目的 通过免疫组织化学(immunohistochemistry,IHC)对肿瘤细胞进行正确的免疫表型分析和精准组织病理学诊断.方法 采用非生物素两步法和全自动免疫组化染色仪进行免疫组化染色,显示CD20在肿瘤组织中表达.结果 全自动免疫组化仪与第一抗体不匹配导致假阳性的发生.结论 在诊断免疫组织化学中使用抗体和全自动免疫组化仪自体配套的二抗试剂盒的相容组合非常重要.
Objective To explore the expressions of Hofbauer cells in placental tissues of early-onset and late-onset severe preeclampsia.Methods A total of 72 severe preeclampsia patients giving birth to their babies in the hospital from March 2009 to December 2015 were collected and divided into early-onset severe preeclampsia group (23 patients,<34 gestational weeks) and late-onset severe preeclampsia group (49 patients,≥34 gestational weeks) according to the onset time of severe preeclampsia.Twenty-two normal pregnant women were selected from the hospital during the same period as control group.Immunohistochemistry was used to detect the expression levels of HMGB1,CD8,and CD163 in Hofbauer cells of placenta.Results HMGB1 was found in cell nucleus of trophocytes and Hofbauer cells,CD68 was found in cell membrance of Hofbauer cells,CD163 was found in cytoplasm and cell membrance of Hofbauer cells.Compared with control group,the numbers of Hofbauer cells with positive CD68 and CD163 in placenta of patients with late-onset severe preeclampsia decreased significantly (P<0.05).Compared with the patients with early-onset severe preeclampsia,the number of Hofbauer cells with positive CD68 in placenta of patients with late-onset severe preeclampsia decreased significantly (P<0.05),but there was no statistically significant difference in CD163 (P>0.05).Conclusion The decrease of Hofbauer cells may be involved in pathogenesis of severe preeclampsia,Hofbauer cell may be an important target of treatment of preeclampsia.
Objective To explore the expressions of high mobility group box B1 (HMGB1),interleukin-6 (IL-6),and transforming growth factor-β1 (TGF-β1) in peripheral blood of patients with severe preeclampsia.Methods ELISA was used to detect the expressions of HMGB1,IL-6,and TGF-β1 in serum of 57 patients with severe preeclampsia and 21 normal late pregnant women (control group).Results Compared with control group,the expressions of HMGB1,IL-6,and TGF-β1 in severe preeclampsia group increased,there were statistically significant differences (P<0.05).HMGB1 expression in patients with early-onset severe preeclampsia was higher than that in patients with late-onset severe preeclampsia,but there was no statistically significant difference (P>0.05).IL-6 expression in patients with early-onset severe preeclampsia was higher than that in patients with late-onset severe preeclampsia,TGF-β1 expression in patients with early-onset severe preeclampsia was lower than that in patients with late-onset severe preeclampsia,there were statistically significant differences (P<0.05).Correlation analysis showed that HMGB1 was positively correlated with IL-6,while HMGB1 was negatively correlated with TGF-β1.Conclusion HMGB1 upregulation may be involved in the pathogenesis of preeclampsia,assessment of cytokines can effectively monitor severe preeclampsia.
Objective:To comfirm the reliability for the neonatal rats model of periventricular leukomalacia caused by lipopolysaccharide.Methods:Sixteen mature pregnant wistar rats were divided into experimental(n=8)and control groups(n=8).On 15 days of gestation,LPS(0.3 mg/kg)and distilled water were injected intraperitoneally to experimental(n=8)group and control group,respectively.Seventy neonatal rats of experimental group and eighty-two in the control group were obtained till the gestation day of 21.Morphological changes and water content in brain tissues were studied.Results:①Periventricular leukomalacia,band-like and spotty coagulation necrosis were found in brain of neonatal rats in the experimental group,but not in the controls;②Oligodendrocytes,mitochondrial obvious swelling,ridge decreased or vacuolization and endoplasmic reticulum expansion were observed in brain of neonatal rats,in the experimental group,but not in the controls;③Water content in neonatal rats' brain of the experimental group[(89.92±4.06)%]was significantly higher than that in the control group[(87.78±5.21)%](P 0.05).Conclusion:Lipopolysaccharide given to maternal rats could result in periventricular leukomalacia and increase the water content in the brain tissues of neonatal rats.
Objective: To study morphological evolution of fetal lung in SD rats and establish continuous systematic morphological datum.Methods: Fetal lungs at embryonic day16(E16),E19,E21,were taken out respectively,and were observed by HE staining under light microscope and electric microscope.Results: At E16,epithelial tract began to appear,which was constructed by tall epithelium,and developed many airway branches.At E19,alveolar space extended gradually,then lamellar body and type-II pneumonocytes began to emerge.At E21,alveolar separation increased gradually,and mesenchyma turned into thiner than before.At the same time the number of lamellar body increased gradually.Conclusion: The fetal lung development is a gradual process.In fact,this process will continue to postnatal stage.The appearance of lamellar body is the sign of differentiation of type-II pneumonocytes,and it increases along with lung development.
