目的 研究乙型肝炎病毒X蛋白(HBx)调节细胞内钙离子可能的分子机制,揭示乙型肝炎病毒(HBV)诱导肝癌的可能途径.方法 培养人胚肾细胞(HEK293),取第2代HEK293细胞共转染HBx基因、钙释放激活钙通道蛋白1(Orai1)基因或者间质相互作用因子1(STIM1)基因,免疫共沉淀(co-IP)实验观察细胞内蛋白结合情况,并使用谷胱甘肽巯基转移酶(GST)pull-down实验研究HBx与Orai1作用位点,使用钙离子测量/成像系统检测HBx蛋白对细胞内钙离子的影响.结果 转染后细胞生长状态良好,co-IP实验结果显示HBx蛋白在细胞内可以与Orai1蛋白结合,GST pull-down实验显示HBx蛋白可以与Orai1蛋白C末端结合,钙离子测量/成像系统检测显示HBx蛋白可升高活细胞钙内流.结论 HBx蛋白通过与细胞膜钙离子通道Oria1结合蛋白的C末端结合,增加细胞钙内流,扰乱细胞内钙离子平衡,从而影响细胞增殖等活性.
Effects factors were investigated on the decrease of polymer HPAM viscosity in the oilfield water for polymer injection production,including Fe2+,dissolved oxygen and corrosion of steel. The result shows that the essential factors are the concentrations of Fe2+ and oxygen for the oilfield reinjection effluent,and both are concerned to the corrosion of wastewater pipeline steel. Corrosion inhibitors with oxygen resistance were presented and evaluated for the viscosity keeping and corrosion resistance. The viscosifying efficiency of 100 mg / L ND-I corrosion inhibitor reaches 93% and corrosion rate with 30 mg / L corrosion inhibitor is less than 0. 076 mm / a.
Generally polymer inhibitor molecule has multi-adsorptive group and is easy to form monolayer or multilayer protective film on metal surface, so that it has some advantages of low dose and high corrosion inhibition efficiency, long time of firm adsorption and low environmental pollution to become one of the important direction of corrosion inhibitor. Some polymer corrosion inhibitors, such as P-containing, N-containing and vinyl polymers were reviewed as well as the corrosion inhibition mechanism, emphasized on the application of quantum chemistry method. Finally the development of polymer corrosion inhibitor was prospected.
王14断块属于高渗透稀油常压断层一岩性油藏。1988年投入开发,随着开发时间的延长,区块逐渐暴露出油井井况问题突出、老井产量递减快、井网不完善等一些问题。针对上述问题,2010年在精细油藏描述的基础上,通过零散井、老井侧钻等手段,分区完善井网,提高储量的控制及动用程度,改善区块开发效果;同时深化单元目标化管理,对区块开发状况及时跟踪分析。2010年王14断块储量控制程度提高20%,增加可采储量12万吨。
