由于腺相关病毒载体(adeno-associated viral vector,AAV)本身的侵染不具有神经元特异性,其在神经系统相关研究中会存在侵染范围不符合试验要求的情况,因此本试验拟研究不同滴度和注射方式对病毒侵染范围的影响.结果显示,在注射较高滴度的AAV2-CMV-EGFP(1.3×101 mg/L)3周后,其侵染范围可由延髄中缝苍白核(raphe pallidus nucleus,RPa)区域延伸至下丘脑室旁核(paraventricular nucleus,PVN)区域;较低滴度(1.3 × 108 mg/L)结合压力注射的方式可以在一定程度上限制AAV的侵染范围,并且载体携带的EGFP荧光基团在637 nm波段不具有明显的假阳性信号.
为检测目前暂未有报道的小鼠下丘脑内3β羟基类固醇脱氢酶1(3β-HSD1)和细胞色素P450 17A1(CYP17A1)的分布及表达,了解其在下丘脑调节激素分泌中的潜在作用.分别使用免疫荧光及Western Blot技术检测小鼠下丘脑内3β-HSD1和CYP17A1的神经元分布及蛋白表达情况.免疫荧光结果显示,3β-HSD1阳性神经元分布于下丘脑背外侧核、腹内侧核以及弓状核,CYP17A1阳性神经元分布于下丘脑腹内侧核及室旁核;Western Blot结果显示,下丘脑内均有3β-HSD1和CYP17A1蛋白表达.Western Blot试验证实,免疫荧光检测到下丘脑内3β-HSD1和CYP17A1分布的真实性.本试验首次检测出下丘脑内类固醇激素合成关键酶3β-HSD1和CYP17A1的免疫阳性神经信号的分布,为进一步研究下丘脑调节激素分泌提供形态学依据.
兽医药理学作为畜牧兽医专业的专业基础课,是联系基础兽医学与临床兽医学的桥梁和纽带.该课程由理论教学和实验教学两部分组成,实验教学为理论教学服务,是夯实理论知识的一种手段.根据高职高专畜牧兽医专业的教学要求和特点,探索并改进兽医药理学课程教学方法,改变传统的填鸭式教学方法,采用多种手段教学以提高学生的自主学习能力、发现问题和解决问题的能力,以激发学生学习积极性,提高教学质量,从而为临床一线培养合格的应用型专业技术人员打下坚实的基础.
试验选择产蛋率一致的63周龄海兰褐壳蛋鸡480只,随机分为4组,每组5个重复,每个重复24只.试验设计4种日粮,对照组饲喂玉米-豆粕型日粮,处理1组在对照组日粮中添加1 g/kg枯草芽孢杆菌,处理2组在对照组日粮中添加1 g/kg菊粉,处理3组在对照组日粮中添加0.5 g/kg枯草芽孢杆菌+0.5 g/kg菊粉,试验共进行13周,包括1周的预饲期.试验结果:与对照组相比,处理组蛋壳重量显著提高,料蛋比显著降低(P<0.05);对照组蛋壳厚度、密度和平均蛋重最低(P<0.05),破蛋率最高(P<0.05).对照组粪钙排泄量最高(P<0.05),但沉积量最低(P<0.05);处理3组较其他三组显著提高了胫骨密度和胫骨钙含量(P<0.05);处理3组较对照组粪、十二指肠和盲肠内容物pH显著降低(P<0.05).对照组较处理组十二指肠、空肠和回肠绒毛高度和隐窝深度显著降低(P<0.05).对照组较处理组回肠和盲肠内容物大肠杆菌含量显著提高(P<0.05),但显著降低了回肠和盲肠内容物乳酸杆菌和双歧杆菌含量(P<0.05).结论:产蛋后期蛋鸡日粮添加枯草芽孢杆菌对蛋鸡生产性能改善效果最好,而添加菊粉对蛋品质改善效果最好.
为了符合新时代社会对人才实践能力、应用能力等方面的要求,以就业为导向发展高职教育,是高职院校培养实用型、高技能型人才的必然选择.而内涵式发展的主要表现和特点中,强调的就是为企事业单位培养和输出合格的高技能型专业人才,从而实现教育的可持续发展.主要论述了高等职业院校的教育特点及内涵式教育发展的特点,以及以就业为导向发展高职院校的内涵式教育的措施.
In this study, the conservative sequence in L and 2B gene of O type FMDV were chose as possible interference sites. Two siRNAs were designed and synthesized. These siRNAs were cloned into the plasmid pSIREN-Shuttle, then the siR-NAs recombined expression vectors were obtained including pShuttle-L and pShuttle-2B. The vectors and the adenovirus vec-tor pAdeno-X were digested with restriction endonuclease I-Ceu I and PI-Sce I and connected in vitro, then recombinant plasmids were obtained. The result of PCR, enzyme-cutting and sequencing suggested that the recombined adenovirus plas-mids pAdeno-L and pAdeno-2B were constructed successfully.
Porcine contagious pleuropneumonia is a respiratory infectious disease which is harmful to the modem pig industry and widely spread all over China.In this study,one strain of Actinobacillus pleuropneumoniae was isolated and cultured from a hog farm in Baotou inlnner Mongolia.Through treatment trial,it was found that the florfenicol and doxycycline had positive effects on the disease.Meantime,based on the cause analysis of infectious Pleuropneumonia,some effective measures were put forward to control the epidemic.
In this paper the study was aimed at discussing the immunoregulation of taurocholic acid (TCA) by analyzing its effects on mice thymocyte apoptosis. The mice thymocyte cells were isolated by gradient centrifugation and cultured under 37 ℃with TCA of different concentration for 12 hours. Through flow cytometry (FCM),the thymocyte apoptosis in early stage were detected quantitatively. Besides,the activity of caspase-9 and caspase-3 were detected through spectrophotometric methods. The results showed that the TCA group 3 (the final concentration:0.3 μg/mL) could dramatically restrain phosphatidylserine from eversion and restrain the cells apoptosis. Meanwhile,TCA group 2 (the final concentration:0.15 μg/mL) could greatly improve the activity of caspase-9 and caspase-3,and induce the cells apoptosis. It indicated that TCA adjusts organism immune function by regulating the mice thymocyte apoptosis.
