实验教学是学生理解掌握理论知识的必要途经,有助于提高学生的学习兴趣、培养学生实践动手能力.植物科学学院农学、植物保护、园艺、资源与环境4个涉农专业开设的机械课程实验课时少、设备少而不全、设备陈旧,并且针对性不强.针对上述现状,研制3套自制教具,并成功应用在植物科学学院4个涉农专业的机械实验课和校开放性创新实验.在实验教学中构建了"引入自制教具的混合教学模式",深受学生欢迎.学生问卷调查表明,学生对教学模式、提高学习兴趣、自学能力、自制教具的适用性等7个问题的提高程度或认可度均在90%以上.自制教具及其混合教学模式大幅提升了课堂效率,教学质量得到显著提高.
本研究旨在比较不同产蛋水平肉种鸡繁殖性能、肠道组织形态、卵巢功能和盲肠微生物区系的差异.试验选用5000只同栋舍37周龄的爱拔益加父母代肉种鸡,记录2周内每只肉种鸡的产蛋率、受精率和孵化率,分别筛选出高和平均2种不同产蛋水平的肉种鸡各90只,设为2个组,分别为平均产蛋率组[AR组,产蛋率(79.34±0.49)%]和高产蛋率组[HR组,产蛋率(90.03±0.34)%],每组10个重复,每个重复9只鸡.预试期2周,正试期6周.结果表明:与AR组相比,HR组肉种鸡产蛋率和入孵蛋孵化率显著升高(P<0.05),血清谷丙转氨酶活性显著降低(P<0.05),空肠绒毛高度显著降低(P<0.05),回肠隐窝深度显著升高(P<0.05),卵巢细胞凋亡率极显著降低(P<0.01),卵巢促凋亡蛋白半胱氨酸天冬氨酸蛋白酶9(caspase 9)、卵泡发育相关蛋白骨形态发生蛋白受体1B(BMPR1B)、家鸡新型类催乳素(PRL-L)和转录因子GATA4表达水平显著升高(P<0.05),盲肠厚壁菌门(Firmicutes)和乳杆菌属(Lactobacillus)相对丰度显著升高(P<0.05),盲肠拟杆菌门(Bacteroidetes)和螺旋体门(Spirochaetes)相对丰度显著降低(P<0.05).肉种鸡盲肠拟杆菌门相对丰度与卵巢BMPR1B和GATA4表达水平呈显著负相关(P<0.05),盲肠厚壁菌门相对丰度与卵巢BMPR1B、GATA4表达水平呈显著正相关(P<0.05),盲肠乳杆菌属相对丰度与卵巢caspase 9、BMPR1B、GATA4、PRL-L表达水平呈显著正相关(P<0.05),盲肠螺杆菌属(Helicobacter)相对丰度与卵巢GATA4表达水平呈极显著正相关(P<0.01).综上所述,本试验结果表明,不同产蛋水平肉种鸡肠道组织形态、盲肠微生物区系和卵巢功能存在显著差异,盲肠厚壁菌门和乳杆菌属相对丰度与高繁殖性能密切相关.
本试验旨在探讨饲粮中添加不同水平芦丁对产蛋后期蛋鸡生产性能、蛋品质及蛋清凝胶特性的影响.选择66周龄的健康罗曼粉壳蛋鸡240只,随机分为4组,每个组6个重复,每个重复10只鸡.各组分别在基础饲粮中添加0(对照)、100、200、400 mg/kg芦丁.试验期8周.结果表明:1)饲粮中添加不同水平芦丁对蛋鸡的产蛋率、平均蛋重、采食量以及料蛋比均没有显著影响( P>0.05) . 2)与对照组相比,第2周、第4周、第8周时,饲粮中添加不同水平芦丁均能显著提高鸡蛋的蛋壳厚度( P<0.05) ;第4 周时,200 mg/kg芦丁组鸡蛋的蛋壳强度显著提高( P<0.05) ;第8周时,400 mg/kg芦丁组鸡蛋的蛋黄比重显著提高( P<0.05) .饲粮中添加不同水平芦丁对鸡蛋的蛋壳颜色、蛋白高度、蛋黄颜色、哈夫单位、蛋黄重和蛋重均没有显著影响(P>0.05). 3)与对照组相比,第2周时,400 mg/kg芦丁组蛋清凝胶的回复性显著提高( P<0.05) ;第8周时,400 mg/kg芦丁组蛋清凝胶的硬度和胶性显著提高( P<0.05) .饲粮中添加不同水平芦丁对蛋清凝胶的胶黏性、弹性、黏聚性均没有显著影响( P>0.05). 4)与对照组相比,第8周时,饲粮添加不同水平芦丁有提高蛋清的起泡性的趋势( P=0.071) .由此可见,饲粮中添加100、200、400 mg/kg芦丁对产蛋后期蛋鸡生产性能没有显著影响,但可以显著增加鸡蛋的蛋壳厚度;饲粮中添加400 mg/kg芦丁可以提高蛋清凝胶的回复性、硬度、胶性和蛋清的起泡性.在本试验条件下,饲粮中芦丁的适宜添加水平为400 mg/kg.
本试验旨在评价添加蛋白酶对肉鸭菜籽粕代谢能值和饲粮养分真利用率的改善效果.选用19周龄以上健康的樱桃谷肉公鸭,采用真代谢能(TME)结合套算法进行代谢试验,选用5种不同蛋白质溶解度的菜籽粕(分别命名为A1、A2、A3、A4、A5),菜籽粕按40%替代玉米,豆粕按35%替代玉米.试验设13个组,包括1个玉米组、1个豆粕组、1个饥饿内源组、5个菜籽粕不加酶组、5个菜籽粕加酶组(蛋白酶制剂添加量为200 mg/kg,蛋白酶制剂中蛋白酶活性为1×105 U/g).每个组设12个重复,每个重复1只鸭.结果表明:1)添加蛋白酶使菜籽粕饲粮干物质(DM)、粗脂肪(EE)、粗蛋白质(CP)、中性洗涤纤维(NDF)、酸性洗涤纤维(ADF)、粗纤维(CF)、粗灰分(Ash)、钙(Ca)、磷(P)真利用率分别提高了3.73%(P<0.05)、9.92%(P<0.05)、8.51%(P<0.05)、15.78%(P<0.05)、17.09%(P<0.05)、2.12%(P>0.05)、8.55%(P<0.05)、2.81%(P>0.05)、12.96%(P<0.05);2)添加蛋白酶可以显著提高菜籽粕的表观代谢能(AME)、TME、氮校正表观代谢能(AMEn)、氮校正真代谢能(TMEn)(P<0.05),提高程度分别为2.01、2.01、1.80、1.80 MJ/kg.综上可知,添加蛋白酶可以改善肉鸭菜籽粕代谢能值和菜籽粕饲粮养分真利用率.
本试验旨在研究苯甲酸对球虫攻毒肉鸡生长性能、免疫功能及血清抗氧化能力的影响.试验采用2×3双因子设计,选用540只1日龄健康爱拔益加( AA)白羽肉公鸡,2种攻毒处理(灌服无菌生理盐水或30 倍球虫疫苗) ,3 种苯甲酸源[饲粮中不添加苯甲酸( N-B) 、添加0.05%肠溶型缓释苯甲酸( ES-B)或0.10%未包被苯甲酸( NC-B) ] ,共计6个组,每组6个重复,每个重复15只鸡.试验期42 d.结果表明:1)饲粮中添加苯甲酸有提高肉鸡42 日龄时体重( BW)和1~42日龄时体增重( BWG)的趋势( P=0.058,P=0.057);球虫攻毒显著降低BW(除14日龄时外) 、BWG和平均采食量( AFI) (除1~14日龄时外) ( P<0.05) ,显著提高15~21日龄时的料重比( F/G) ( P<0.05) ,且N-B组的生长性能下降幅度大于ES-B和NC-B组. 2)饲粮中添加苯甲酸显著提高肉鸡21日龄时的血清免疫球蛋白M( IgM)含量、胸腺指数和42日龄时的血清免疫球蛋白A( IgA)含量( P<0.05) ,有降低42日龄时血清一氧化氮( NO)含量的趋势( P=0.068) ;球虫攻毒显著提高42日龄时的血清白蛋白( ALB)含量,显著降低21日龄时的血清碱性磷酸酶( ALP)活性和42日龄时的血清IgM含量( P<0.05) ;饲粮中添加苯甲酸与球虫攻毒对21日龄时的血清IgM含量有显著交互作用( P<0.05) ,表现为在球虫攻毒条件下,饲粮中添加苯甲酸提高血清IgM含量,且以ES-B组效果最佳. 3)饲粮中添加苯甲酸显著提高肉鸡21日龄时的血清谷胱甘肽过氧化物酶( GSH-Px)活性( P<0.05) ,显著降低血清丙二醛( MDA)含量( P<0.05) ;饲粮中添加苯甲酸与球虫攻毒对42日龄时的血清MDA含量和GSH-Px活性有显著交互作用( P<0.05) ,表现为在球虫攻毒条件下,饲粮中添加苯甲酸提高血清GSH-Px活性及降低血清MDA含量,且以NC-B组效果最佳.由此得出,饲粮中添加苯甲酸可提高肉鸡的生长性能;在球虫攻毒条件下,饲粮中添加苯甲酸可通过改善机体健康和免疫功能,从而缓解球虫攻毒造成的生长性能下降,且以添加0.05% ES-B的改善效果较好.
本试验旨在探讨一种新型的维生素D添加剂,即25-羟基维生素D3( 25-OH-D3 )与维生素D3 共晶对肉鸭生长性能、免疫功能及胫骨发育的影响.选取360只1日龄健康樱桃谷肉公鸭,随机分为3个组,每组6 个重复,每个重复20 只. T1 组在基础饲粮中添加69. 0 μg/kg 25-OH-D3与66.0 μg/kg维生素D3 物理混合物,T2组在基础饲粮中添加135.0 μg/kg 25-OH-D3与维生素D3 共晶,T3组在基础饲粮中添加67. 5 μg/kg 25-OH-D3 与维生素D3 共晶.试验期35 d,分为1~14日龄和15~35日龄2个阶段.结果表明:1)与T1组相比,T2组肉鸭35日龄体重以及15~35 日龄和1~35 日龄平均日增重( ADG)和平均日采食量( ADFI)显著提高( P<0.05),35日龄胸肌肉色亮度值显著增加(P<0.05) ,35日龄脾脏指数显著降低( P<0.05) ,14日龄血清Ⅰ型胶原C端肽含量和35日龄血清钙含量显著升高( P<0.05) ,35日龄胫骨粗灰分含量及胫骨生长板宽度和宽度系数显著增加( P<0.05) . 2)与 T1 组相比,T3 组肉鸭1~35 日龄ADG和15~35日龄ADFI显著提高( P<0.05),14日龄血液嗜酸细胞百分率和35日龄血液单核细胞计数显著增加( P<0.05),14日龄血清钙和降钙素含量显著降低( P<0.05) ,14日龄胫骨钙含量以及35日龄胫骨体积和粗灰分含量显著降低( P<0.05),35日龄胫骨生长板厚度、厚度系数、宽度系数和横截面积显著增加( P<0.05) . 3)与T2组相比,T3组肉鸭35日龄胸肌剪切力显著提高( P<0.05) ,14日龄法氏囊指数和35日龄脾脏指数显著提高( P<0.05) ,14日龄血液中性粒细胞百分率、单核细胞百分率、嗜碱细胞百分率以及单核细胞计数显著升高( P<0.05),14日龄血液淋巴细胞百分率显著降低( P<0.05) ,14日龄血清降钙素含量和35日龄血清钙和Ⅰ型胶原C端肽含量显著降低( P<0.05) ,14日龄胫骨粗灰分和磷含量以及35日龄胫骨钙含量显著降低( P<0.05),14日龄胫骨生长板宽度和宽度系数显著增加( P<0.05).由此可见,饲粮中添加25-OH-D3 与维生素D3 共晶对于肉鸭的生长性能、免疫功能及胫骨发育有显著影响,且具有剂量依赖性,表现为饲粮中添加135.0 μg/kg 25-OH-D3 与维生素D3 共晶效果更佳.
本试验旨在通过实验室分析和代谢试验建立基于菜籽饼粕养分的代谢能值预测方程,为生产提供便捷的估测菜籽饼粕代谢能值的方法.试验共收集25种菜籽饼粕样品,选用19周龄以上健康的樱桃谷肉公鸭进行代谢试验.代谢试验分3批进行,前2批代谢试验均设13个组:玉米组、玉米-豆粕组、绝食内源组和10个菜籽饼粕组;第3批代谢试验设8个组:玉米组、玉米-豆粕组、绝食内源组和5个菜籽饼粕组.前2批代谢试验均使用156只,而第3批代谢试验使用96只,每组设12个重复,每个重复1只鸭.待测饲粮由豆粕或菜籽饼粕替代玉米组成,其中豆粕按35%替代玉米,菜籽饼粕按40%替代玉米.采用真代谢能(TME)结合套算法评定菜籽饼粕的代谢能值,强饲量为试验鸭体重的2%,排空时间和排泄物收集时间均为36 h,每批之间设14 d的恢复期.采用逐步回归法建立菜籽饼粕养分与代谢能值的回归方程.结果表明:1)25种菜籽饼粕的总能(GE)平均值为17.91 MJ/kg[16.48~20.04 MJ/kg,变异系数(CV)=5.24%],干物质(DM)含量平均值为89.72%(86.20%~93.73%,CV=2.21%),粗蛋白质(CP)含量平均值为44.56%(33.91%~50.70%,CV=8.86%),粗脂肪(EE)含量平均值为5.63%(1.21%~10.92%,CV=61.83%),粗灰分(Ash)含量平均值为7.58%(6.90%~8.48%,CV=6.04%),粗纤维(CF)含量平均值为12.89%(7.98%~16.18%,CV=15.08%),中性洗涤纤维(NDF)含量平均值为35.86%(20.30%~62.73%,CV=28.21%),酸性洗涤纤维(ADF)含量平均值为23.23%(12.75%~40.10%,CV=28.12%),钙(Ca)含量平均值为0.78%(0.22%~1.29%,CV=31.01%),磷(P)含量平均值为1.07%(0.85%~1.32%,CV=8.24%).2)25种菜籽饼粕的表观代谢能(AME)平均值为10.21 MJ/kg(8.28~12.51 MJ/kg,CV=17.87%),TME平均值为10.96 MJ/kg(9.00~13.39 MJ/kg,CV=16.66%),氮校正表观代谢能(AMEn)平均值为9.04 MJ/kg(7.28~10.75 MJ/kg,CV=17.42%),氮校正真代谢能(TMEn)平均值为9.71 MJ/kg(7.87~11.51 MJ/kg,CV=16.21%).3)通过逐步回归分析建立菜籽饼粕AME、TME、AMEn、TMEn的预测方程:AME=0.0107PS+0.2265EE+8.3528(R2=0.7081,P<0.01);TME=0.0114PS+0.2239EE+9.1033(R2=0.7047,P<0.01);AMEn=1.0916GE+0.0193PS-0.2340DM-0.1589CF+0.0599ADF+10.1119(R2=0.8582,P<0.05);TMEn=0.7945GE-0.2143CF-1.7817(R2=0.7626,P<0.01).由此可见,樱桃谷青年公鸭对不同来源菜籽饼粕的代谢能值存在差异,可利用菜籽饼粕的养分构建其代谢能值的预测方程.
课程思政是高校实现"三全育人"的重要途径.将课程思政元素融入生物统计学实验教学中,结合典型案例,促进了本科生对专业知识的理解,有效实现了对本科生的价值引领.
Background MicroRNAs (miRNAs) is a class of functional regulator of tumorigenesis of human cancer including hepatocellular carcinoma (HCC). However, the potential clinical significance of serum exosomal miR-320d in HCC has not been elucidated. Methods Real-time reverse transcription PCR was used to detect the expression pattern of serum exosomal miR-320d in patients with HCC, and the correlation between the deregulation of serum exosomal miR-320d and the clinical outcome of HCC was explored. The biological function of exosomal miR-320d in HCC was also investigated. Results Our results showed that the expression levels of exosomal miR-320d were remarkably reduced in the serum samples of HCC patients and the culture medium of HCC cell lines compared with their respective controls. Serum exosomal miR-320d could differentiate the HCC patients from healthy controls with high accuracy. In addition, its level was remarkably increased in the HCC patients who had received surgical treatment. Moreover, reduced serum exosomal miR-320d was associated with advanced tumor stage, positive lymph node metastasis, and poorly differentiated tumors. HCC patients with lower serum exosomal miR-320d had shorter overall and disease-free survival. Low serum exosomal miR-320d was identified to be an independent unfavorable prognostic factor for HCC. Finally, overexpression of miR-320d inhibited the proliferation and invasion of HCC cells, and BMI1 was demonstrated to be a direct target of miR-320d. Conclusion Taken together, serum exosomal miR-320d could be a potential non-invasive biomarker for the diagnosis and prognosis of HCC.
高阶思维能力是当前社会对人才新的要求.问题解决能力、批判性思维能力、创新性思维能力是高阶思维能力中的三种重要能力.笔者对这三种高阶思维能力进行了技能体系分析,并结合“牧草生产学”综合性实验,分别给出了具体的能力培养策略,即基于混合式教学模式的“牧草生产学”综合性实验教学,通过树立培养学生高阶思维能力的总体目标、设计利于思考的思辨性学习活动、给予学生思维发散与聚合机会、培养本科生理清问题脉络的日常习惯、建立公平合理的考核机制,激发了本科生对实验内容的学习兴趣.
动物科学和动物医学实验研究常用的统计分析方法有t检验、卡方检验、非参数检验、单因素方差分析、两因素方差分析、多因素方差分析、协方差分析、回归分析、相关分析以及主成分分析和聚类分析等多元统计分析.统计分析方法的选择要考虑分析目的、因素和水平数量、试验设计类型、资料类型、变量数量和类型、统计分析方法的前提条件等.本研究对常用的统计分析方法的分类、选择以及如何在SPSS上的实现等进行详细分析,以期为动物科学和动物医学实验研究提供参考.
Freezing-thawing and saline-alkaline are the major abiotic stress for the pasture in most high-latitude areas, which are serious threats to the yield of pasture. In this study, the osmotic adjustment substances, membrane lipid peroxidation, and antioxidant enzymes activities of rye (Secale cereale L., cv. Dongmu-70) seedlings under different treatments: CK (no treatment), SC (Na2CO3 treatment), FT (freezing-thawing treatment), and FT+SC (combined Na2CO3 and freezing-thawing treatments), were investigated. At the freezing stage, the content of MDA and proline, the activity of APX, SOD, and POD increased with the decrease of the temperature in the leaves of rye seedlings in FT and FT+SC treatments and reached the maximum value at − 5 °C. In addition, the content of protein and H2O2, CAT activity reached the maximum value at 0 °C; the damage is larger under low temperature stress at 0 °C and − 5 °C in rye seedling. At the thawing stage, the content of MDA and H2O2 in seedling leaves decreased in FT and FT + SC treatments. These results demonstrated that proline content and antioxidant enzymes activities could play an important role in protecting cytomembrane and scavenging ROS respectively in rye under alkaline salt stress and freezing-thawing stress. The result also indicated rye seedlings were subjected to a freezing-thawing stress which resulted in a reversible (recoverable) injury.
采用星点设计效应面法考察了乙醇体积分数(0,17%,40%,63%,80%)、提取温度(60℃,68.5℃,80℃,91.5℃,100℃)和提取时间(15,24.5,37.5,50.5,60 min)对可溶性糖含量的影响,研究了蒽酮比色法测定可溶性糖含量的最佳提取方法.利用Design-Expert.8.0.6软件对实验数据进行二次多项式模型拟合.结果表明:乙醇体积分数和提取时间对可溶性糖含量有显著影响(P<0.05),提取温度无影响(P>0.05),影响次序为:乙醇体积分数>提取时间>提取温度.可溶性糖的最佳提取工艺为:乙醇体积分数54%,提取温度100℃,提取时间60 min.
高阶能力是近年来我国教学改革的目标导向之一.借助CiteSpace软件对知网收录的高阶能力高质量研究文献进行量化分析,理清了不同时段的研究热点.通过对文献内容进行系统梳理,得出我国高阶能力培养理论研究还需关注国际发展的新动向,实践研究有待进一步深入开展,评价研究更需引入国际教育测评领域的前沿方法进行科学测度.
Chloride deicing salt stress usually coincides with the event of freeze–thaw, and the short-term adaptation of Dongmu-70 Secale cereale L. seedlings to these stresses was investigated in this paper. The chloride deicing salt and the freeze–thaw (FT) simulation experiments were carried out in the lab and alternation refrigerator. The changes of soluble sugar, soluble protein, relative conductivity (RC), malondialdehyde (MDA), and catalase (CAT) activity in seedlings were studied under freeze–thaw stress (10, 5, 0, − 5, 0, 5, and 10 °C) and 0, 200, 400, and 600 mmol L−1 of chloride deicing salt stress (CK, D1, D2, and D3). The results indicated that the content of physiological index in different treatment groups rose first and then decreased within a freeze–thaw cycle. During the recovery phase (T8: 24 h after freeze–thaw stress and T9: 6 days after freeze–thaw stress), there was significant difference either in MDA and CAT activity between D2 × FT and D2 or in RC, MDA, soluble sugar, and CAT activity between D3 × FT and D3. The seedlings showed different adaptability under different intensities of combined stress, and the sequence of the changes in physiological index can be patterned as D × FT > FT > D > CK. Freeze–thaw and chloride deicing salt complex stress exhibited a synergistic effect on the plant, which indicates that the snow-melting operation would be more harmful in spring and autumn to plants than in winter.
为充分调动学生的学习兴趣,改善传统实验课枯燥乏味的状态,笔者根据ARCS(Attention,Relevance,Confidence,Satisfaction)理论对草地资源学实验课进行了混合式教学设计,从注意(Atten-tion)、关联(Relevance)、自信(Confidence)和满意(Satisfaction)四个方面详细剖析了草地资源学实验课的教学策略,并进行了教学效果评价.结果 表明:在ARCS理论指导下的草地资源学实验课教学设计提高了学生的成绩,同时也提升了学生们的学习动机.说明基于ARCS理论的教学设计能够对草地资源学实验教学起到积极的促进作用.
The application of deicing agents often coincides with freeze-thaw (FT); however, the physiological effects of the sodium chloride deicing agent and concentrated calcium-magnesium acetate (CMA) deicing agent on plants in the FT environment are not well investigated. To explore the physiological response characteristics of white clover seedlings under different kinds of deicing agents and FT stress, the changes in the content of total soluble protein, soluble sugar, malondialdehyde (MDA), superoxide dismutase (SOD) and peroxidase (POD) of white clover seedlings were determined under the simulated stress of FT and deicing agent (DA). The results indicated that the soluble protein content in the white clover seedlings decreased, whereas the contents of MDA and soluble sugar, and the activities of SOD and POD increased under the FT stress. In the FT cycle, the content of soluble sugar under single FT stress, and the activities of SOD and POD of the three FT groups all showed a negative correlation with temperature. The MDA showed a significant correlation with other indicators, and the correlations among the other above physiological indexes were not significant (p < 0.05). The combined stress of FT + DA can cause increased plant damage, such as lipid peroxidation and osmotic potential imbalance. As an environmentally friendly DA, concentrated CMA was less harmful to plants than sodium chloride DA, but still brought damage to plants inevitably. Our results and corresponding discussion highlight the new findings on the effects of two kinds of deicing agents on plants under FT stress.
针对吉林大学农学、植物保护、农业资源与环境、生物技术、园艺5个涉农专业开设的机械课程中对内燃机的启动原理理解难的问题,研制了柴油机启动系统实验装置.该装置采用液晶显示屏显示,能实现人力启动和电力启动,其中电力启动采用触摸屏操作.该实验装置在实验教学和开放性创新实验中已进行了应用,提高了学生的学习兴趣、实践动手能力、创新意识.
Epithelial-mesenchymal transition (EMT) and angiogenesis is involved in tumor progression after radiofrequency ablation (RFA). ATPase inhibitory factor 1 (IF1) is a bad predictor of prognosis. Sorafenib inhibited EMT of hepatocellular carcinoma (HCC) after RFA. Whether IF1 promotes the EMT and angiogenesis of HCC and attenuates the effect of sorafenib after insufficient RFA is investigated. In this study, higher expression of IF1 was found in residual tumor after insufficient RFA. Hep3B or Huh7 cells after insufficient RFA were designated as Hep3B-H or Huh7-H cells in vitro. Hep3B-H or Huh7-H cells exhibited enhanced capacities of colony formation, migration, and increased expression of EMT associated markers and IF1 compared with Hep3B or Huh7 cells. IF1 knockdown in Hep3B-H or Huh7-H cells decreased the colony formation and migratory capacity, and IF1 overexpression in Hep3B or Huh7 cells increased these capacities. IF1 in HCC cells directly and indirectly affected angiogenesis of TAECs after insufficient RFA. IF1 promoted HCC cells growth and metastasis after insufficient RFA. IF1 increased HCC cells resistance after insufficient RFA to sorafenib. Higher IF1 expression indicated poor disease survival in HCC patients after sorafenib therapy. NF-κB activation induced by IF1 attenuated the effect of sorafenib on HCC cells after insufficient RFA. Our results demonstrated that IF1 promotes the EMT and angiogenesis, and attenuates HCC cell sensitivity to sorafenib after insufficient RFA through NF-κB signal pathway.
INTRODUCTION:MicroRNAs (miRNAs) are key modulators for gene expression via inducing translational repression or target gene degradation. miR-133a-3p was reported to stimulate or inhibit cancer progression but its role in hepatocellular carcinoma (HCC) remains to be explored.METHODS:Quantitative real-time PCR (RT-qPCR) was utilized to explore miR-133a-3p expression level in HCC cells. Dual-luciferase activity reporter assay was used to validate the direct interaction between miR-133a-3p and coronin-like actin-binding protein 1C (CORO1C). In addition, we analyzed the expression levels of miR-133a-3p and CORO1C in HCC tissues and normal tissues on the UCALAN website. Functional assays including cell counting kit-8 assay, colony formation assay, flow cytometry analysis and transwell invasion assay were conducted to explore the biological functions of miR-133a-3p in HCC.RESULTS:miR-133a-3p was found to have downregulated expression in HCC tissues and cells. Meanwhile, we showed that low miR-133a-3p levels were correlated with poorer overall survival of HCC patients. Overexpression of miR-133a-3p suppressed HCC cell growth and invasion but promoted cell apoptosis via targeting CORO1C.DISCUSSION:Our results revealed a novel mechanism of miR-133a-3p in regulating HCC progression and provided evidence that miR-133a-3p functions as a tumor suppressor in HCC.
