As artificial intelligence technology gains widespread adoption in biomedicine, the exploration of integrating biofluidic Raman spectroscopy for enhanced disease diagnosis opens up new prospects for the practical application of Raman spectroscopy in clinical settings. However, for systemic lupus erythematosus (SLE), origin Raman spectral data (ORS) have relatively weak signals, making it challenging to obtain ideal classification results. Although the surface enhancement technique can enhance the scattering signal of Raman spectroscopic data, the sensitivity of the SERS substrate to airborne impurities and the inhomogeneous distribution of hotspots degrade part of the signal. To fully utilize both kinds of data, this paper proposes a two-branch residual-attention network (DBRAN) fusion technique, which allows the ORS to complement the degraded portion and thus improve the model's classification accuracy. The features are extracted using the residual module, which retains the original features while extracting the deep features. At the same time, the study incorporates the attention module in both the upper and lower branches to handle the weight allocation of the two modal features more efficiently. The experimental results demonstrate that both the low-level fusion method and the intermediate-level fusion method can significantly improve the diagnostic accuracy of SLE disease classification compared with a single modality, in which the intermediate-level fusion of DBRAN achieves 100% classification accuracy, sensitivity, and specificity. The accuracy is improved by 10% and 7% compared with the ORS unimodal and the SERS unimodal modalities, respectively. The experiment, by fusing the multimodal spectral, realized rapid diagnosis of SLE disease by fusing multimodal spectral data, which provides a reference idea in the field of Raman spectroscopy and can be further promoted to clinical practical applications in the future.
A dual-target molecular parallel detection method is proposed to detect multiple pesticides on a single sensor based on a double quantum dot/porous silicon optical biosensor. We fabricated porous silicon Bragg devices with broader forbidden bands to enhance the fluorescence from various quantum dots. Different quantum dots were labeled on complementary strands of nucleic acid aptamers and reacted with immobilized aptamers on the pore walls of the porous silicon pores. Finally, upon the addition of the pesticides of acetamiprid and profenofos, the complementary strands of the pesticides were replaced due to the stronger specific binding of the pesticides to their respective nucleic acid aptamers, and the fluorescence of the quantum dots was reduced. The detection of different pesticides was accomplished by monitoring the changes in the fluorescence of different quantum dots before and after the biological reaction. The detection limits were 2.64 nM for acetamiprid and 1.05 nM for profenofos. Our method was sensitive, cost-effective, and fast. Furthermore, our method could be applied to the efficient detection of multiple classes of chemical contaminants and nontargeted risk agent screening.
Cystic echinococcosis (CE) is a grievous zoonotic parasitic disease. Currently, the traditional technology of screening CE is laborious and expensive, developing an innovative technology is urgent. In this study, we combined serum fluorescence spectroscopy with machine learning algorithms to develop an innovative screening technique to diagnose CE in sheep. Serum fluorescence spectra of Echinococcus granulosus sensu stricto-infected group (n = 63) and uninfected E. granulosus s.s. group (n = 60) under excitation at 405 nm were recorded. The linear support vector machine (Linear SVM), Quadratic SVM, medium radial basis function (RBF) SVM, K-nearest neighbor (KNN), and principal component analysis-linear discriminant analysis (PCA-LDA) were used to analyze the spectra data. The results showed that Quadratic SVM had the great classification capacity, its sensitivity, specificity, and accuracy were 85.0%, 93.8%, and 88.9%, respectively. In short, serum fluorescence spectroscopy combined with Quadratic SVM algorithm has great potential in the innovative diagnosis of CE in sheep.
Cervical cancer is the most common reproductive malignancy in the female reproductive system. The incidence rate and mortality rate of cervical cancer among women in China are high. In this study, Raman spectroscopy was used to collect tissue sample data from patients with cervicitis, cervical precancerous low-grade lesions, cervical precancerous high-grade lesions, well differentiated squamous cell carcinoma, moderately differentiated squamous cell carcinoma, poorly differentiated squamous cell carcinoma and cervical adenocarcinoma. The collected data were preprocessed using an adaptive iterative reweighted penalized least squares (airPLS) algorithm and derivatives. Convolutional neural network (CNN) and residual neural network (ResNet) classification models were constructed to classify and identify seven types of tissue samples. The attention mechanism efficient channel attention network (ECANet) module and squeeze-and-excitation network (SENet) module were combined with the established CNN and ResNet network models, respectively, to make the models have better diagnostic performance. The results showed that efficient channel attention convolutional neural network (ECACNN) had the best discrimination, and the average accuracy, recall, F1 and AUC values after five cross-validations could reach 94.04%, 94.87%, 94.43% and 96.86%, respectively.
Ag2O-Ag-PSi (porous silicon) surface-enhanced Raman scattering (SERS) chip was successfully synthesized by electrochemical corrosion, in situ reduction and heat treatment technology. The influence of different heat treatment temperature on SERS performance of the chip is studied. The results show that the chip treated at 300 degrees C has the best SERS performance. The chip was composed of Ag2O-Ag nano core shell with a diameter of 40-60 nm and porous silicon substrate. Then, the optimized chip was used to perform SERS test on serum samples from 30 healthy volunteers and 30 early breast cancer patients, and the baseline was corrected by LabSpec6 software. Finally, the data were analyzed by principal component analysis combined with t-distributed Stochastic Neighbor Embedding (PCA-t-SNE). The results showed that the accuracy of the improved substrate combined with multivariate statistical method was 98%. The shelf life of the chips exceeded six months due to the presence of the Ag2O shell. This study provides a basis for developing a low-cost rapid and sensitive early screening technology for breast cancer.
Breast cancer is one of the most common cancers in women worldwide, and it seriously threatens people’s lives and health. Breast Imaging Reporting and Data System is developed as a standardized system or tool for reporting breast mammograms, where different grades of diagnosis and treatment are critical to the survival rate and survival time of patients. Efficient computer-aided diagnosis of breast tumors based on computer vision models can better assist physicians in selecting effective treatment options, thereby reducing patient mortality. Therefore, early detection and early treatment are of great significance to patients with breast disease. In this study, a new image enhancement framework, called Image Negatives and Contrast Limited Adaptive Histogram Equalization Image Enhancement, was created for the first time based on the comparison of a set of multiple data preprocessing methods for detecting normal, benign, and probably benign breasts. The ResNet-50 pre-trained neural network was used for feature extraction and the classification results were compared on K-nearest neighbor, Random Forest, and Support Vector Machine classifiers. The evaluation indexes adopted in this paper include confusion matrix, precision, sensitivity, F1 Score, etc. These evaluation indexes can be used to evaluate the model in a very comprehensive and accurate way. The experiments show that the KNN classifier has the best classification result, the classification accuracy is 85%, and the AUC is 0.89. It is proved that mammography, as a non-invasive screening tool, has certain practical significance in effectively evaluating tumor grade and its clinical application.
Parotid tumors are among the most prevalent tumors in otolaryngology, and malignant parotid tumors are one of the main causes of facial paralysis in patients. Currently, the main diagnostic modality for parotid tumors is computed tomography, which relies mainly on the subjective judgment of clinicians and leads to practical problems such as high workloads. Therefore, to assist physicians in solving the preoperative classification problem, a stacked generalization model is proposed for the automated classification of parotid tumor images. A ResNet50 pretrained model is used for feature extraction. The first layer of the adopted stacked generalization model consists of multiple weak learners, and the results of the weak learners are integrated as input data in a meta-classifier in the second layer. The output results of the meta-classifier are the final classification results. The classification accuracy of the stacked generalization model reaches 91%. Comparing the classification results under different classifiers, the stacked generalization model used in this study can identify benign and malignant tumors in the parotid gland effectively, thus relieving physicians of tedious work pressure.
Surface-enhanced Raman spectroscopy (SERS), as a rapid, non-invasive and reliable spectroscopic detection technique, has promising applications in disease screening and diagnosis. In this paper, an annealed silver nanoparticles/porous silicon Bragg reflector (AgNPs/PSB) composite SERS substrate with high sensitivity and strong stability was prepared by immersion plating and heat treatment using porous silicon Bragg reflector (PSB) as the substrate. The substrate combines the five deep learning algorithms of the improved AlexNet, ResNet, SqueezeNet, temporal convolutional network (TCN) and multiscale fusion convolutional neural network (MCNN). We constructed rapid screening models for patients with primary Sjögren's syndrome (pSS) and healthy controls (HC), diabetic nephropathy patients (DN) and healthy controls (HC), respectively. The results showed that the annealed AgNPs/PSB composite SERS substrates performed well in diagnosing. Among them, the MCNN model had the best classification effect in the two groups of experiments, with an accuracy rate of 94.7% and 92.0%, respectively. Previous studies have indicated that the AgNPs/PSB composite SERS substrate, combined with machine learning algorithms, has achieved promising classification results in disease diagnosis. This study shows that SERS technology based on annealed AgNPs/PSB composite substrate combined with deep learning algorithm has a greater developmental prospect and research value in the early identification and screening of immune diseases and chronic kidney disease, providing reference ideas for non-invasive and rapid clinical medical diagnosis of patients.
In recent years, Raman spectroscopy combined with deep learning techniques has been widely used in various fields such as medical, chemical, and geological. However, there is still room for optimization of deep learning techniques and model compression algorithms for processing Raman spectral data. To further optimize deep learning models applied to Raman spectroscopy, in this study time, accuracy, sensitivity, specificity and floating point operations numbers(FLOPs) are used as evaluation metrics to optimize the model, which is named RamanCompact(RamanCMP). The experimental data used in this research are selected from the RRUFF public dataset, which consists of 723 Raman spectroscopy data samples from 10 different mineral categories. In this paper, 1D-EfficientNet adapted to the spectral data as well as 1D-DRSN are proposed to improve the model classification accuracy. To achieve better classification accuracy while optimizing the time parameters, three model compression methods are designed: knowledge distillation using 1D-EfficientNet model as a teacher model to train convolutional neural networks(CNN), proposing a channel conversion method to optimize 1D-DRSN model, and using 1D-DRSN model as a feature extractor in combination with linear discriminant analysis(LDA) model for classification. Compared with the traditional LDA and CNN models, the accuracy of 1D-EfficientNet and 1D-DRSN is improved by more than 20%. The time of the distilled model is reduced by 9680.9s compared with the teacher model 1D-EfficientNet under the condition of losing 2.07% accuracy. The accuracy of the distilled model is improved by 20% compared to the CNN student model while keeping inference efficiency constant. The 1D-DRSN optimized with channel conversion method saves 60% inference time of the original 1D-DRSN model. Feature extraction reduces the inference time of 1D-DRSN model by 93% with 94.48% accuracy. This study innovatively combines lightweight models and model compression algorithms to improve the classification speed of deep learning models in the field of Raman spectroscopy, forming a complete set of analysis methods and laying the foundation for future research.
Cervical cancer is one of the most common cancers with a long latent period and slow onset process. Early and accurate identification of the stage of cervical cancer can significantly improve the cure rate and patient survival time. In this study, we collected 699 Raman spectral data of tissue sections from 233 different patients. We analyzed and compared the characteristics and differences of the mean Raman spectra of the seven tissues and pointed out the main differences in the biochemical composition of the seven tissues. In this study, 1D hierarchical convolutional neural network (H-CNN) is proposed by integrating the prior knowledge of hierarchical classification relations with the research of deep learning in Raman spectroscopy. H-CNN is based on CNN and is added with three network branches. Hierarchical classification is performed from coarse to fine for tissue samples of cervicitis, Low-grade Squamous Cell Carcinoma, High-grade Squamous Cell Carcinoma, Well Differentiated Squamous Cell Carcinoma, Moderately Differentiated Squamous Cell Carcinoma, Poorly Differentiated Squamous Cell Carcinoma and cervical adenocarcinoma. To evaluate the recognition performance of H-CNN, we compared it with traditional methods such as Bayesian classifier (NB), decision tree classifier (DT), support vector machine classifier (SVM) and CNN. The experimental results show that H-CNN can accurately identify different classes of tissue sections and has apparent advantages in several aspects such as recognition accuracy, stability and sensitivity compared with the other four traditional recognition methods. The classification Macro-Accuracy of H-CNN can reach 94.91%, Macro-Recall can reach 95.31%, Macro-F1 can reach 95.23%, and Macro-AUC can reach 97.35%. The hierarchical classification method proposed in this study can diagnose patients more accurately. This could lay the foundation for further research on Raman spectroscopy as a clinical diagnostic method for cervical cancer.
The aim of this study was to explore the feasibility of Raman spectroscopy combined with computer algorithms in the diagnosis of primary Sjögren syndrome (pSS). In this study, Raman spectra of 60 serum samples were acquired from 30 patients with pSS and 30 healthy controls (HCs). The means and standard deviations of the raw spectra of patients with pSS and HCs were calculated. Spectral features were assigned based on the literature. Principal component analysis (PCA) was used to extract the spectral features. Then, a particle swarm optimization (PSO)-support vector machine (SVM) was selected as the method of parameter optimization to rapidly classify patients with pSS and HCs. In this study, the SVM algorithm was used as the classification model, and the radial basis kernel function was selected as the kernel function. In addition, the PSO algorithm was used to establish a model for the parameter optimization method. The training set and test set were randomly divided at a ratio of 7:3. After PCA dimension reduction, the specificity, sensitivity and accuracy of the PSO-SVM model were obtained, and the results were 88.89%, 100% and 94.44%, respectively. This study showed that the combination of Raman spectroscopy and a support vector machine algorithm could be used as an effective pSS diagnosis method with broad application value.
Purpose Skin cancer is one of the ten most common cancer types in the world. Early diagnosis and treatment can effectively reduce the mortality of patients. Therefore, it is of great significance to develop an intelligent diagnosis system for skin cancer. According to the survey, at present, most intelligent diagnosis systems of skin cancer only use skin image data, but the multi-modal cross-fusion analysis using image data and patient clinical data is limited. Therefore, to further explore the complementary relationship between image data and patient clinical data, we propose multimode data fusion diagnosis network (MDFNet), a framework for skin cancer based on data fusion strategy. Methods MDFNet establishes an effective mapping among heterogeneous data features, effectively fuses clinical skin images and patient clinical data, and effectively solves the problems of feature paucity and insufficient feature richness that only use single-mode data. Results The experimental results present that our proposed smart skin cancer diagnosis model has an accuracy of 80.42%, which is an improvement of about 9% compared with the model accuracy using only medical images, thus effectively confirming the unique fusion advantages exhibited by MDFNet. Conclusions This illustrates that MDFNet can not only be applied as an effective auxiliary diagnostic tool for skin cancer diagnosis, help physicians improve clinical decision-making ability and effectively improve the efficiency of clinical medicine diagnosis, but also its proposed data fusion method fully exerts the advantage of information convergence and has a certain reference value for the intelligent diagnosis of numerous clinical diseases.
Echinococcosis is a severe zoonotic parasitic disease, and it is continuing to be a significant public health issue. The course of the disease is usually slow, and patients often remain asymptomatic for years. There is no standardized and widely accepted treatment, so early and accurate diagnosis is essential. Herein, this study utilized vibrational spectroscopic techniques, namely Raman and Fourier Transform Infrared (FTIR) spectroscopy, to quickly and accurately distinguish hepatic echinococcosis (HE) patients' serum from the healthy group. Serum samples were collected from HE patients as well as healthy control subjects, and then the Raman and FTIR spectra of the two groups were recorded. After a series of pre-processing, support vector machines (SVMs) were then used to establish the classification models for the two spectral data sets. The performance of each diagnostic model was evaluated using leave-one-out cross-validation (LOOCV) and hold-out validation methods, respectively. For the distinction between HE and healthy groups, these two spectroscopic techniques had achieved satisfactory classification results, and the diagnostic capabilities of the Raman technique were comparable to that of the FTIR method. The results demonstrate that vibrational spectroscopy has great potential in the rapid and accurate detection of HE and is expected to make up for the shortcomings of the existing clinical diagnosis methods.
In this paper, a new method for pesticide detection based on porous silicon microcavities was proposed. First, the aptamer of acetamiprid was fixed in the functionalized porous silicon microcavity. Then, the complementary strand of the aptamer modified with graphene quantum dots was hybridized with the aptamer, and the effective refractive index of the porous silicon microcavity increased. When acetamiprid was added, it bound to the aptamer due to its stronger specificity. The aptamer separated from the complementary strand of the coupled graphene quantum dots and bound to acetamiprid, resulting in a decrease in the effective refractive index, and its central wavelength of reflection spectrum was blue shifted, and the blueshift increased with the increase of acetamiprid concentration. The relationship between the change in central wavelength and the concentration of acetamiprid was analysed, and the detection limit of acetamiprid was 151 nm.
Breast cancer is a malignant tumor with the highest incidence rate in women. Current diagnostic methods are time-consuming, costly, and dependent on physician experience. In this study, we used serum Raman spectroscopy combined with multiple classification algorithms to implement an auxiliary diagnosis method for breast cancer, which will help in the early diagnosis of breast cancer patients. We analyzed the serum Raman spectra of 171 invasive ductal carcinoma (IDC) and 100 healthy volunteers; The analysis showed differences in nucleic acids, carotenoids, amino acids, and lipid concentrations in their blood. These differences provide a theoretical basis for this experiment. First, we used adaptive iteratively reweighted penalized least squares (airPLS) and Savitzky-Golay (SG) for baseline correction and smoothing denoising to remove the effect of noise on the experiment. Then, the Principal component analysis (PCA) algorithm was used to extract features. Finally, we built four classification models: support vector machine (SVM), decision tree (DT), linear discriminant analysis (LDA), and Neural Network Language Model (NNLM). The LDA, SVM, and NNLM achieve 100% accuracy. As supplementary, we added the classification experiment of the raw data. By comparing the experimental results of the two groups, We concluded that the NNLM was the best model. The results show the reliability of the combination of serum Raman spectroscopy and classification models under large sample conditions.
Diabetes mellitus is a global public health problem, and the epidemic situation in China is particularly serious. The prevalence of the disease has been increasing in recent years, and the number of patients is the highest in the world. Diabetes has become another chronic non-communicable disease that seriously endangers the health of our people after cardiovascular and cerebrovascular diseases and tumors. In this study, urine sample data were collected from 37 diabetic patients and 37 healthy volunteers using Raman spectroscopy. The collected data were preprocessed using an adaptive iterative reweighted penalized least squares (airPLS) algorithm and a polynomial Savitzky-Golay smoothing algorithm. After extracting features using principal component analysis (PCA) dimensionality reduction algorithm, ResNet, support vector machine (SVM) and linear discriminant analysis (LDA) classification models were selected to classify and identify diabetic patients and healthy controls. The results show that ResNet has the best discrimination effect, and the average accuracy, recall and F1-score can reach 84.28%, 86.20% and 84.02% respectively after five cross-validations, and the area under the subject working characteristic (ROC) curve is 0.93. The experimental results show that the model established in this paper is simple to operate, highly accurate and has good reference value for rapid screening of diabetes.
In recent years, liver cancer has caused great harm to human health. This study aims to use serum Raman spectroscopy combined with deep learning algorithms to classify liver cancer patients and control groups. For improving the robustness of models, we added equal proportions of Gaussian white noise of 5, 10, 15, 20, 25 dBW to enhance the data, and compared the results of convolutional neural networks and long short-term memory networks which show that the convolutional neural network combined with 10-fold data enhancement was better. The accuracy was 96.95%, indicating the great potential of this experimental model in liver cancer detection.
Keratitis and conjunctivitis are the most common ocular diseases, their symptoms are similar and easy to confuse, however infectious conjunctivitis is highly contagious. If misdiagnosed, it may worsen the disease and pose a threat to public health.This is a preclinical study to propose a method for rapid and accurate screening of keratitis and conjunctivitis by combining tear Raman spectroscopy with deep learning models that may be applied to clinical applications in the future.The tears of 16 cases of keratitis patients, 13 cases of conjunctivitis patients and 46 cases of healthy subjects were collected, and their Raman spectra were compared and analyzed. By adding different decibels of Gaussian white noise to expand the data, the performance of the tear Raman spectra with a large sample size in the deep learning model was discussed. Principal component analysis (PCA), partial least squares (PLS) and maximum correlation minimum redundancy (mRMR) were used for feature extraction. The processed data were imported into convolutional neural network (CNN) and recurrent neural network (RNN) depth models for classification. After the data were enhanced and processed by PLS, the highest classification accuracy of healthy subjects and keratitis patients, healthy subjects and conjunctivitis patients, and keratitis and conjunctivitis patients reached 94.8%, 95.4%, and 92.7%, respectively. The results of this study show that the use of large sample tear Raman spectra data combined with PLS feature extraction and depth learning algorithms may have great potential in clinical screening of keratitis and conjunctivitis.
In this paper, we investigated the feasibility of using serum Raman spectroscopy and multivariate analysis method to discriminate echinococcosis and liver cirrhosis from healthy volunteers. Raman spectra of serum samples from echinococcosis, liver cirrhosis, and healthy volunteers were recorded under 532 nm excitation. The normalized mean Raman spectra revealed specific biomolecular differences associated with the disease, mainly manifested as the contents of β carotene in the serum of patients with echinococcosis and liver cirrhosis were lower than those of healthy people. Furthermore, principal components analysis (PCA), combined with linear discriminant analysis (LDA), was adopted to distinguish patients with echinococcosis, liver cirrhosis, and healthy volunteers. The overall diagnostic accuracy based on the PCA-LDA algorithm was 87.7 %. The diagnostic sensitivities to healthy volunteers, patients with echinococcosis, and liver cirrhosis were 92.5 %, 81.5 %, and 89.1 %, and the specificities were 93.2 %, 96.1 %, and 92.4 %, respectively. This exploratory work demonstrated that serum Raman spectroscopy technology combined with PCA-LDA diagnostic algorithm has great potential for the non-invasive identification of echinococcosis and liver cirrhosis.
Glioma has a low cure rate and a high mortality rate. Therefore, correct diagnosis and treatment are essential for patients. This research aims to use mid-infrared spectroscopy combined with machine learning algorithms to identify patients with glioma. The glioma infrared spectra and the control group serum are smoothed and normalized, then the principal component analysis (PCA) algorithm is used to reduce the data dimensionality, and finally, the particle swarm optimization-support vector machine (PSO-SVM), backpropagation (BP) neural network and decision tree (DT) model are established. The classification accuracy of the three models was 92.00 %, 91.83 %, 87.20 %, and the AUC values were 0.919, 0.945, and 0.866, respectively. The results show that PCA-PSO-SVM has a better classification effect. This study shows that mid-infrared spectroscopy combined with machine learning algorithms has great potential in the application of non-invasive, rapid and accurate identification of glioma patients.
