Parkinson's disease (PD) is one of the key neurodegenerative disorders caused by a dopamine deficiency in the striatum due to the death of dopaminergic (DA) neurons of the substantia nigra pars compacta. The initially discovered A53T mutation in the alpha-synuclein gene was linked to the formation of cytotoxic aggregates: Lewy bodies in the DA neurons of PD patients. Further research has contributed to the discovery of beta- and gamma-synucleins, which presumably compensate for the functional loss of either member of the synuclein family. Here, we review research from 1-methyl-4-phenyl-1,2,3,6-tetrahydropyridine (MPTP) toxicity models and various synuclein-knockout animals. We conclude that the differences in the sensitivity of the synuclein-knockout animals compared with the MPTP neurotoxin are due to the ontogenetic selection of early neurons followed by a compensatory effect of beta-synuclein, which optimizes dopamine capture in the synapses. Triple-knockout synuclein studies have confirmed the higher sensitivity of DA neurons to the toxic effects of MPTP. Nonetheless, beta-synuclein could modulate the alpha-synuclein function, preventing its aggregation and loss of function. Overall, the use of knockout animals has helped to solve the riddle of synuclein functions, and these proteins could be promising molecular targets for the development of therapies that are aimed at optimizing the synaptic function of dopaminergic neurons.
Целью данной работы являлась оценка выживаемости популяции зрелых дофаминергических (ДА-ергических) нейронов чёрной субстанции двух альфа-синуклеин нокаутных линий мышей Abel-KO и ΔFlox-KO, а также бессинуклеиновых животных abg-КО в условиях МФТП-токсического моделирования паркинсонического синдрома. Методы исследования: Водный раствор нейротоксина МФТП вводили 3-месячным мышам внутрибрюшинно в дозе 30 мг/кг ежедневно в течение 5 дней по субхроническому протоколу. Через 21 день после последней инъекции МФТП у животных извлекали головной мозг, фиксировали в холодном растворе Карнуа и парафинизировали для последующего приготовления гистологических препаратов на ротационном микротоме Leica RM2265 (Leica Biosystems, Германия). Иммуногистохимическое окрашивание проводили антителами против тирозингидроксилазы (моноклональные антитела мыши, Sigma, разведение 1:2000). Сравнительный морфометрический анализ популяции ДА-ергических нейронов чёрной субстанции выполнен с учётом поправки Аберкромби. Результаты: Установлено, что в условиях дефицита альфа-синуклеина мыши устойчивы к потере ДА-ергических нейронов в компактной части ЧС после введения МФТП. При генетической делеции всех трёх синуклеинов чувствительность ДА-ергических нейронов ЧС к токсическому действию МФТП не отличается от таковой у животных с немодифицированным геномом. Заключение. На основании проведённого морфометрического анализа предполагается, что особенности чувствительности к нейротоксину МФТП у альфа-синуклеин нокаутных линий мышей обусловлены повышением функциональной активности (замещением) бета-синуклеина, оптимизирующего захват ДА синаптическими везикулами. The aim of this study was to assess survival of mature dopaminergic (DAergic) neuronal population in the substantia nigra pars compacta (SNpc) of two alpha-synuclein knockout mice strains (Abel-KO and ΔFlox-KO) and of non-synuclein animals (abg-KO) in MPTP-induced parkinsonism. Material and methods: MPTP water solution was administered to 3-month-old mice intraperitoneally (30 mg/kg daily for 5 days) according to a subchronic protocol. On the 21st day after the last MPTP injection, the brain was excised, fixed in cold Carnoy’s solution and paraffined for the subsequent preparation of histological samples on a Leica RM2265 rotary microtome (Leica Biosystems, Germany). Immunohistochemical staining was performed with antibodies against tyrosine hydroxylase (mouse monoclonal antibodies, Sigma, dilution 1:2000). A comparative morphometric analysis of substantia nigra dopaminergic neurons was performed using the Abercrombie correction. Results: MPTP-treated alpha-synuclein deficient mice were resistant to the loss of DAergic neurons in the SNpc. Genetic deletion of all three synucleins restored the sensitivity of SNpc DAergic neurons to the MPTP toxicity, which did not differ from the sensitivity of wild type animals. Conclusion: Based on the morphometric analysis, it was assumed that the specific features of MPTP sensitivity in alpha-synuclein knockout mice are due to an increased functional activity (substitution) of beta-synuclein, which optimizes the capture of DA by synaptic vesicles.

Данный метод особенно актуален в отношении использования моделей in vivo для понимания патофизиологических механизмов расстройств центральной нервной системы, включая эпилепсию, рассеянный склероз и нейродегенеративные заболевания. Одним из ключевых среди нейродегенеративных расстройств является болезнь Паркинсона (БП) - хроническое прогрессирующее заболевание, вызванное недостатком дофамина в стриатуме головного мозга в результате гибели дофаминергических (ДА) нейронов чёрной субстанции (ЧС). В настоящее время для воспроизведения у животных основных характеристик БП, включая двигательные нарушения, прогрессирующую потерю ДА-ергических нейронов в компактной части черной субстанции и образование телец Леви, широко используют соединение 1-метил-4-фенил-1,2,3,6-тетрагидропиридин (МФТП). В этом обзоре будут охарактеризованы популярные экспериментальные модели для изучения БП, в которых в рамках исследований вводили нейротоксин МФТП, с целью систематизировать уже имеющийся научный опыт, а также способствовать развитию моделирования паркинсонического синдрома. The use of experimental animal models to understand human diseases and the potential treatments has been extremely valuable and has led to many significant advances in medicine. In contrast to isolated in vitro models, the use of animals as a model system has proven to be more productive for studying of multi-factorial diseases or diseases that affect several human systems. This method is particularly relevant for the use of animal models to understand the pathophysiology of central nervous system disorders such as meningitis, multiple sclerosis, and neurodegenerative disorders. Parkinson’s disease (PD), a chronic progressive illness, caused by a lack of dopamine (DA) in the striatum due to the death of DAergic neurons in the substantia nigra (SN), is one of the neurodegenerative key disorders. Nowadays, 1-methyl-4-phenyl-1,2,3,6-tetrahydropyridine (MPTP) is a widely used compound to reproduce the main characteristics of PD in animals, which include motor disorders, progressive loss of DA-neurons in the substantia nigra pars compacta and the formation of Lewy bodies. In this review, we observe MPTP-treated popular mouse models for the PD studying in order to systematize the existing scientific experience, as well as to promote the development of parkinsonian syndrome modeling.
Целью данных исследований явилось изучение влияния гимантана (N-(2-адамантил)-гексаметиленимина гидрохлорида) на уровень провоспалительных цитокинов IL-1β, IL-6, интерферона-γ (ИФН-γ) и фактора некроза опухоли-α (ФНО-α) в нигрокаудатном комплексе мышей на ранней и поздней клинической фазе экспериментального паркинсонического синдрома (ПС), для выяснения его антипаркинсонического эффекта. Методы исследования: Раннюю и позднюю клиническую фазу ПС создавали у мышей линии C57BL/6J внутрибрюшинным введением пронейротоксина 1-метил-4-фенил-1,2,3,6-тетрагидропиридина (МФТП) в дозах 12 мг/кг или 20 мг/кг по 4 инъекции с интервалом 2 часа, соответственно. Гимантан вводили мышам внутрибрюшинным в дозе 20 мг/кг, предварительно каждый раз за 30 мин до введения МФТП. Содержание цитокинов в структурах мозга мышей определяли методом иммуноферментного анализа с использованием тест-систем производства «Cloud-Clone Corporation», США и считывающего устройства «ИФА-reader» прибора «ImmunoChem-2100», США. Результаты: Показано, что уровень IL-1β, IL-6, ИФН-γ и ФНО-α в нигрокаудатном комплексе мозга мышей возрастает как на ранней, так и на поздней фазах развития ПС. Предварительное применение гимантана снижало в нигрокаудатном комплексе мышей содержание цитокинов IL-1β, ИФН-γ и ФНО-α на ранней фазе, и только одного из 4 изученных (IL-6) - на поздней фазе развития ПС. Предполагается, что антипаркинсонический эффект гимантана на ранней клинической стадии МФТП-индуцированного ПС осуществляется, в том числе, за счёт снижения уровня провоспалительных цитокинов в нигростриатной системе, предупреждая снижение жизнеспособности дофаминергических нейронов. The aim of this work was to study the effect of Hemantane (N-(2-adamantyl)-hexamethyleneimine hydrochloride) on the level of pro-inflammatory cytokines IL-1β, IL-6, interferon-γ (IFN-γ), and tumor necrosis factor-α (TNF-α) in the nigrocaudate complex of mice in early and late clinical phases of experimental Parkinsonian syndrome (PS) to elucidate its antiparkinsonian effect. Material and methods: The early and late clinical phases of PS were created in C57BL / 6J mice by 4 intraperitoneal injections at 2-h intervals of a proneurotoxin, 1-methyl-4-phenyl-1,2,3,6-tetrahydropyridine (MPTP), at a dose of 12 mg/kg or 20 mg/kg. Hemantane was injected intraperitoneally at a dose of 20 mg/kg 30 min before each MPTP administration. Concentrations of cytokines in mouse brain structures were measured by enzyme-linked immunosorbent assay (ELISA) using Cloud-Clone Corporation (USA) test systems and an ImmunoChem-2100 (USA) ELISA reader. Results: Concentrations of IL-1β, IL-6, IFN-γ, and TNF-α in the nigrocaudate complex were increased both in the early and late phases of PS. Prior administration of hemantane reduced the content of IL-1β, IFN-γ, and TNF-α in the nigrocaudate complex at the early phase and the content of only one of the 4 studied cytokines (IL-6) at the late phase of PS. It was assumed that the antiparkinsonian effect of hemantane at the early clinical stage of MPTP-induced PS involves a decrease in proinflammatory cytokines in the nigrostriatal system, which prevents the impairment of the viability of dopaminergic neurons.
Aggregated forms of α-synuclein are core components of pathohistological inclusions known as Lewy bodies in substantia nigra (SN) neurons of patients with Parkinson’s disease (PD). The role of α-synuclein in selective loss of SN dopaminergic neurons (DNs) in PD is studied in mice knocked out in the α-synuclein gene. The new mouse strain delta flox KO with a constitutive knockout of the α-synuclein gene models the end point of in vivo deletion of the α-synuclein gene in mice with a conditional knockout and has no foreign sequence in the modified genomic locus, thus differing from all other α-synuclein knockout mouse strains. The effect of the neurotoxin 1-methyl-4-phenyl-1,2,3,6-tetrahydropyridine (MPTP), which is used to model PD, was compared between delta flox KO mice and mice of the well-known α-synuclein knockout strain AbKO. Subchronic MPTP administration, which models early PD, was found to reduce the dopamine content and to change the ratio of dopamine metabolites in the striatum to the same levels in delta flox KO, АbKO, and wild-type mice. Overt locomotor defects were not observed after MPTP treatment, but gait testing in a CatWalk XT (Noldus) system revealed identical gait deviations in mice of the two strains and control wild-type mice. Based on the findings, a similar mechanism of neurotoxic damage to DNs was assumed for delta flox KO and AbKO mice.
Previous studies of the alpha-synuclein null mutant mice on the C57Bl6 genetic background have revealed reduced number of dopaminergic neurons in their substantia nigra pars compacta (SNpc). However, the presence in genomes of the studied mouse lines of additional genetic modifications that affect expression of genes located in a close proximity to the alpha-synuclein-encoding Snca gene makes these data open to various interpretations. To unambiguously demonstrate that the absence of alpha-synuclein is the primary cause of the observed deficit of dopaminergic neurons, we employed a recently produced constituent alpha-synuclein knockout mouse line B6(Cg)-Sncatm1.2Vlb/J. The only modification introduced to the genome of these mice is a substitution of the first coding exon and adjusted short intronic fragments of the Snca gene by a single loxP site. We compared the number of dopaminergic neurons in the SNpc of this line, previously studied B6(Cg)-Sncatm1Rosl/J line and wild type littermate mice. A similar decrease was observed in both knockout lines when compared with wild type mice. In a recently published study we revealed no loss of dopaminergic neurons following conditional inactivation of the Snca gene in neurons of adult mice. Taken together, these results strongly suggest that alpha-synuclein is required for efficient survival or maturation of dopaminergic neurons in the developing SNpc but is dispensable for survival of mature SNpc dopaminergic neurons.
In the present study, we analyzed the uptake of radiolabeled dopamine by intact synaptosomes and purified synaptic vesicles isolated from the dorsal striatum of mice with constitutive inactivation of all three synuclein-coding genes and wild-type mice. Synuclein deficiency substantially compromised the uptake of this neurotransmitter by synaptic vesicles but had no effect on synaptosomal dopamine uptake.
Провели сравнительный анализ захвата радиоактивно меченого дофамина интактными синаптосомами и очищенными синаптическими везикулами, выделенными из дорзального стриатума мышеи с полной инактивацией генов, кодирующих все три синуклеина, и мышеи дикого типа. Отсутствие синуклеинов привело к существенному снижению эффективности захвата этого нейромедиатора синаптическими везикулами, но не повлияло на захват дофамина синаптосомами.
Lesion of the dopaminergic neurons of the nigrostriatal system is a key feature of Parkinson's disease (PD). Alpha-synuclein is a protein that is a major component of Lewy bodies, histopathological hallmarks of PD, and is involved in regulation of dopamine (DA) neurotransmission. Previous studies of knockout mice have shown that inactivation of alpha-synuclein gene can lead to the reduction in number of DA neurons in the substantia nigra (SN). DA neurons of the SN are known to be the most affected in PD patients whereas DA neurons of neighboring ventral tegmental area (VTA) are much less susceptible to degeneration. Here we have studied the dynamics of changes in TH-positive cell numbers in the SN and VTA during a critical period of their embryonic development in alpha-synuclein knockout mice. This precise study of DA neurons during development of the SN revealed that not only is the number of DA neurons reduced by the end of the period of ontogenic selection, but that the way these neurons are formed is altered in alpha-synuclein knockout mice. At the same time, DA neurons in the VTA are not affected. Alpha-synuclein exerts a modulating effect on the formation of DA neurons in the SN and has no effect on the formation of DA neurons in VTA, the structure that is much less susceptible to degeneration in PD brain, suggesting a potential role of alpha-synuclein in the development of the population of DA neurons in substantia nigra.
