A rare strain of Toxoplasma gondii typeⅢ has been isolated from a rabbit of Shanghai in our laboratory,and named it as SHR.Due to the specificity of this strain in china,we analysed the therapeutic effect of azithromycin,sulfachloropyrazine sodium,acetylspiramycin and paromomycin against the infection of SHR strain.Mice were inoculated intraperitoneally with 1×104tachyzoites of SHR strain.Drugs were administrated three hours post-infection,once per day for 5consecutive days,and the first dose was reduplicated.The survival time and survival rate of mice were observed for continued 20 days.The results indicated that the azithromycin showed the best therapeutic effect,acetylspiramycin came the second.The Sulfaclozine sodium can only delay the survival time,while paromomycin showed no therapeutic effect.The results revealed that azithromycin has the best treatment for the mice infected with SHR.
A double antibody sandwich ELISA for detection of Trypanosomaevansivariant surface glucoprotein(VSG)antigen was established by using two monoclonal antibodies against Trypanosoma evansi VSG antigen.The result of sensitivity showed when positive serum was diluted to 3200 doubles,its D450nm value was still greater than the critical value of positive and negative,and specificity test result showed that there was no cross reaction with the antigens of Theileria,Toxoplasma gondii,Chlamydiaand Fasciola gigantica.82 clinical sera were detected by this double antibody sandwich ELISA and its positive rate was 9.76%.The experiment results demonstrated that this double antibody sandwich ELISA for detection of Trypanosoma evansiVSG antigen had good detecting sensitivity and specificity,and could be used as an effective method for clinical detecting buffalo Trypanosoma evansi disease.
本试验旨在培育大片形吸虫囊蚴,着重研究大片形吸虫在中间宿主体内的发育过程和临床病理变化.人工培养和孵化大片形吸虫虫卵,用孵化出来的大片形吸虫毛蚴感染中间宿主小土蜗螺,收集大片形吸虫囊蚴,再用囊蚴感染试验小鼠.结果显示,囊蚴经口感染试验小鼠后1周即可在肝脏找到虫体,幼虫可在小鼠体内发育生存7~8周,随着时间的推移和感染囊蚴数量不同,可给小鼠肝脏造成不同程度的病变,甚至造成死亡.剖检小鼠可见肝脏质地脆,颜色发黄,脾脏肿大等严重病理变化.因此,可利用小鼠作为大片形吸虫幼虫感染的试验动物,进行片形吸虫病的早期诊断、免疫预防和治疗等方面的研究.
