Objective To observe the expression of vimentin (Vim) after silence of interleukin-1β (IL-1β) in rats with spinal cord contusion (SCC). Methods The model of SCC was established in 30 Sprague-Dawley rats with Allen's method. The rats were randomized into vector group (n=15) and silence group (n=15), which were injected blank lentivirus vector and vector of IL-1β siRNA, respectively; and divided in three, seven and 28 days subgroups. The relationship between IL-1βand Vim was predicted with GeneMANIA bioinformatics. The expression of Vim protein and mRNA in spinal cord was detected with immunohistochemistry and real-time quantitative polymerase chain reaction. Results GeneMANIA bioinformatic analysis indicated that there was some direct and indirect relationship between IL-1β and Vim. The Vim protein and mRNA expressed in the spinal cord, and was less in the silence group than in the vector group (t>2.875, P<0.05). Conclusion Silence of IL-1β can inhibit the expression of Vim in SCC rats, which may promote the recovery of spinal cord function.
目的 探讨白细胞介素-6 (IL-6)在脊髓损伤SD大鼠白质中的定位及表达变化,分析IL-6对大鼠神经行为学变化的影响.方法 66只成年SD大鼠,随机分为假手术组和脊髓顿挫损伤(Spinal cord contusion,SCC)术后12h、3d、5d、7d、14d组.用BBB神经功能评分评价脊髓顿挫损伤后大鼠的运动功能;采用RT-qPCR技术检测IL-6 mRNA含量变化;采用免疫组化染色,定位观察IL-6在脊髓白质中的分布.结果 SCC后大鼠后肢运动功能消失,BBB评分显示术后5d、7d、14d较Sham组明显降低(P<0.05).RT-qPCR表明IL-6 mRNA在SCC后12h表达峰值,与假手术组相比差异有统计学意义(P<0.05) HL-6 mRNA在第5d明显下降,与12h相比较具有显著性差异(P<0.05).免疫组化结果显示:IL-6主要分布在脊髓白质区的小神经胶质细胞和神经纤维.结论 SCC后大鼠后肢运动功能障碍随时间推移而得到一定的改善,其机制可能与IL-6的表达减少相关,因此,脊髓白质中IL-6的表达与运动功能恢复有关.
Objective To observe the effects of Salvia Miltiorrhiza on repair area of skin wound and expression level of vascular endothelial growth factor(VEGF)in skin wound repair model of rat.Methods 36 SD(Sprague-dawley)rats were randomly divided into control group and Salvia Miltiorrhiza group,18 rats in each group,all rats were subjected to the full-thickness skin excision.The Salvia Miltiorrhiza group was treated with Salvia Miltiorrhiza injection to repair the wound,and the control group was treated with the same dose of physiological saline.The wound unrepaired area was measurement of all the animal models,and the pathologicchange and the expression change of VEGF were observed by HE staining and immunohistochemistry.Results Determination of unrepaired area of skin wound showed that after injury 1d,3d,7d control group and Salvia Miltiorrhiza group of rat skin wounds unrepaired area was gradually reduced,but it was lower than the control group in the Salvia Miltiorrhiza group after injury 3d,7d.In the Salvia Miltiorrhiza group the erythrocyte sedimentation in vessels was lighter than the control group and the number of new capillaries was more than the control group by HE staining after 3d of injury.Immunohistochemical staining showed that VEGF was located in animal skin epidermal cells,sebaceous gland,hair follicle,neutrophils,macrophages,fibroblasts,but fibroblast was in the majority.After injury 1d,3d,the VEGF expression intensity in the Salvia Miltiorrhiza group was higher than the control group(P<0.05).Conclusion Salvia Miltiorrhiza can promote the repair of traumatic skin in rats by increasing the expression level of VEGF.
目的 通过慢病毒介导的RNA技术干扰IL-6的表达,探究脊髓顿挫损伤后Caspase-3的表达变化.方法 Allen's打击法制备脊髓顿挫伤(SCC)模型,BBB评分确认造模成功;免疫组织化学和Q-PCR技术定位和定量测定IL-6和Caspase-3的表达变化;Gene MANIA网站预测IL-6与Caspase-3的联系;慢病毒介导的RNA干扰技术构建IL-6干扰模型后,Q-PCR技术定量测定Caspase-3的表达变化.结果 SCC后SD大鼠BBB评分显著低于假手术组(P<0.05);IHC结果显示:IL-6和Caspase-3均在在脊髓白质胶质细胞表达;Q-PCR结果显示:IL-6在损伤后12h表达显著增加,3d、28 d表达持续下降;Caspase-3在损伤后12h、3d、28 d表达持续下降.生物信息学分析结果:IL 6和Caspase 3在大鼠体内存在共定位关系.干扰组Caspase-3表达比空载组显著降低(P<0.05).结论 在SCC后,抑制IL-6表达可降低Caspase-3表达水平.
Objective To explore the expression of aquaporin (AQP)-4 in white matter of spinal cord after spinal cord contusion (SCC). Methods 88 adult Sprague-Dawley rats were assigned randomly to sham operation group and SCC group. The model was established by Al-len's method. BBB sore was used to assess the motor function of rats. The relative expression of AQP-4 mRNA was determined by Q-PCR technique. The localization of AQP-4 was observed by immunohistochemistry. Results BBB score showed motor dysfunction in SCC group, and it increased 7 and 14 days after SCC (t>5.061, P<0.001). The level of AQP-4 mRNA decreased on the 1st and 3rd days (t>50.44, P<0.001), and increased on the 5th day (t=-3.968, P=0.001), and lasted until the 28th day (t=-4.227, P=0.001) compared with that on the 3rd day. The immunohistochemistry showed AQP-4 was located on the process of glial cell and vascular endothelial cells in white matter of spi-nal cord. Conclusion AQP-4 may play various roles at different stages in SCC.
目的:探究神经生物电刺激对原发性痛经(Primary dysmenorrhea,PD)大鼠P物质(Substance P,SP)的表达变化的影响.方法:将SD大鼠随机分为非模型对照组、模型对照组和实验组3组.实验组给予股部皮下注射已烯雌酚,每天一次,连续10d,第1天0.8 mg·只-1,第2-9天0.4 mg·只-1,第10天0.8 mg·只-1;第11天腹腔注射缩宫素2U·只-1;对实验组大鼠进行神经生物电刺激干预,并在1h、3h和7h后采用扭体评分进行运动评价.模型对照组只注射与实验组相同剂量的已烯雌酚与缩宫素,不进行神经生物电刺激干预,其余操作不变.非模型对照组不注射已烯雌酚与缩宫素,不进行神经生物电刺激干预,其余操作不变.免疫组织化学技术检测各组大鼠脊髓后角中SP的定位及表达变化,采用免疫荧光技术探究SP与脑源性神经营养因子(Brain derived neurotrophic factor,BDNF)在脊髓后角的共定位情况.结果:扭体评分显示实验组大鼠运动增加幅度明显小于模型对照组(P<0.05).SP主要分布于脊髓后角Ⅱ板层,所有时间点实验组SP表达均低于模型对照组,在3h表达差异最大(P<0.05).免疫荧光双标技术显示SP和BDNF在脊髓后角共表达.结论:神经生物电刺激能抑制SP的表达,影响原发性痛经,其机制可能与SP及BDNF共同作用有关.
Objective To explore the change of apoptosis factor Caspase-3 and Bcl-2 in the injured segment of rat with spinal cord injury after inhibiting lentivirus expression of inflammation factor TNF-α. To study the relationship between Caspase-3, Bcl-2, Bax and TNF-α in spinal cord injury. Mthods Spinal cord contusion model was prepared by Allen method. The relation between tumor necrosis factor alpha and Bcl-2, was predicted by the method of GeneMANIA bioinformatics. The RNA which was packaged by lentivirus constructed the RNA interference model of tumour necrosis factor alpha. After interference of tumor necrosis factor alpha, we used the method of QRT-PCR to assays the mRNA expression of Caspase-3 and Bcl-2 in spinal cord and detect of the localization of Caspase-3 and Bcl-2 by immunohistochemistry. Statistical analysis with SPSS17.0. Results SD rats had paraplegia and urinate retentaion because of spinal cord injury. The result of QRT-PCR showed that in the seventh day after SCC, the expression of Caspase-3 reduced significantly (P < 0.05) and Bcl-2 did not change significantly (P > 0.05). Immunohistochemistry experiment results showed that Caspase-3 Bcl-2 and Bax immunoreactive cells were observed in the neurons and glial cells of both white matter and gray matter in the spinal cord. The results were the same with QRT-PCR.. Conclusion TNF-α in rats after SCC can effectively regulate the ratio of Bcl-2 and Bax , and then regulate the expression of Caspase-3 , which may affect the function of apoptosis and function recovery after spinal cord injury.
目的 探究大鼠脊髓损伤后磷脂酰乙醇胺结合蛋白1 (PEBP1)的表达和意义.方法 本研究采用105只Sprague-Dawley雌性大鼠,随机分为假手术组和实验组.采用Allen's法制备大鼠脊髓损伤模型,应用免疫组织化学方法检测PEBP1在脊髓中的定位变达,采用实时定量PCR和蛋白免疫印迹检测PEBP1的mRNA和蛋白在脊髓中的表达变化.结果 免疫组织化学结果显示PEBP1在正常脊髓后角的神经胶质细胞的胞浆和胞核中表达,在脊髓损伤后与sham组相比PEBP1免疫阳性细胞数明显减少.此外,实时定量PCR显示PEBP1的mRNA在大鼠脊髓损伤后12h,1d,3d,7d,14d和28 d的表达量明显降低,与sham组相比差异具有统计学意义(P<0.01);免疫印迹实验显示,与sham组相比,PEBP1蛋白在大鼠脊髓损伤后的7d,14 d和28 d表达量降低,差异具有统计学意义(P<0.05).结论 脊髓损伤后PEBP1的mRNA和蛋白表达水平均降低,在脊髓后角的表达明显减少.提示脊髓损伤后PEBP1可能参与调控神经胶质细胞的存活和凋亡,进一步影响脊髓损伤后的感觉功能.
目的:观察大鼠脊髓顿挫伤后脑衰反应蛋白2(CRMP-2)在脊髓组织中的分布及基因表达变化.方法:采用Allen's重物法打击SD大鼠,制备脊髓顿挫伤模型.采用BBB评分方法评估大鼠后肢运动功能;采用免疫组织化学技术(IHC)观察CRMP-2在损伤脊髓中的定位,实时定量PCR (qRT-PCR)方法观察CRMP-2在大鼠脊髓顿挫伤后的基因表达变化.结果:脊髓顿挫伤后,BBB评分结果表明大鼠后肢运动功能明显下降,在损伤后1d,7d,14 d,21 d和28 d的BBB评分逐渐恢复,但是仍然低于对照组.免疫组织化学染色结果显示:CRMP-2在脊髓前角运动神经元和神经胶质细胞中广泛表达,在脊髓损伤后12 h,3d,7d,14 d和28 d,CRMP-2免疫阳性细胞数相比sham组均有明显的减少.定量qRT-PCR结果显示:脊髓顿挫伤后CRMP-2基因表达下降,并持续至术后28 d.结论:脊髓顿挫伤后,CRMP-2大量存在于脊髓前角运动神经元及神经胶质细胞中,在损伤脊髓中的基因表达持续下降.结果提示脊髓顿挫伤后CRMP-2可能影响神经细胞的生长,分化和迁移.
目的 对固体硅胶假体与多孔高密度聚乙烯(Medpor)假体用于隆颏进行系统评价.方法 计算机检索Pubmed文献数据库、中国知网中文数据库、万方数据库.收集所有相关使用生物材料隆颏术的临床报道,行Meta分析,以进行系统评价.结果 (1)硅胶感染率合并值为0.00,95%可信区间为[0.00;0.01],Medpor的感染率合并值为0.00,95%可信区间为[0.00;0.01],差异无统计学意义(P>0.05).(2)硅胶假体位置偏移率合并值为0.03,95%可信区间为[0.00;0.08],Medpor假体位置偏移率合并值为0.00,95%可信区间为[0.00;0.01],差异有统计学意义(P<0.01).(3)硅胶假体取出率合并值为0.17,95%可信区间为[0.00;0.34],Medpor的假体取出率合并值为0.01,95%可信区间为[0.00;0.02],差异有统计学意义(P<0.01).(4)硅胶假体骨质吸收发生率合并值为0.15,95%可信区间为[0.04;0.26],Medpor假体的骨质吸收发生率合并值为0.00,95%可信区间为[0.00;0.01],差异有统计学意义(P<0.01).结论 生物材料为隆颏术的发展带来了新进展,而Medpor与硅胶假体各有优缺点,应当根据患者不同需求及情况作出合适的选择.
目的:探讨在电刺激干预下大鼠脊髓损伤节段神经生长因子(NT4)和增殖细胞核抗原(PCNA)的表达及对脊髓损伤的影响.方法:成年雌性SD大鼠78只,随机分为3组:假手术组(n=6)、对照组(n=36)和实验组(n=36).Allen's法复制SD大鼠T9脊髓钝挫伤模型.实验组脊髓钝挫伤后给予夹脊穴和足三里穴位神经电刺激干预.神经行为学评分后,运用免疫组化和蛋白质印迹(Westem blot)来检测NT4、PCNA的表达变化.结果:行为学观察,实验组大鼠术后1周BBB评分显著高于对照组(P<0.05),低于假手术组(P<0.05).实验组第1-3天NT4表达显著升高(P<0.05),第7天达到峰值并且其表达明显高于对照组(P<0.05).实验组第1-3天PCNA表达降低,第7天达到最大差异(P<0.05).结论:电刺激干预下,脊髓组织中神经元中的NT4表达增强,电刺激能改善运动功能恢复.神经电刺激后PCNA表达减少,神经电刺激对神经干细胞的分化作用大于神经干细胞的再生增殖.
Objective To evaluate the effect of rehabilitation training on rats with spinal cord injuries by Meta-analysis.Methods Articles were searched from PubMed( ~ 2013),CNKI( 1989 ~ 2013),WanFang Data( ~ 2013),VIP( 1989 ~ 2013),quality of included article was assessed with Jadad scale,and available data was analyzed with RevMan 5.0 software.Results 287 related articles were identified,but only 11 eligible articles were included.The Meta-analysis of BBB score indicated that the rehabilitation training groups were better than control groups.The BBB score[weighted mean difference( WMD) = 1.87,95% CI( 1.50,2.33),Z = 10.02,P 0.01].There was significant diffence between two groups.Conclusion Rehabilitation training can improve the recover of hindlimb function.
Objective:To study the impact of transcutaneous electrical stimulation(TES) on the expressions of glial fibrillary acidic protein(GFAP) and nerve growth factor (NGF) after spinal cord injury(SCI) in rats. Method:Seventy-two adult male SD rats were randomly divided into 3 groups: normal group(A group, n=8), TES group(B group,n=32) and model group(C group,n=32). The incomplete SCI on T9 model was made by Allen's method. TES group was given electroacupuncture on the acupoint at 2cm above the injury part skin and right shank for 7 days. Immunohistochemistry and Western blotting techniques were used to detect the expressions of GFAP and NGF after BBB evaluation. Result:The BBB score in TES group increased to 21.00 (P0.05) compared with model group, The BBB scores in each SCI group were significantly less than that in normal group (P0.05). The result of immunohistochemistry and Western blotting within 7 days of the experiment showed in TES group compared with model group, the expression of GFAP reduced and the lowest value appeared at the 5th day (P0.05); NGF expression kept increasing continuously(P0.05). Conclusion:Within 7 days after SCI, TES can inhibit the expression of GFAP and stimulate the synthesis of endogenous NGF. So it may create a favorable microenvironment for nerve regeneration and reduce the formation of glial scar.
