IntroductionArchaeal communities play a crucial role in marine ecosystems, yet our understanding of their ecological and functional traits remains incomplete. This study focuses on northern Hangzhou Bay to fill gaps in knowledge regarding the biogeography and functionality of archaeal groups.MethodsWe utilized a high-throughput sequencing dataset based on the 16S rRNA gene to characterize the archaeal community, aiming to identify biogeographic patterns and assess the influence of environmental factors on community structure.ResultsThe predominant phyla identified were Woesearchaeota, Thaumarchaeota, Euryarchaeota, and Crenarchaeota. Archaeal community structure in sediments showed a geographical pattern along the environmental gradient, influenced by factors such as salinity, ammonium, total phosphorus, pH, and total nitrogen content. Network analysis revealed nonrandom co-occurrence patterns, with associations changing along the salinity gradient. Additionally, this study directly proved the existence of dispersal limitation in this strongly connected marine ecological system through null model analyses. Variation in the archaeal community was attributed to both environmental constraints and stochastic processes due to dispersal limitation. Furthermore, our results revealed that the key biogeochemical functions of the archaeal community also exhibited a clear salinity gradient, the functional differences appear to be influenced by salinity, and the critical roles of archaeal diversity were highlighted.DiscussionAll these findings enhance our understanding of microbial ecology and element transformation in estuarine environments. The highlighted roles of archaeal diversity and the influence of environmental factors on community structure and function underscore the complexity of marine microbial ecosystems.
细胞生物学作为生物学专业的核心课程,在生命科学知识体系的建构中具有重要作用.本课程组基于细胞生物学课程特点,构建了基于SPOC平台的混合式教学模式,开展了线上线下混合式教学的改革.文章从细胞生物学混合式教学模式的构建,实施和评价等方面对混合式智慧教学模式进行初步的探究.
The use of photocatalysis to address environmental pollution and energy shortage is an attractive choice. Herein, we successfully synthesized a novel 3D interconnected porous carbon-rich g-C3N4 catalyst via facile thermal polymerization to enhance photocatalytic hydrogen production and photodegradation of dye contaminants. Enhanced hydrogen evolution (1956.23 μmol g−1 h−1) and photocatalytic RhB degradation (96.74%) efficiency were achieved with the as-obtained catalysts. Based on the photocatalytic experimental data and characterization analyses, an enhancement mechanism was proposed. The 3D interconnected porous structure endowed the g-C3N4 with numerous active sites and a large specific surface area, and the carbon modification facilitated the separation and transfer of the photoinduced charge carriers. Nanoshape engineering and the carbon-rich structure showed a synergetic effect in increasing photocatalytic performance. This study offers an applicable methodology for the exploitation of an economical catalyst to alleviate environmental pollution and energy shortages.
为探讨人脐带间充质干细胞源外泌体对黑色素瘤细胞生长的影响,利用差速超速离心法提取人脐带间充质干细胞源外泌体,并用透射电子显微镜、Western blot方法鉴定外泌体形态、粒径和蛋白质;利用CCK-8法、细胞划痕试验、Transwell试验检测不同浓度人脐带间充质干细胞源外泌体对黑色素瘤细胞的增殖、迁移和侵袭能力的影响.结果表明:人脐带间充质干细胞源外泌体呈杯托状,平均粒径70 nm左右,表达标志性蛋白CD63和CD9.不同浓度人脐带间充质干细胞源外泌体对黑色素瘤细胞增殖、迁移和侵袭能力有促进作用,且呈时间、剂量依赖性,并在人脐带间充质干细胞源外泌体浓度为300μg·mL-1时达到最高,而在浓度为400 pg·mL-1时促进率开始明显下降.
在全课程育人背景下,高校“课程思政”建设是一个不断发展与完善的过程,旨在对学生进行知识传授和综合能力培养的同时,实现对学生的德行培养和价值塑造。然而,在实际教育教学过程中,很多思政要素尚没有充分挖掘。本文对高校“课程思政”建设的重点与难点进行了分析,以期为我国高校“课程思政”建设提供参考。
目的:探究金银花叶总黄酮的最佳提取方法及其抗氧化能力.方法:分别采用乙醇回流提取法、索氏提取法和超声辅助浸提法提取金银花叶总黄酮,并用ABTS自由基清除率评估其抗氧化活性.结果:不同提取方法获得供试品中总黄酮含量有差异,其中回流提取法所得总黄酮含量最高;金银花叶总黄酮清除ABTS自由基的能力明显强于维生素C.结论:金银花叶总黄酮具有较强的抗氧化活性.
为探究怀菊不同部位内生细菌群落多样性及分布情况,本文应用Illumina Miseq高通量测序技术,分析怀菊茎、叶、花内生细菌高通量测序质控后有效序列数目有差异,分别聚成54、54和51个OTUs.菊花不同部位多样性指数中的Shannon指数花>茎>叶、Simpson指数叶>茎>花,Streptophyta属为内生细菌的优势菌属.证明怀菊不同部位内生细菌的菌群组成及分布存在一定差异.
目的:比较红螺红茶、碧螺春绿茶和紫芽普洱茶(生茶)3种常见茶叶提取物对黑色素瘤细胞增殖的影响及对大肠杆菌、金黄色葡萄球菌的抑菌效果.方法:3种茶叶均采用超纯水煮沸浸提法,过滤获得茶叶浸提物溶液.CCK-8法测茶叶提取物对细胞增殖的影响;通过药敏片扩散法研究不同茶叶浸提物对大肠杆菌、金黄色葡萄球菌的抑菌效果.结果:3种茶叶提取物对黑色素瘤细胞的生长都有一定的抑制效果,紫芽普洱茶(生茶)>碧螺春绿茶>红螺红茶.大肠杆菌对红螺红茶、碧螺春绿茶为中敏,对紫芽普洱茶生茶(生茶)高敏;金黄色葡萄球菌对红螺红茶高敏,对碧螺春绿茶、紫芽普洱茶生茶(生茶)极敏.结论:3种茶叶对黑色素瘤细胞生长都有一定的抑制效果;3种茶叶均有抑菌效果,紫芽普洱茶(生茶)>碧螺春绿茶>红螺红茶,同一种茶叶提取物对金黄色葡萄球菌的抑菌效果要优于大肠杆菌.
对新乡医学院三全学院生命科学技术学院三个专业五门核心课程的原有实验项目进行分析,找出缺陷和不足,在此基础上对实验项目进行重新设计,并在实际运用中取得了良好的实践效果.
