目的:探讨HIF-1α通过调节TP53INP1蛋白对乳腺癌细胞迁移及侵袭能力的影响.方法:通过免疫组化检测60例乳腺癌标本中TP53INP1和HIF-1α的表达情况及分析其临床病理资料.通过体外构建HIF-1α及共转染HIF-1α和TP53INP1细胞模型,迁移侵袭实验和划痕实验检测HIF-1α和TP53INP1对乳腺癌细胞迁移侵袭能力的影响;Western blot实验研究共同下调HIF-1α和TP53INP1对MMP2、VEGF蛋白表达水平的影响.结果:免疫组化实验显示HIF-1α在人乳腺癌组织中高表达,TP53INP1呈低表达,两者之间具有相关性且均与淋巴结转移有关(P<0.05).迁移侵袭实验及划痕实验显示在乳腺癌MDA-MB-231细胞和MCF-7细胞中下调HIF-1α抑制了迁移侵袭及愈合能力,MCF-7细胞中共同转染下调HIF-1α和TP53INP1质粒之后逆转了乳腺癌细胞的迁移侵袭能力及愈合能力(P<0.05).Western blot实验显示shHIF-1α和TP53INP1共转染后在MDA-MB-231细胞中MMP2表达水平高于下调HIF-1α表达组.共转染shHIF-1α和shTP53 INP1质粒后在MCF-7细胞中,VEGF和MMP2表达高于下调shHIF-1α组(P<0.05).结论:HIF-1α可能通过下调TP53INP1的表达促进乳腺癌细胞的迁移侵袭能力.
Objective: To determine the expression of TAZ and its role in angiogenesis in gastric carcinoma. Methods: Immunohistochemical staining was performed to investigate the expression of TAZ and to determine whether a direct relationship exists between TAZ and β-catenin. Transfection with TAZ overexpression plasmid in MKN28 cells was conducted to induce exogenous expression of TAZ and a TAZ knockdown plasmid was transfected into MGC803 cells to reduce TAZ levels. The effects on endothelial cell formation, proliferation, and migration were determined by Matrigel three-dimensional culture, MTT proliferation assay and Transwell migration assay. In addition, the expression of TAZ and β-catenin in transfected gastric cancer cells was detected by Western blot. Results: Immunohistochemistry showed that TAZ protein was expressed in 64 of 150 gastric cancer sample tissues (43%), TAZ was localized in the nucleus, and its expression was associated with tumor grade, TNM stage, metastasis, and microvessel density (MVD) (P<0.05). In addition, the expression frequency of β-catenin in the TAZ positive group was 67.2%, which was significantly higher than that in the TAZ negative group, and the expression of TAZ was positively correlated with β-catenin. After transfection, TAZ overexpression increased the expression of β-catenin and enhanced HUVECs tube formation, proliferation, and migration. In the MGC803 cells transfected with the knockdown plasmid, β-catenin levels were decreased and HUVECs motility was inhibited. Conclusions: TAZ may promote angiogenesis in gastric cancer by promoting β-catenin expression.
Objective:To investigate the effect and mechanism of Jiedu Quyu prescription on drug resistance reversal for MCF-7/ADM human breast cancer cells.Method:The 3-(4,5-dimethylthiazol-2-yl)-2,5-diphenyltetrazolium bromide (MTT) was performed to test the effect of Jiedu Quyu prescription on proliferation of MCF-7/ADM cells.Flow cytometry was used to detect the concentration of intracellular rhodamine (Rh)-123,Real-time PCR and Western blot were used to analyze the expression of genes and proteins,including multiple drug resistant 1 (MDR1) and breast cancer resistant protein (BCRP).Result:After treatment with 1.25,2.50 g·L-1 Jiedu Quyu prescription,the reverse fold (RF) of MCF-7/ADM was 1.7 and 3.0 respectively (P < 0.05);the concentration of intracellular Rh-123 was enhance for 1.8 and 2.5 times respectively as compared with control group (P < 0.05).The protein and mRNA expression levels of MDR1 and BCRP were down-regulated obviously (P < 0.05);MDR1 mRNA expression level was decreased by 35.5% and 56.0% respectively (P < 0.05),while BCRP mRNA expression level was decreased by 41.6% and 49.5% respectively (P < 0.05).Conclusion:Jiedu Quyu prescription could enhance the sensitivity of human breast cancer cell line MCF-7/ADM to adriamycin,and reverse drug resistance of MCF-7/ADM.Its mechanism may be correlated with down-regulation of MDR1 and BCRP,as well as inhibition of intracellular drug efflux.
Objective To discuss observe the clinical effect of yanshu parenteral solution adjuvant therapy in the early metaphase endometrial carcinoma.Methods Chose 54 patients whose endometrial pathology indicated the early metaphase endometrial carcinoma,One group used conventional therapy,the other combined conventional therapy with yanshu parenteral solution.Results The near future effect rate and the remission rate of the observe group is 18.52%,51.85% respectively,which better than the control group,and the adverse reaction is few.Conclusion Applying yanshu parenteral solution to the early metaphase endometrial carcinoma as a adjuvant therapy had a better clinical effect.
