Background. The physiological regulation of the uterine contractile activity changes with age, which leads to an increased number of prolonged labor and emergency caesarean sections in women giving birth at the age of 35+. One of the modern approaches to correct the function of the reproductive system is the use of from cell cultures. CM from glial cell culture contains neurotrophic factors that play an important role in maintaining the contractile function of the uterus. Current cell culture technologies include cryopreservation. Objective: to research experimentally the effect of CM obtained from intact and cryopreserved cultures of glial cells on the contractile activity of the uterus in rats of different reproductive ages. Materials and methods. The monolayer cell culture was obtained from the dorsal root ganglia of neonatal piglets and cryopreserved in the presence of cryoprotectant dimethyl sulfoxide. CM from native and cryopreserved cultures were collected for 28 days, after which fractions with a molecular weight of < 30 kDa were obtained from them by ultrafiltration. Rats at the age of 6 and 14 months, which corresponds to reproductive age and late reproductive age (LRA), were intraperitoneally injected with 0.2 ml of media from intact (ICM) or cryopreserved (CCM) cultures for 9 days. On the 30th – 32nd day after the end of the administration of CM animals were slaughtered and the uterine contractile activity was determined by the organ bath method, the relative area of myometrium and density of myocytes by histological method, the average area of labeling with specific antibodies to smooth muscle actin by immunohistochemical method. The statistical significance of differences was assessed by the Mann–Whitney test. Results. It was found that spontaneous, OT-, and KCl-induced tension of isometric contraction of the uterus in intact LRA rats decreased by 19, 20, and 14%, respectively, compared with intact reproductive aged animals. After the introduction of ICM and CCM in LRA animals, normalization of isometric contraction parameters was observed. This effect was realized against the background of an increase in the area of the myometrium, the density of myocytes, and actin expression. Conclusions. Intra-abdominal administration of CM from glial cell culture increases the uterine contractile activity in LRA rats. This effect is realized by increasing the relative area of the myometrium, the density of myocytes, and the area of expression of smooth muscle actin. The effect of media from intact and cryopreserved cultures on the contractile activity of the uterus was similar, which makes it possible to use low-temperature culture storage technologies to obtain CM without losing its biological effect.
The aim. The article is devoted to the methods used in combustiology practice to assess the condition of patients and predict general and local complications. Results. With severe thermal trauma, there is a risk of developing life-threatening conditions in the victims, in particular, sepsis and multiple organ failure. Informative for assessing the course of burn disease is a modified scale of physiological disorders, which takes into account the syndrome of systemic inflammatory response, endogenous intoxication during periods of toxemia and septicotoxemia, and concomitant pathology. The development of intoxication in severe burn injuries requires determining the degree of severity of the process and its assessment in dynamics. The toxicity of blood plasma is studied by the resistance of red blood cell membranes in the acid hemolysis reaction, indicators of the red blood cell intoxication index, and the content of universal markers of intoxication, in particular, medium-weight molecules. Methods for biosensor indication of cytotoxic factors have also been developed. Assessment of the severity of the condition of victims with burns and monitoring the effectiveness of therapy is carried out taking into account the level of procalcitonin and presepsin. To determine the effect of the wound process on the formation of early sepsis in patients with burns in the capillary zone of thermal damage, the content of homocysteine, endothelin and nitric oxide is studied. To assess the severity of the condition of burn patients and predict the development of complications, the values of integral hematological indices are informative. Reliable data on the depth of a burn wound are obtained using thermotopometry and pH measurement of burn wounds, the method of magnetic resonance imaging. In order to assess violations and dynamics of blood supply restoration in the area of thermal damage, the method of laser Doppler flowmetry and optical tissue oximetry is used. Determination of interstitial pressure in burns of the extremities and intra-abdominal pressure in common burns is a way to predict the development of compartment syndrome. To monitor the patient's glycemic status, it is proposed to determine the level of glycosylated hemoglobin. Microbiological research data is used to predict the risk of developing general complications of burn disease, as well as – together with cytological data and methods for determining the maturity of granulation tissue – to assess the readiness of granulating wounds for autodermoplasty. Conclusion. The development of new effective ways to predict and prevent the development of complications in burns is promising
Всього в урбоценозах Харкова зареєстровано 66 видів стафілінід з 37 родів. Staphylinus caesareus та Drusilla canaliculata відзначені як еудомінанти, а 15 видів виявилися домінантами чи субдомінантами в більшості біотопів. Зростання числа видів відзначено з кінця квітня (12–15), і сягає максимуму в травні-червні (28–32 види). У липні– серпні відзначено по 10 та 16 видів відповідно, а восени – до 9 видів. Висока активність жуків характеризувалась одним–двома піками відзначеними з квітня до середини липня (але з максимумом в кінці травня). С другої половини літа чисельність стафілінід знижувалась, і досягала мінімуму восени. Перше збільшення чисельності жуків (кінець квітня-травень), обумовлено масовим виходом жуків, що перезимували. Другий пік чисельності (кінець червня-середина липня чи серпень) пов'язано як з появою жуків нового покоління, так і міграціями ряду видів стафілінід (особливо в літній період). Розглянуті сезонні зміни чисельності стафілінід у різних урбоценозах: парках, лісопарку, насадженнях центру та околиць міста і на присадибних ділянках. Висока чисельність стафілінід в центрі міста відзначена з квітня-травня (максимум) до початку червня. У парках та в Лісопарку, пік активності стафілінід спостерігали з другої половини квітня до середини травня, а значне зниження –з кінця червня до початку-середини серпня. Максимум чисельності Staphylinidae в насадженнях околиць міста характеризувалося одним-двома пікамим – з середини травня до червня та в першій половині червня, а різке зниження відзначено з середини літа. На присадибних ділянках пік чисельності в спостерігали у другій половині травня (2018) та у другій половині квітня (2019), після чого активність Staphylinidae істотно знижувалась. Можна припустити, що підвищення чисельності стафілінід весною (кінець квітня-травень) у всіх біотопах обумовлено виходом, активним пошуком їжі і розмноженням особин, що перезимували (в першу чергу Staphylinus caesareus), а в ряді випадків (в середині червня) і появою жуків нового покоління (наприклад, для Drusilla canaliculata). В подальшому, зниження активності, обумовлено смертністю більшості жуків після відкладання яєць, міграціями в інші біотопи в зв'язку з погіршенням умов, наприклад, низькою вологості ґрунту і підвищенням температури влітку. Максимальні показники чисельності (на прикладі S. саesareus) навесні зареєстровано на півтора-два місяці раніше, ніж такі статевого індексу. Максимальна частка самиць відзначена з другої половини літа і до початку осені.
Species composition, the ecological structure and the occurrence characteristics of weevils (Curculionoidea) in the stratobios of the main urban habitats of Kharkiv were studied. 59 species from 41 genera and three families were registered. Curculionoidea comprises 31.5% of the coleopterofauna of stratobios. Approximately 10 species are attributed to dominants (2–3 species are eudominants). 40 species are ranked as random and 9 species are rare. 4 species of Curculionoidea (Cyanapion columbinum, Curculio rubidus, Otiorhynchus albidus, Tropiphorus micans) are recorded for the first time in the Left bank forest-steppe zone of Ukraine. The values of the main indices of species diversity were low, which may indicate a significant oligodominance of Curculionoidea in all urban habitats. Most of species are herba- and dendrobionts (14–17), but almost all of them are registered as random (27) or rare (4) elements. 26 species associated with stratobios (among which most dominants elements are recorded). Species associated with meadows (21) and forests, or eurytopic species (14) are dominants in the samples; by trophic links – oligo- (32) and polyphagous species (24 ) were prevailed. 45 species prefer mesophilic habitats. The number of eurytopic mesophilous polyphagous stratogeobionts were maximal. The largest number of species (25–33) were documented in vegetation of the outskirts and household plots of city compared to the city parks and plantings of the center (18–19), but minimumly (12) in forest. The spectrum of ecological groups was also minimal in a forest, but the maximum is in the plantings of periphery and household plots of the city. Faunistic similarity was slightly more than 0.20, which may indicate specific and significant differences in the species composition of weevils in different urban habitats of Kharkiv.
During hundreds of years, in large cities man has been forming a specific urban environment with original species composition of insect communities, including the most diverse group of predatory beetles – Staphylinidae family. Kyiv, Kharkiv, Dnipro are the three most populated cities of Ukraine. In the urban cenoses of these cities, over 140 species from 66 genera of Staphylinidae have been recorded. The total of 69 species (43 genera) were recorded in Kyiv, 67 (39 genera) in Dnipro and 66 (37 genera) in Kharkiv. Among them, , eight species in the catalogue of Palearctic staphylinds had not been previously recorded for Ukraine: Arpedium quadrum Grav., Atheta laticeps Thomson, Medon apicalis Kraatz, Ocalea rivularis Müll., Philonthus salinus Kiesenwetter, Quedius invreae Gridelli, Tasgius pedator Grav. and Xantholinus gallicus Coiffait. By number, common species accounted for 29 in Dnipro, 21 in Kyiv and 19 in Kharkiv. In all the metropolises, two species (Staphylinus caesareus Cederhjelm and Drusilla canaliculata (F.)) were identified as eudominants. Dominants and subdominants comprised 18–25 species. Almost two thirds of the fauna of staphylinids of the cities was classified as rare species. The lowest faunistic similarity was seen between the staphylinids of the urban cenoses of Dnipro and those in Kyiv and Kharkiv (15.3% and 17.5%), similarity was higher for Staphylinidae of Kyiv and Kharkiv (36.0%). Similarity by common species of staphylinids for Kyiv and Kharkiv equaled 73.9%, 28.2% for Dnipro and Kyiv and 37.1% for Dnipro and Kharkiv. The article offers a review of species diversity, number and ecological structure (biotopic confinedness, hygrothermal preference) of typical species of staphylinids in different urban cenoses of Kharkiv. Differences in qualitative-quantitative and ecological structures of staphylinids are related to the diversity of the conditions in a particular urban cenoses. The commonest representatives of the family in the metropolises were polytopic mesophilous carnivorous species.
To date, about a 120 hundred clown beetle species from 32 genera are known from Ukraine. The validity of a species new to science ( Eudiplister ukrainicus Pozharov, 2011) described from the agroecosystems of Poltava region require verification. Based on personal and literature-derived data, 39 species of clown beetles have been registered in the transformed ecosystems of Ukraine. Of these, 12 species in six genera are known from Kharkiv City: Margarinotus (4 species), Hister (3 species), Atholus (2 species), and Gnathoncus, Hypocacculus , Saprinus with one species each. Individual abundance of the clown beetles in the Kharkiv urban coenoses averaged 0.2% out of the total number of collected ground-dwelling beetles. Only one species, Margarinotus purpurascens Herbst, 1792, reached the status of subdominant in some habitats, the other species being rare. Eight species were found at the garden-plots, and seven species in the city parks (six species were recorded from the Karpovskyi Sad, the other parks hosted three or four species each). Central and suburban loans were inhabited by three species only. In terms of habitat distribution, three species of collected clown beetles belonged to the group of meadow dwellers, three species to the steppe dwellers, and six species were classified as generalists. In terms of station preferences, we distinguished six copro-necrobiont species, four coprobionts, one botrobiont ( Gnathoncus suturifer ), and one stratobiont ( Margarinotus ventralis ) inhabiting decaying plant and fungi residues. Faunisitc similarity of the histerid assemblages of the urban ecosystems of Kharkiv and field agroecosystems of Forest-steppe zone of Ukraine is high. In our collection, eleven species out of twelve occurred in both city and fields, M. ventralis was the only species found exclusively in the city. This suggests that the ways of the fauna formation in the above-mentioned transformed ecosystems has been identical. Therefore, we suppose some species known from the agroecosystems ( Hister bissexstriatus Fabricius, 1801, H. helluo Truqui, 1852, H. quadrimaculatus Linnaeus, 1758, H. unicolor Linnaeus, 1758, M . carbonarius Hoffmann, 1803, Eudiplister planulus Menetries, 1848 and S. tenuistriatus Marseul, 1862) to be found in the urban areas as well. A total of 320 species from 42 families were registered in the herpetobios (stratobiont and epigeobiont beetles) of the urbocenosis of Kharkiv. Among these more than 160 species are recorded for the first time for this metropolis. Four species of ground beetles (Notiophilus germinyi Fauvel, Clivina fossor L., Laemostenus terricola Herbst and Masoreus wetterhali Gyll.) were not listed for Ukraine in the Catalogue of Palearctic Coleoptera. The species Omosita japonica Reitter, 1874 (Nitidulidae) is new for Ukranian fauna. Moreover, 93 species from 17 families are the dominant species (at least in one of the urban ecosystems). Among these, eudominants are represented with almost 20 species, but the number of eudomonants did not exceed ten species in separate urban cenoses. Carabidae (80), Staphylinidae (36), Curculionidae (32) and Tenebrionidae (8 species) dominate both, in species composition and abundance. The total part of these groups reached at least 90% of Coleoptera, recorded in urban-cenosis. Separate species of Silphidae, Dermestidae, Elateridae, Chrysomelidae, Leiodidae, Coccinellidae, Nitidulidae, Scarabaeidae, Histeridae and Lucanidae were sporadically usual in most plots. They accounted almost 9% of the total quantity of all beetles in transformed cenoses. Among these, a greater number of species are found within the families of Scarabaeidae (33), Chrysomelidae (16), Elateridae (14), Histeridae (12), Coccinellidae (11) and Nitidulidae (11), Silphidae (6) and Dermestidae (6). The rest of the families are represented with 1–4 species. All beetles registered in herpetobios belong to three groups. Typical inhabitants of herpetobios prevailed (about 200 species from 22 families, at least 90% of all Coleoptera). The proportion of species sporadically associated with herpetobios (more than 40 species from 10 families) was about 5% of all beetles. Random components of herpetobios are represented by more than 70 species from 10 families, and their total share did not exceed 3% of all Coleoptera. A preliminary taxonomic review of coleopterofauna was carried out in different urbocenoses. The number of species in urbocenosises ranged from 101 (Forest Park) to 124 (urban parks). In other green plantings of the city, the species diversity reached 105–118 species. The number of dominant beetles in different areas ranged from 37 to 60 species.
A total of 54 species in 35 genera of Curculionidae are registered in the urbocenoses of Kharkiv city. Only three species, Exomias pellucidus, Otiorhynchus raucus and Ot. ovatus, are recorded as dominants. Some other species (Urometopus nemorum, Sciaphobus squalidus and Glocianus punctiger) are sporadically noted in some sites as subdominants. The total number of species turned out to be higher in the herpetobios of plantings at periphery (31), slightly lower in private farmlands of the city (21), parks and plantings of the center (17 each), but minimal in suburban forests (11 species). The highest quantity of specimens were observed for the plantations of the center and margins of city: nevertheless, minimal quantity of specimens was registered at parks and woods. The maximum number of species (including all cenoses) was recorded from the end of April to first decade of May (28 species). From May to the end of June 15–18 species were registered. Ten species were recorded at July, and only six species at August. A slight rise was noted from the end of August to mid-September (11 species). In biotopes where Exomias pellucidus appeared to be the monodominant, the seasonal dynamic density of weevils reached a peak at late May or early June, but the significant decline was observed from early June to July. At the sites with several dominants, two or three peaks were registered: at spring (May) and at mid-summer (end of June or beginning of July). Furthermore, the number of weevils decreased gradually, but some species of Curculionidae were rather abundant even from the end of summer to September–October. The spring increase is reasoned by high density of species in the genus Otiorhynchus; the peak in early summer (maximum) is caused by the increased activity of majority of dominant species; the autumn peak is also caused by the high number of species of the genus Otiorhynchus. The level of sex index (by the example of E. pellucidus) differed significantly at all plots. It was higher for the plantations at the periphery of the city (0.70), and it was minimal at private farmlands (0.20). Relative conjugacy of sex index and dynamic density were not observed: the maximum abundances of quantity of Curculionidae were recorded 20–30 days earlier than the maximal numbers of the sex index.
This article presents a general overview of the superfamily Scarabaeoidea in five parks of Kharkov city and some adjacent agrocenosises. The species composition includes 32 species from four families of Scarabaeoidea. The Scarabaeidae family dominated in biodiversity and abundance (26 species from 16 genera). Six species (Dorcus parallelopipedus, Platyderes caraboides, Anoplotrupes stercorosus, Aphodius melanostictus, Pleurophorus caesus and Onthophagus coenobita) were sporadically registered as common, but in separate urban cenosises the number of species did not exceed one or three. The species composition and abundance of lamellicorn beetles was higher in large parks with only slight recreational disturbance. Here from 13 to 20 species were registered. On lawns in the center and in other districts of the city Scarabaeoidea species met very rarely. In the agrocenosises on the outskirts of Kharkov city – 16 species are noted, almost half of which were registered as common. The faunistic similarity of different urban cenosises was at low to medium levels (an average 0.20–0.47). These indices were slightly higher (0.38–0.67) for areas with lower recreational pressure. The indices of faunal similarity of separate parks and agrocenoses were lower (0.18–0.56, but on average – 0.33). At the level of common species, there was no faunistic similarity. The significance of such differences was due to both the low number of most Lamellicorn species and some of their ecological characteristics. The main differences were observed in the ratio of trophic groups and features of the spatial distribution of the dominant species. Smaller differences were observed on biotopic characteristic of most species. There were no differences in the hygropreference of the scarabeid species. In the parks herpetobiont saprophagous species dominated (mainly coprophagous and dendrophagous). In the agrocenoses, dendrochordobiontic phytophagous species predominated. The peculiarities of the ratio of different ecological groups of Scarabaeoidea in urban and agrocenosises are discussed.
