped cells were visible at 10~11 days,and swirly growth halos were observed at 16 days.Tissue adhesion + double enzyme digestion method requires only 5 days for cells to migrate out of the tissue block,spindle cells can be seen on the 6th day,and the swirly growth halo can be observed on the 10th day.The comparison of the two methods shows that the tissue adhesion and double enzyme digestion method can quickly separate and obtain bovine fibroblasts,and is the best method to obtain bovine fibroblasts.

河南省的南阳牛、郏县红牛是由河南省经过长期不断选育而形成的地方优良品种.近年来,由于牛肉需求不断加大以及对牛肉品质要求的提高,同时由于河南肉牛遗传改良基础好,培育具有优良肉质的新品种成为当前河南省育种产业的首要任务,与此同时,河南肉牛产业也凸显了一些新的问题.因此,本文综述了河南省肉牛遗传育种现状、遗传育种相关研究进展以及遗传育种存在的问题,从而为下一步河南省地方牛种质研究及创新利用提供对策建议.
种公牛的精液品质除了与饲养管理、个体差异、疾病等有关外,受气温的影响也是很大的.为了探讨气温对种公牛精液品质的影响,笔者从2007年至2009年连续3年进行观察和实验,以精子活力、畸形率、顶体完整率为主要指标对生产记录进行了统计分析,结果如下.
该试验分别通过向精液中添加适量催产素和在输精前5分钟向母猪子宫内注入适量催产素,以探讨外源性催产素对猪受胎率的影响。试验选用大白后备母猪、经产母猪(3胎次)各36头,按胎次随机分为6组,试验Ⅰ组、试验Ⅱ组、对照A组为后备母猪,每组12头,试验Ⅲ组、试验Ⅳ组、对照B组为经产母猪,每组12头。试验Ⅰ组、试验Ⅲ组母猪在输精前5分钟分别向子宫内注入10国际单位的催产素,试验Ⅱ组、试验Ⅳ组母猪以含有催产素的稀释精液进行输精。结果为:试验Ⅰ组、试验Ⅲ组的受胎率均高达91.67%、窝均产仔数为7.57头和11.88头,均比试验Ⅱ组、试验Ⅳ组要高。经方差分析可知:各组的受胎率、窝均产仔数、窝均产活仔数差异显著(P<0.05),由此表明输精前5分钟向子宫内注入10国际单位的催产素对提高母猪受胎率和增加产仔数有明显的促进作用。
