The eukaryotic expression plasmids of full-length and truncated swine TRIM56 gene were constructed, and the expression and localization of fusion proteins were confirmed. According to the sequence and structural characteristics of swine TRIM56 gene, the primers of full-length and four truncations were designed. The total RNA of porcine alveolar macrophages 3D4/21 was extracted. The full-length and truncated genes were amplified by RT-PCR and cloned into pXJ41 eukaryotic expression vector to construct their full-length and truncated eukaryotic expression plasmids. The full-length and truncated eukaryotic expression plasmids of swine TRIM56 were transfected into HEK-293T cells, and the protein expression was detected by Western blotting. The plasmids were also transfected into porcine alveolar macrophages 3D4/21 and the protein localization was detected by IFA. The results of double enzyme digestion and sequencing showed that the full-length and truncated eukaryotic expression plasmids of swine TRIM56 were successfully constructed. Western blotting results showed that the protein of swine TRIM56 was about 82 kDa, and the bands of truncated TRIM56 proteins were consistent with the expected size. IFA results showed that the full-length and four truncated proteins of swine TRIM56 were mainly localized in the cytoplasm. Our research laid a foundation for further study on the antiviral effect and molecular mechanism of swine TRIM56 protein and different domains.
猪流行性腹泻是导致仔猪腹泻的重要传染病,及时准确地诊断对该病的防控具有重要意义.2021年初,湖南娄底某规模化猪场仔猪发病,临床以呕吐、腹泻和消瘦等症状为主,剖检后取肠组织样品送至实验室进行诊断,提取核酸后以PCR/RT-PCR法检测PEDV、TGEV、PORV和PDCOV等病原核酸,仅检出PEDV核酸,未检测到其他病原核酸.通过进一步序列分析,结果表明,该毒株的S1基因与国内流行的变异株同源性最高,提示该猪场流行的毒株为变异株.
子宫蓄脓多为发情后期的一种疾病,是未绝育成年母猫的常见疾病.其特征是子宫内膜异常并继发病原菌感染.文章阐述了 一例母猫发生闭锁型子宫蓄脓的诊断、治疗过程,通过手术治疗该病猫康复.
此试验旨在利用现有神经性犬瘟热病例病料通过鸡胚培养的方法分离提取犬瘟热病毒,并研究出鸡胚培养犬瘟热病毒的最佳条件.同时提取的病毒可以用于后续抗体制备.试验共进行六次鸡胚培养,使用犬瘟热快速检测试纸进行鉴定,成功分离鉴定了 CDV病毒.结果表明:鸡胚培养犬瘟热病毒的方法是可行的,犬瘟热病料研磨液接种鸡胚的较佳接种部位是尿囊腔接种,每个鸡胚接种犬瘟热病料研磨上清液量最佳接种量0.25mL,但鸡胚培养犬瘟热病毒成功率较低,病毒液浓度不高.
兽医外科与手术学是一门应用性极强的学科,掌握该门课程需要学生拥有较高的综合素质.然而传统的教学模式,以灌输式教育为主,教学效果不甚理想.通过课堂教学、实验教学、实践教学、技能大赛中提高及适应社会5个方面对该门学科的教学新方式进行了探究与讨论,旨在提高兽医外科与手术学的教学质量,不断向社会输送高水平人才.
小动物疾病学是培养学生宠物临床实践技能的重要环节,是将兽医学基础理论与宠物临床实际操作相结合的关键课程之一,在兽医专业教学中占有重要地位,在全国农业院校中均受到重视.本文介绍了小动物疾病学课程的现状、课程改革的必要性以及改革与实践,提高了学生对该门课的兴趣和修课率,也提高了毕业生执业兽医资格考试该门课程的得分率和整个考试的通过率,为国家培养新的兽医体制下的合格宠物医疗人才.
为了建立快速检测猪流行性腹泻病-毒(PEDV)的胶体金免疫层析方法(GICA),本研究用PEDV免疫BALB/c小鼠,制备杂交瘤细胞.分别采用间接ELISA和表面荧光吸附法(CSFIA)筛选,共得到33株能够稳定分泌抗PEDV单克隆抗体(iAb)的杂交瘤细胞株.通过制备小鼠腹水并纯化后测得其中21株细胞分泌的MAb的ELISA效价在1∶1.6×104以上,且均仅与PEDV反应,不与TGEV和PRV反应,特异性较强.采用双抗体夹心ELISA法筛选出能够配对检测PEDV的MAb有5株,以其中的4H7为检测抗体、5A9为俘获抗体初步建立了检测PEDV的GICA.该GICA特异性试验结果显示,除了PEDV检测结果为阳性外,该方法对伪狂犬病病毒、猪瘟病毒、猪繁殖与呼吸综合征病毒和传染性胃肠炎病毒的检测结果均为阴性,特异性较强;该方法针对PEDV的最低检测限为7.8× 103 TCID50/mL,敏感性较高;批内和批间重复性检测样品结果无明显差异,具有较好的重复性;将该方法和RT-PCR方法同时检测115份临床样品,该方法与RT-PCR阳性符合率为93%.本实验制备的PEDV MAb,以及基于该AMbs初步建立的检测PEDV的GICA为开发快速检测PEDV的胶体金免疫层析试纸条奠定了基础.
为提高鸡细管精液冷冻质量,采用2%、4%、6%、8%、11%的甘油浓度,离液氮面9种熏蒸高度,两步冷冻法,5℃水浴解冻3 min,离心去甘油等,通过活力与人工输精检测冷冻效果.结果表明:(1)以甘油浓度为主效应,8%~11%各组间差异不显著(P>0.05),但11%组效果最优;以冷冻速率作为主效应,6~8高度处各组间差异不显著(P>0.05),其中第6高度处效果最优;甘油浓度-冷冻速率交互作用下“浓度”、“速率”、“浓度-速率”对活力有极显著的影响(P<0.01);去除甘油前与去除后的精子活力成正相关,去甘油导致活力下降程度达50%左右;(2)以受精率为指标,6%甘油浓度组的最高(77.63%),与4%、8%组差异不显著(P>0.05);11%组显著低于4%、6%、8%组(P<0.05);(3)采用2%甘油浓度组的黑丝羽乌骨鸡冻精,对白莱航鸡和伊萨鸡母鸡进行人工输精,孵化效果无显著性差异(P>0.05),但白莱航鸡略高于伊萨鸡.研究结果提示,采用6%甘油浓度,冷冻装置的第6高度处对鸡精液进行冷冻效果最好.
本试验旨在研究饲粮中添加表皮生长因子(EGF)对断奶仔猪血清生化指标、血清游离氨基酸和小肠黏膜水解氨基酸含量的影响.选取体重相近、健康的21日龄断奶仔猪42头,随机分成3组,每组14个重复,每个重复1头仔猪.Ⅰ组为对照组,饲喂基础饲粮;Ⅱ组和Ⅲ组分别饲喂在基础饲粮中添加200和400μg/kg EGF的试验饲粮.试验期为14 d.结果表明:1)饲粮中添加EGF对断奶仔猪平均日增重、平均日采食量和料重比没有显著影响(P>0.05).2)断奶后7d,Ⅱ组血清尿酸含量显著高于Ⅰ组(P<0.05),而Ⅱ组和Ⅲ组血清免疫球蛋白A含量显著低于Ⅰ组(P<0.05).断奶后14 d,Ⅲ组血清肌酐含量显著低于Ⅰ组(P<0.05),且Ⅲ组血清谷草转氨酶/谷丙转氨酶值显著低于Ⅰ组和Ⅱ组(P<0.05).饲粮中添加EGF对断奶仔猪血清尿素氮、葡萄糖、总蛋白、白蛋白、球蛋白、免疫球蛋白M、免疫球蛋白G、胰岛素样生长因子-1、生长激素含量,白蛋白/球蛋白,谷草转氨酶、谷丙转氨酶活性没有显著影响(P>0.05).3)断奶后7d,Ⅱ组血清牛磺酸、丙氨酸、瓜氨酸和组氨酸含量显著低于Ⅲ组(P<0.05),Ⅱ组血清精氨酸、谷氨酸和丝氨酸含量显著低于Ⅰ组和Ⅲ组(P<0.05),各组之间其他血清氨基酸含量无显著差异(P>0.05).4)断奶后14d,与Ⅰ组和Ⅲ组比,Ⅱ组空肠黏膜缬氨酸和回肠黏膜半胱氨酸含量显著升高(P<0.05),Ⅱ组空肠黏膜丝氨酸、谷氨酸、丙氨酸、半胱氨酸、异亮氨酸、苯丙氨酸和组氨酸含量也有升高趋势(P<0.10).综上所述,饲粮中添加高剂量EGF能够降低断奶仔猪血清免疫球蛋白A等生化指标含量,而饲粮中添加低剂量EGF能够降低血清和小肠黏膜部分氨基酸含量.
The object of this study was to investigate the effects of oral administration with glutamate on organ indices, blood biochemical indexes, blood lipid levels and antioxidant capacities of the suckling piglets. Forty-eight healthy pig-lets (Duroc × Large White × Landrace;1 d of age) with similar initial weight and birth date (1. 55 ± 0. 20 kg;1 d of age) were selected from six sows (8 piglets per sow) and randomly assigned to 4 dietary groups in terms of body weight (BW). Pigltes were marked by ear tags after birth. The piglets were oral administration with 0. 18 g/kg BW of sodium chloride (CN group), 0. 06 g/kg BW monosodium glutamate (LMG group), 0. 50 g/kg BW monosodium glutamate ( MMG group) and 1. 00 g/kg BW monosodium glutamate ( HMG group) , respectively. On day 7 of the trial, one piglet was randomly selected from each group, and the samples were collected after blood collection, and the others were slaughtered on the day 21. The results showed that there were no significant differences on the heart, liver, spleen and kidney index at day 7 and day 21. Oral administration with high dose of glutamate increased serum glutamine synthetase activity and hemoglobin content in suckling piglets on day 7 as well as serum glutamine synthetase activity on day 21. Serum endotoxin ET content and ATPase activity in sucking piglets were significantly reduced by oral administration with high dose of glutamate. Oral administration with medium dose of glutamate reduced the serum free fatty acid content on day 7 and 21, and also affected the serum HDL levels on 21 days. Oral administration with medium dose of glutamate increased serum total antioxidant capacity and reduced the activities of serum glutathione transferase and superoxide dis-mutase and the content of malondialdehyde on day 7. On day 21, the MMG and HMG groups had lower serum glutathi-one transferase activity and malondialdehyde content and greater serum superoxide dismutase activity than CN group. The results of the present experiment indicate that oral administration with glutamate can affect blood biochemical indices, blood lipid levels and improve the antioxidant capacities of suckling piglets.
为探讨异源脂质体包裹血红蛋白(Liposome encapsulated hemoglobin,LEH)复苏急性失血性休克猫的临床效果,利用脂质体包裹猪血红蛋白制作异源LEH,将6只猫随机分为3组,分别为自身红细胞(RBC)组,LEH组和生理盐水组,建立猫急性失血性休克模型,并维持休克30 min后输入2倍失血体积的乳酸林格液恢复渗透压.分别用自身RBC、LEH、生理盐水再进行复苏,观察比较3组治疗前后的参数指标.LEH组相比于生理盐水组,血氧饱和度、血红蛋白含量有明显上升;生化指标谷草转氨酶(AST)、谷丙转氨酶(ALT)、血肌酐(CREA)、尿素(UREA)中所表达的损伤更低,接近于RBC组的复苏效果.试验结果表明:复苏3h后,异源LEH可以修正急性失血性休克猫的氧亏和减少低灌注引起的多器官损伤.

The experiments reported in this research communication aimed to plot the expression pattern of Sirt3, a master regulator of energy metabolism and antioxidation defence, in the liver of dairy goats during perinatal period. Ten healthy dairy goats in late pregnancy were chosen, and needle biopsy was applied to collect liver samples at 1-week intervals. Protein levels of hepatic Sirt3 were analysed by western-blotting. Serum enzyme activities of manganese superoxide dismutase (Mn-SOD) and non-esterified fatty acids (NEFA) levels were measured, and their correlation with Sirt3 mRNA levels was also estimated. Compared with >3-week before parturition (BP), Sirt3 proteins were significantly reduced at 1-week after parturition (AP) and 2-week AP (P < 0·05), but increased on the day of parturition (P < 0·01). Correlation analysis revealed a positive association between hepatic Sirt3 mRNA levels and serum enzyme activity of Mn-SOD (r = 0·46), but a negative association between that and serum NEFA levels (r = −0·41). These data indicate that the decreased hepatic expression of Sirt3 might be one of the reasons that dairy goats undergo oxidative stress after parturition.
2010—2016年,笔者对治疗过的具有代表性的有一定操作意义的17例中大型野生动物的外伤进行了治疗,取得了较好的治疗效果。并在野生动物治疗过程中对治疗方法、保定方式、用药方法及动物习性方面进行汇总分析,为大中型野生动物的外伤治疗提供治疗参考。
脂质体包裹血红蛋白(LEH)有望克服人类和动物的用血障碍.文中以国内外研究LEH的成果为基础,对LEH的发展历史、创新改进、临床运用进行了综述,为脂质体包裹血红蛋白的发展提供参考.
围产期是反刍动物普通病及传染病高发时期,其最显著的病理特征是能量负平衡.肝脏是能量代谢最重要的器官,肝脏样品的采集在围产期疾病诊断及发病机制研究方面尤为重要.用围产期山羊作为试验动物,采用穿刺活检技术进行肝脏样本采集.标准穿刺活检操作对9头围产期山羊进行70次穿刺取样,结果显示,肝脏采出率为100%,死亡率为0,流产率为0;血常规检查及肝功能检测显示血液学指标及转氨酶无明显异常.说明此法安全可靠,可用于山羊肝脏的活检取样.
《兽医外科学》是兽医专业本科教育的必修课程之一,具有较强的实践性和应用性.《兽医外科学》在传统教育模式下,无法向社会输送小动物临床专业型人才.因此,在《兽医外科学》的教育过程中,应注重小动物医学专业人才的培养,提高学生的学习兴趣,增强学生的实践能力,为兽医行业输送合格的毕业生.
The aim was to explore the effects of different kinds of dilution and thawing devices on the N,N-dimethylformamide (DMA) pellet frozen semen of Black Silkies.Firstly,the motility and fertility of the frozen semen thawed by different dilutions were compared;Then,the motility of the frozen semen was compared when the pellets were thawed using different tube and different number.Finally,the motility and fertility of the sperm thawed by three kinds of thawing devices (thermostat water bath,hotfunnel and hotplate) were tested.The results showed that:①There was similar order of the sperm motility and fertility in the different dilution groups (LR>F>B> L),and there was significant difference among those groups (P<0.05).②The motility was the best when the frozen semen was thawed with large thin-wall glass tube at 60 ℃.③The best temperature range of the 3 devices was different.The highest motility for thermostat water bath was 50 to 60 ℃ (0.51 to 0.59),and thermostat hotfunnel was 40 to 45 ℃ (0.42 to 0.46),while thermostat hotplate was 50 to 55 ℃ (0.61 to 0.63).There was no significant difference of the motility in the optimum temperature range for each device (P>0.05).④The fertility of the different devices in their best thawing temperature was 26.91% (55 ℃,thermostat hotplate),23.08% (60 ℃,thermostat water bath),20.93% (40 ℃,thermostat funnel),respectively,and there was no significant differences among those groups (P>0.05).Therefore,the efficiency thawing condition for the Black Silkies frozen semen was the LR diluent,DMA cryoprotectant,pellet freezing,thawed in the thermostat hotplate at 54.9 ℃.
T-2可引起细胞发生毒性作用,抑制DNA、RNA及蛋白质合成,还可造成细胞活性下降,细胞内LDH、ALT外漏.T-2可诱导DNA损伤,进而导致DNA碎片化,而DNA碎片化是检测细胞凋亡的指标之一.对于细胞凋亡的机制,尚未研究透彻,但已发现与细胞凋亡相关的蛋白有Caspases家族、Bcl-2家族、p53蛋白等.这些蛋白分别在细胞凋亡的不同时期发挥重要作用.目前,未见相关文献报道T-2/HT-2诱导鸡肝细胞损伤、氧化应激及细胞凋亡.
The PAM isolated from healthy piglets,were incubated as the proportion of 1 ∶ 10,1 ∶ 3 and 1 ∶ 1 with Mycoplasma hyopneumoniae (Mhp) strain 232.After incubation 12 h,PAM was harvested and detected by qPCR methods to determine the relative transcription of MHC Ⅱ mR-NA,and the optimal incubation ratio (ROI) of MHC Ⅱ and Mhp.PAM and Mhp were incubated by the ROI,and the 10% RPMI-1640 solution,instead of Mhp,was incubated with PAM as the control group.PAM were harvested at 12 h post incubation (12 hpi),24,36 and 48 hpi,respectively.qPCRs were used to detect mRNA of the exogenous antigen processing and presenting moleculars of PAM such as:MHC II,SLA-DM,Li and CD86,toll like receptor 2 (TLR2) and TLR6.The mRNA expressions of MHC II,SLA-DM,Li,CD86,and TLR6 genes were significantly increased (17-380 times) at 12 hpi,was lower 0.043-0.391 times then the control groups at 24 hpi,greatly reduced at 36 hpi and were not detected at 48 hpi.It showed that the early Mhp infection (12 h) can cause to increase the mRNA expression of the exogenous antigen processing and presenting moleculars of PAM and in the later stage of Mhp infection (24 h),the mRNA expression were inhibited,indicating that Mhp infection in the early stage can lead to enhance imunity level of the animal body through PAM,and in the later stage Mhp infection can cause immune suppression.
