目的:探讨核转运蛋白基因(karyopherin,KPNA)2在皮肤恶性黑素瘤临床预后中的作用.方法:应用GEO数据库数据分析,比较KPNA2 mRNA在正常皮肤组织、恶性黑素瘤中表达的差异,并进一步利用两种在线数据库进行生存分析,分析KPNA2 mRNA与皮肤恶性黑素瘤的预后之间的关系.结果:KPNA2 mRNA水平在正常皮损组织中明显要低于恶性黑素瘤组织,差异有统计学意义(P<0.05);同时生存分析显示KPNA2 mRNA表达水平与皮肤恶性黑素瘤的预后具有的显著相关性,其表达水平高则预后差(P<0.05).结论:KPNA2与皮肤恶性黑素瘤临床预后密切相关,为皮肤恶性黑素瘤治疗提供潜在靶点.
The definition of oocytes in vitro maturation (IVM) is that the maturation in vitro of immature cumulus-oocyte complexs (COCs) collected from antral follicles. IVM was firstly used in human in 1991, but it developed very slowly. Now researchers pay more and more attention to IVM. C-type natriuretic peptide (CNP) can sustain the arrest of oocytes at germinal vesicle (GV) stage. According to this, biphasic IVM system based on CNP is established, and it works effectively in multi-species. Recently, it was proved that this kind of IVM system can improve human oocytes maturation rate and the developmental competence of mature oocytes. This article summarizes the action principle of CNP and the research advances of biphasic IVM system. We wished to investigate the effect of silencing of the human papillomavirus 18 (HPV18) E7 gene on the apoptosis and proliferation of a human cervical cancer cell (HeLa) line by small interfering RNAs (siRNAs).Two HPV18-E7 gene-specific siRNAs were designed and transfected into HeLa cells,and denoted "siE7-1 group" and "siE7-2 group".Non-transfected HeLa cells were used as the "normal control" group (NC).Non-functional scramble siRNA was transfected using the same method and named as the "SCR group".Reverse transcription-polymerase chain reaction and western blotting were used to detect expression of the E7 gene at mRNA and protein levels.Flow cytometry was used to detect apoptosis after silencing of the E7 gene.A colony-formation assay and CCK-8 assay were used to monitor cellular proliferation.Expression at mRNA and protein levels of the E7 gene in siE7-1 and siE7-2 groups decreased significantly compared with that of the NC group (P<0.01).Percent apoptosis in siE7-1 and siE7-2 groups was 17.78% and 36.44% at 48 h after transfection,which was significantly different compared with that of the NC group (P<0.01),thereby implying apoptosis induction after silencing of the E7 gene.Results of the CCK-8 assay and colony formation assay were consistent with each other,and showed proliferation suppression in siE7-1 and siE7-2 groups compared with the NC group (P<0.01).These data suggest that siRNA interference of the E7 gene can induce apoptosis and inhibit the proliferation of HeLa cells.Therefore,the E7 gene could be a potential target site in the treatment of cervical cancer.
Objective:To explore the risk factors associated with ectopic pregnancy after Assisted Reproduction Technologies (ART).Methods:We retrospectively evaluated all the cycles underwent IVF /ICSI from January 2001 to December 2008.One hundred and forty-six of 3005 pregnancies (4.89%) were ectopic pregnancy,following 7798 IVF /ICSI embryo transfers.Multivariable logistic regression was employed for multi-factor analyses.Results:The infertility associated with tubal factor such as hydrosalpinx,past medical history of controlled ovarian hyperstimulation (COH),the thickness of endometrium before hCG injection,the quality of transplanted embryos was correlated with ectopic pregnancy.Conclusion:The infertility associated with tubal factor is the main correlative factor with EP in IVF /ICSI programs;hydrosalpinx,past medical history of COH can increase the risk of ectopic pregnancy;on the other hand,increase the endometrium's thickness and the quality of transplanted embryo will decrease the risk.
Objective:To investigate the effects of different kinds of laparoscopic managements of hydrosalpinx on ovarian reserve and analyze the mechanisms.Methods:A total of 111 hydrosalpinx patients were treated by laparoscopy.This was a prospective study,including 48 cases with pure salpingostomy(group A),33 cases with proximal tubal division and distal salpingostomy(group B) and 30 cases with salpingectomy(group C).Other 20 non-hydrosalpinx women undergoing laparoscopic with unexplained factor infertility or uterine disease were defined as control group(group D).Results:Three months after operation,the number of small antral follicles(2-6 mm) in groups A,B and C was more than preoperation(P<0.05);RI of ovarian artery was lower than preoperation in group B(P<0.05).There was no significant difference between preoperation and postoperation in group D.Postoperative RI of groups A,B and C was significantly different compared with each other(P<0.05).Conclusion:Hydrosalpinx can impair ovarian reserve function,three kinds of clinical laparoscopic managements mentioned above can improve ovarian reserve.The proximal tubal division and distal salpingostomy of hydrosalpinx may be the most conducive to improve the function of ovarian reserves.
