Objective:To investigate the effects of the differentiation of regulatory T cells (Tregs) on radiation-induced lung injury and its mechanism.Methods:A mouse model that inhibits Tregs was established. C57BL/6 mice were divided into the control group, simple irradiation group, irradiation+IgG group, and irradiation+CD25 group according to the random number table method. Each group comprised 12 mice. The mice in the simple irradiation group, irradiation+IgG group, and irradiation+CD25 group were given a single 20 Gy X-ray full chest irradiation. The mice in the irradiation+IgG group and irradiation+CD25 group were intraperitoneally injected with IgG antibody and CD25 antibody every week, respectively. Six mice were killed at 4 and 8 weeks after irradiation respectively. Flow cytometry was used to detect the percentage of CD25 +Foxp3 +Tregs in the lung tissue of each group of mice to identify whether the model was established successfully. Then, Western blot was used to detect the expression of neuropilin 1 (NRP1) in the lungs of the mice in the control group and irradiation group. Immunofluorescence was used to detect the percentage of CD25 +NRP1 +Tregs. Photos were taken to observe the skin damage of each group of mice. Hematoxylin eosin staining was used to detect the pathological changes of lung tissue. The secretion of cytokines, namely, transforming growth factor-beta 1 (TGF-β1), interleukin (IL)-17A, interferon-γ (IFN-γ), IL-2, and IL-4, in the lung tissue of each group was detected by enzyme-linked immunosorbent assay. Independent sample t-test was used for comparison between the two groups. Results:The results of flow cytometry showed that the percentage of CD25 +Foxp3 +Tregs ((1.73±0.04)%, (2.13±0.15)%) in the lung tissue of mice in the simple irradiation group was significantly higher than that in the control group ((1.14±0.02)%, (1.70±0.06)%) ( t=-26.680, -4.545; P=0.000, 0.010) at 4 and 8 weeks. After the inhibition of Treg cells, the percentage of CD25 +Foxp3 +Tregs ((0.72±0.14)%, (0.27±0.02)%) in the lung tissue of mice in the irradiation+CD25 group was significantly lower than that in the irradiation group alone ( t=5.296, 37.538; both P=0.000). The result of Western blot showed that the expression of NRP1 protein in the lung tissue of mice in the irradiation group increased significantly ( t=-7.341, -9.127; both P=0.000). The results of immunofluorescence showed that the proportion of CD25 +NRP1 +Tregs in the lung tissue of mice in the irradiation group was higher than that in the control group at 4 and 8 weeks after irradiation. The proportion of CD25 +NRP1 +Tregs in the irradiation+CD25 group was lower than that in the irradiation group ( t=8.926, 14.457; P=0.001, 0.000). The skin lesions observed were severe in the simple irradiation group. The skin in the irradiated+CD25 group was almost intact at 4 weeks, and hair removal and peeling still occurred at 8 weeks. Hematoxylin eosin staining showed that relative to that in the control group, the lung tissue structure of the mice in the irradiated group was destroyed, the alveolar wall was thickened, and the extracellular matrix was increased at 4 and 8 weeks after irradiation. The lung tissue of the irradiated+CD25 group was intact and the alveolar wall was slender. As indicated by the changes in inflammatory factors, relative to that in the control group, the secretion of IL-17A and IL-4 in the lung tissue of mice increased in the simple irradiation group ( t=-8.492, -15.796; P=0.001, 0.000) at 4 weeks. After 8 weeks of irradiation, the levels of TGF-β1 and IL-17A increased significantly ( t=-11.072, -7.167; P=0.000, 0.002), and the levels of IL-2 decreased at 4 and 8 weeks. IFN-γ secretion increased at 4 weeks ( t=-27.393, P=0.000) and decreased at 8 weeks. Relative to that in the simple irradiation group, TGF-β1 and IL-17A decreased ( t=6.037, 4.524, 5.496, 4.772; all P=0.000), IFN-γ increased ( t=-7.006, -12.565; P=0.002, 0.000) at 4 and 8 weeks, and IL-2 and IL-4 decreased at 4 weeks and increased significantly at 8 weeks ( t=2.866, -9.090, 8.833, -7.191; all P=0.000) in the irradiation+CD25 group. Conclusion:The differentiation of Tregs occurs in the lung tissue of mice with radiation-induced lung injury and promotes the development of radiation-induced lung injury by secreting TGF-β1 pro-inflammatory factor while interfering with the balance of helper T cell (Th)1/Th2 type cytokines.
The delineation of brain tumor margins has been a challenging objective in neurosurgery for decades. Despite the development of various preoperative imaging techniques, the current methodology is still insufficient for clinical practice. We present an intraoperative optical intrinsic signal imaging system for brain tumor surgery and establish a data processing procedure model to localize tumors. From the experimental result of a glioblastoma patient, we observe a relative small oscillation of ΔHbD in tumor region and speculate that vessels in tumor region have poor ability to provide oxygen. 目的 分析妊娠合并甲状腺疾病对新生儿的影响,为临床提供参考.方法 选择2014年1月-2017年12月在松原市中心医院接受治疗的354例妊娠早期合并甲状腺功能异常妇女为研究对象,按照甲状腺病情是否控制分为控制组(201例)和未控制组(153例),跟踪两组孕妇妊娠结局和新生儿情况.结果 控制组孕妇血压过高、血糖过高以及贫血发生率分别为7.46%、4.48%、3.98%,未控制组为15.03%、16.34%、9.15%,两组比较,差异有统计学意义(P<0.05);控制组孕产妇流产率以及新生儿低体重率分别为1.49%、2.49%,未控制组为5.23%、7.96%,两组比较,差异有统计学意义(P<0.05).结论 早期妊娠合并甲状腺疾病对新生儿以及妊娠结局能够产生一定的负面影响,在临床应引起足够的重视,积极控制病情,促使母婴安全健康,尤其是在妊娠早期,应尽可能将妊娠期女性甲状腺功能控制在正常范围内.
目的 探讨托伐普坦治疗急性心肌梗死(AMI)合并心力衰竭的应用疗效及对心肾功能的影响.方法 选取AMI合并心力衰竭患者80人,随机分成托伐普坦组和常规治疗组,常规治疗组给予心力衰竭常规药物治疗:硝酸酯类药物、利尿剂、血管紧张素转化酶抑制剂(ACEI)/血管紧张素受体拮抗剂(ARB)、β受体阻滞剂等.托伐普坦组在常规抗心力衰竭治疗的基础上应用托伐普坦7.5~15.0 mg,每日1次,共观察7 d.通过症状改善情况、心脏超声心功能指标左心室射血分数(LVEF)、左心室舒张末期内径(LVEDD)、血清脑钠肽(BNP)水平、尿量、尿素氮(BUN)、肌酐(CR)、血清钠水平等指标评估其疗效及对心肾功能的影响.结果 两组心力衰竭症状均明显改善,总有效率差异无统计学意义(P>0.05),但托伐普坦组较常规治疗组症状明显改善比例更高(P<0.05).两组患者LVEF治疗前后均未见明显变化(P>0.05).两组治疗后血清BNP水平均明显下降(P<0.05),托伐普坦组与常规治疗组比较差异有统计学意义(P<0.05).两组治疗前后BUN、CR均未见明显变化(P>0.05).托伐普坦组7 d平均24 h尿量较常规治疗组明显增加(P<0.05).托伐普坦组治疗后血清钠水平较常规治疗组明显升高(P<0.05).结论 托伐普坦治疗AMI合并心力衰竭疗效良好,对心肾功能无不良影响.
目的 探讨护理风险管理在老年心肺系统疾病患者护理中的应用价值及效果.方法 选取我院2015年5月~2017年5月期间收治的246例老年心肺疾病患者作为临床研究对象,按照随机数字表法将其随机分为观察组和对照组,各123例.对照组采用常规护理管理模式,观察组应用护理风险管理模式,比较两组患者的护理疗效、护理风险事件发生率及护理满意度.结果 根据统计分析结果,经过不同的护理管理之后,观察组患者护理疗效合格率为96.75% ,明显高于对照组的86.18% ,差异有统计学意义(χ2=9.807,P=0.01);住院期间对照组发生差错型护理风险事件8例,护理操作不当5例,投诉型风险事件5例,意外型风险事件9例,护理风险事件发生率21.95% ;观察组差错型护理风险事件2例,护理操作不当1例,投诉型风险事件1例,意外型风险事件3例,护理风险事件发生率5.69% .观察组护理风险事件发生率明显低于对照组,差异有显著统计学意义(χ2=13.652,P=0.00);对照组患者对于护理工作的满意度为78.86% ,观察组为93.50% ,观察组患者满意程度高于对照组,差异有统计学意义(χ2=15.303,P=0.00).结论 护理风险管理能够有效提高老年心肺系统疾病患者的护理合格率,降低护理风险事件发生率,提高患者的护理满意度,值得临床推广应用.
目的 检测子痫前期血清降钙素原(PCT)和C-反应蛋白(CRP)水平,为子痫前期的临床诊断、严重程度及预后评估提供依据.方法 选择2015年1月-2015年10月吉林省妇幼保健院子瘤前期患者80例(研究组),轻度、重度各40例,随机选取同期正常妊娠晚期孕妇50例(正常对照组).结果 研究组患者血清PCT、CRP及阳性率分别为(2.38±0.57)μg/L、(22.96±1.13) mg/L和88.75%、83.75%,均高于正常对照组,差异有统计学意义(P<0.05);严重患者血清PCT、CRP及阳性率分别为(3.45±1.17) μg/L、(24.24±1.20) mg/L和95.00%、90.00%,均高于轻度患者,差异有统计学意义(P<0.05),研究组治疗后患者血清PCT、CRP及阳性率分别为(0.52±0.36)μg/L、(8.05±0.79) mg/L和10.00%、6.25%,均低于治疗前,治疗前后比较,差异有统计学意义(P<0.05).结论 子痫前期患者血清PCT、CRP显著升高,且随着严重程度而上升,治疗后血清PCT、CRP均下降,有助于子痫前期患者的早期诊断和预后评估.
现代研究表明,补阳还五汤对脑缺血患者具有神经保护作用,可有效改善其神经行为学功能、减少脑缺血面积、降低梗死率和缺血再灌注损伤〔1〕;并可以刺激神经增殖、调节血管内皮生长因子的表达〔2〕,促进神经细胞的生长和分化、并抑制神经细胞凋亡〔3〕。本文就近年来有关补阳还五汤改善脑缺血作用机制的研究进行综述。
Objective To investigate the effectiveness, tolerability and safety of carboplatin in combination with temozolomide (TMZ) dose-dense regimen for the treatment of recurrent glioblastoma (GBM).Methods From February 2013 to March 2014, 9 adult patients with recurrent GBM received carboplatin in combination with temozolomide dose-dense regimen at the Department of Radiotherapy, the First Hospital of Jilin University were collected.The specific regimen: carboplatin 350 mg/m2 at day 1;TMZ 150 mg · m-2 · d-1, at day 1-7 and day 15-21, and 28 days were a circle.The follow-up time ranged from 5 to 16 months.Results Four cycles of chemotherapy were performed in 7 patients;5 cycles of chemotherapy were performed in 2 patients.The main toxic reactions were hematological toxic reactions,gastrointestinal reactions, and fatigue.Three patients had grade Ⅰ-Ⅱ of bone marrow suppression;6 patients had grade Ⅲ-Ⅳ of bone marrow suppression.All patients had mild to moderate gastrointestinal reactions including anorexia and vomiting.All the above adverse reactions were alleviated after symptomatic treatment;none of them had treatment-related deaths.Five patients died of tumor progression.The progression-free survival rate of 6 months was 44% (4/9);the imaging remission rate (PR) was 44% (4/9);the symptom improvement rate was 67% (6/9);and the 1-year survival rate was 22% (2/9).Conclusions Carboplatin in combination with TMZ dose-dense regimen for the treatment of recurrent GBM was effective.Most of the patients with 4-5 cycles of chemotherapy could tolerate it.No severe irreversible adverse reactions occurred.
目的 用Meta分析来评价姜在预防术后恶心呕吐方面的临床效果.方法 通过检索Pubmed、Web of science、Google scholar等数据库,搜集有关用姜来预防术后恶心呕吐的随机对照实验研究,应用R统计软件(R-3.1.3)对数据进行分析.结果 经过评价和筛选,纳入8篇随机对照研究.Meta分析结果显示:与对照组相比,姜具有预防术后恶心呕吐的作用,并且与对照组相比有统计学意义.结论 就目前已有的文献来看,姜在预防术后恶心呕吐上安全有效.尚有待开展高质量的多中心、随机、盲法、对照的临床研究进一步证实.
The 2-hydroxyl of -cyclodextrins can be selenized,producing an artificial simulation GPX enzyme of selenium which has wide application prospects.In order to carry on the further research,a lot of products were needed.Middle scale preparation of 2-SeCD was done based on the experiments of the original research and the amount was expanded for original 50 times.7 experimental results showed that the technologies were perfect and a large number of products were obtained.The structure of product obtained by middle scale preparation was verified by a series of characterization experiments such as carbon and hydrogen analysis,IR,NMR mass spectrometry analysis,etc.GPX activity of 2-SeCD was measured as 5.2 U/mol.The results of the biological effect test showed that low concentrations of 2-SeCD can reduce cell death but high concentrations of 2-SeCD inhibit cell growth.Apoptosis of Hela cells induced by 2-SeCD was found through morphological observation and DNA ladder detection,moreover,its mechanism was identified as the intrinsic mitochondrial apoptosis pathway by relative activity of caspase-3 assay.
岩斜区肿瘤向上可侵及岩尖、小脑幕切迹、Meckel囊、鞍旁及海绵窦,向下外可累及内听道甚至颈静脉孔;向中线可累及脑干及椎、基底动脉,肿瘤体积巨大时可累及一侧的第Ⅲ~Ⅺ对脑神经等重要结构,而肿瘤又多为良性,其手术切除一直是神经外科的难点[1],经岩乙状窦前入路和颞枕经小脑幕-经岩嵴入路都是治疗岩斜区域的肿瘤手术入路,二者的显露范围有一定的重叠。
In content based image retrieval,computation complexity will increase seriously as the demension of the image feature increases.When reducing the vector dimension with locally linear embedding algorithm,the number k of nearest neighbors should be determined.In this paper we propose a variable k neighbors LLE(VK-LLE) method based on the distribution of the image feature.The VK-LLE method can reduce the vectors dimension with keeping their original topology structure into a lower dimension space.Experiment shows that the proposed VK-LLE method can achieve higher precision rate in content based image retrieval problem.
目的:探讨妊娠中晚期凝血4项指标变化。方法:选择正常体检非孕女性50例,孕中期80例,孕晚期105例,采用德国BE公司四通道半自动凝血分析仪测定PT、APTT、TT、FIB 4项指标,对结果进行分析。结果:妊娠中晚期FIB含量明显增高,PT、APTT时间明显缩短,TT无明显变化。结论:对妊娠中晚期孕妇进行凝血4项指标检测可早期发现凝血功能的改变,对于预防DIC的发生和及时治疗有重要意义。
<正>胆石症是我国的常见病。目前的治疗方法均属于有创性手段,且均有禁忌证或相对禁忌证。因此,胆石病的非手术疗法是当前临床研究的热点。本研究采用我科研制的计算机辅助控制胆囊结石自动灌注溶石仪(DRY-03A胆石溶解仪)进行体内外溶石试验,为进一步研发和应用提供依据。
