To further research the biodiversity of Orbilia,a survey was conducted from September to October in 2013.Results:O.corculispora was reported for the first time in China.Its anamorph was isolated from the ascospores of O.corculispora.The connection between anamorph O.corculispora and its teleomorph was established.
The effect of preharvest calcium sprays and low temperature treatment on storage quality of Yangshuo kumquat fruits with preharvest calcium sprays three times and then storage at 4 and 25 ℃ when the fruits were mature were studied.The results showed that the rotten rate and weight loss rate,the fruit firmness,TSS,TA,TS and Vc content of kumquat fruits treated by CaCl2 and 4 ℃ low temperature storage for 60 days were 30.00 %(absolute value)and 3.98 %(absolute value) lower,38.48 %,2.50 %(absolute value),0.11 %(absolute value),4.58 %(absolute value)and 52.56 % higher than that stored at 25 ℃ for 60 days without calcium spray,respectively.The preharvest calcium sprays or low temperature storage could restrain the occurrence of rotten and weight loss,delay the decrease of the fruit firmness,TSS,TA,TS and Vc content,combination of both was the best treatment in maintaining the quality of kumquat fruits.
The protein expressions under low temperature stress were studied using proteomics method.The results showed that the expression of DLPIP1 protein was up-regulated. The full length of DLPIP1 eDNA obtained by RT-PCR was 1 132 bp with a 900 bp open read frame which encoded a putative DLPIP1 pro- tein with 299 amino acids. Comparison of the amino acid sequence homology among DLPIP1 proteins from 21 different species indicated that DLPIP1 protein had a range of 90% to 93% identity with homologues of other plants. Bioinformatics analysis demonstrated that DLPIP1 exhibited a typical structure with seven membrane-spanning domains and an internal symmetry showing two highly conserved NPA motifs and possessing the MIP family signal consensus sequence. The DLPIP1 amino acids showed high identity with the PIP plasmalemma subfamily of other 21 plant species by homology comparison analysis. Quantitative real-time PCR results showed that the DLPIP1 expressed in root, stem and leaf, while the amount of ex- pressions were different in different organs. The mRNA of DLPIP1 was the most abundant in root, the least in leaf and stem. Furthermore, the mRNA of DLPIP1 changed with time extension under low temperature stress. These results suggested that DLPIP1 might be involved in function of chilling stress.
The dynamic changes of the Yangshuo kumquat fruits qualities were compared under rain shelter cultivation covered with plastic film and in open field cultivation as control from December 20,2010 to April 15,2011.In order to understand the effects of rain shelter cultivation on quality of Yangshuo kumquat(Fortunella crassifolia Swingle cv.Yangshuo) fruits on tree storage in Yangshuo county.The results showed that the storage period on tree of Yangshuo kumquat under rain shelter cultivation could extend to mid-April of the next year and the good fruit rate still could reach 98.9%.The rain shelter cultivation could not noly delay the decrease of the fruit firmness and maintain higher VC content of later period,but also increased its soluble solids content,TSS-TA ratio and TS-TA ratio.During the whole storage period on the tree,the fruit firmness under rain shelter cultivation was 10.08% to 14.26% higher than that in open field cultivation,its soluble solids content was 2.12% to 5.86% higher than the control;titratable acid content was 13.64% to 14.29% lower than the control;soluble sugar content,TSS-TA ratio and TS-TA ratio of fruit under rain shelter cultivation were all higher than that of control.The VC content of kumquat fruits on the tree storage increased during the first two months and reached maximum in the second month,then reduced rapidly,but the VC content of kumquat fruits under rain shelter cultivation was 8.69% to 14.73% higher than that of control.
RACE technique is a rapid and effective method for cloning gene 5' end and 3' end,and one of main methods to obtain full-length genes.However,RACE technique has its own shortcomings.We further improved the previous RACE protocols and obtained a modified RACE technique which is simple,rapid,high-efficient and low-cost.Our modified RACE technique is suitable for obtaining large number of genes 5' end and can be used in ordinary laboratories.
The technique of two-dimensional electrophoresis in Nanfeng mandarin peel proteomics was established and optimized by phenol-extraction method.The two dimension electrophoresis profiles of Nanfeng mandarin peel proteins were good repeatability and high resolution.The differential expression proteins of Nanfeng mandarine peel during storage were separated with 2-DE and then analyzed with Imagemaster 2D PDQuest.The result showed that the number of detected differential expression protein spots in Nanfeng mandarin peel under storage was 45,33 proteins were up-regulated,and 12 proteins were down-regulated.It was presumed that these proteins were related to the storability of Nanfeng mandarin peel.
[Objective]The research aimed to explore the tolerance and accumulation of Agaricus bisporus to lead and cadmium.[Method]A.bisporus was cultivated in medium with different concn.of lead and cadmium,and the lead and the cadmium contents in fruiting body and mycelia of A.bisporus were measured by the flame atomic absorption spectrophotometry.[Result]In the tested concn.range,the bioaccumulation of lead and cadmium in A.bisporus were increased with increasing lead and cadmium concn.in the medium,the contents of lead and cadmium were different in different parts of fruiting body and they were in the order of lamellae pileus stipe,being 19.42,16.42 and 15.30 mg/kg for the lead content,and 38.56,35.73 and 11.80 mg/kg for the cadmium content resp.The bioaccumulation amount of lead in A.bisporus was generally higher under the combined stress than under the single lead stress,but the bioaccumulation amount of cadmium was lower than that under the single cadmium stress.[Conclusion]A.bisporus had a high tolerance and accumulation to the lead and cadmium.
