[目的]探讨miR-145在配对的非小细胞肺癌(non-small cell lung cancer,NSCLC)患者肿瘤组织和癌旁组织中的表达情况,分析其与临床病理因素和预后的关系.[方法]应用实时定量聚合酶链反应(q-RT-PCR)检测miR-145在配对的NSCLC患者肿瘤组织和癌旁组织中的表达水平.[结果] miR-145在NSCLC患者肿瘤组织中的表达水平明显低于癌旁组织(P<0.001).miR-145诊断NSCLC的ROC曲线下面积为0.895.miR-145在Ⅲ~Ⅳ期和中低分化的NSCLC患者肿瘤组织中的表达水平明显低于Ⅰ~Ⅱ期和高分化的肿瘤组织(P<0.05).miR-145低表达组的总生存率明显低于miR-145高表达组(P<0.05).[结论]miR-145在NSCLC组织中的表达下调,在NSCLC的发生中可能发挥抑癌基因的作用,可能是NSCLC的诊断及预后标志物之一.
Objective To observe the protein change and find potential biomarkers of myocardial ischemia in a rat model using surface-enhanced laser desorption /ionization time of flight mass spectrometry(SELDI-TOF MS).Such research may be useful for the diagnosis of acute cardiac ischemia and sudden cardiac death.Methods Sprague-Dawley rats were divided as three groups:operation group,sham operation group,non-operation group.The myocardiums were collected 5min~6h after the coronary artery ligation.Protein extracts were processed by CM10 ProteinChip and analyzed by SELDI-TOF MS.Results Three novel ischemia related peaks with m /z 6304Da,8337Da and 8375Da were only found in operation groups with the 100% positive rate and 100% specificity.Three ischemia related peak height increasing peaks were found:two of them,peak 6658Da and 6876Da increased in ≥15min groups,while peak 8577Da increased in ≥5 min groups.Conclusion Three novel myocardial ischemia related peaks 6304Da、8337Da、8375Da are specific for the myocardium ischemia.They appeared as early as 5 min after the onset of LAD liagation and may serve as the bismarkers of acute myocardial ischemia or ancillary ischemia sign for SCD.
