目的:观察滋肾填精中药对肾阴虚型体外受精-胚胎移植(in vitro fertilization-embryo transfer,IVF-ET)患者子宫内膜同源框基因10(Hox 10)mRNA表达的影响,从基因层面探讨滋肾填精法对子宫内膜容受性的作用机理.方法:收集120例进入IVF-ET周期的不孕症患者,其中80例为肾阴虚型患者,随机分为观察组40例和对照组40例,另外40例单纯因输卵管因素者为空白组.3组患者均采用常规长方案进行超促排卵治疗,观察组在此基础上于降调节前1周期的月经第3~21天、降调节当月月经第3~14天及降调节后月经第3天服用二至天癸颗粒至取卵日.于降调节当天(监测排卵后第7天)采用RT-PCR法检测注射人绒毛膜促性腺性激素(human chorionic gonadotropin,HCG)日3组患者子宫内膜中Hox 10 mRNA的表达;比较观察组、对照组患者中医证候积分变化情况,评定3组患者注射HCG日子宫内膜厚度及分型、子宫内膜容积、子宫动脉搏动指数(pulsatility index,PI)和阻力指数(resistance index,RI)、血清激素水平及临床妊娠率等.结果:1)观察组肾阴虚证候积分低于对照组(P<0.05),肾阴虚症状改善.2)HCG日子宫内膜厚度、容积观察组高于对照组(P<0.05),与空白组比较差异无统计学意义(P>0.05),对照组低于空白组(P<0.05).3)子宫内膜分型中A型+B型比例观察组高于对照组(P<0.01),略高于空白组,但差异不明显(P>0.05),对照组低于空白组(P<0.05).4)观察组HCG日PI和RI均低于对照组(P<0.05),与空白组相比PI无明显差异,与对照组相比空白组PI和RI均降低(P<0.05).5)3组患者HCG日血清雌二醇(estradiol,E2)、血清黄体生成素(luteinizing hormone,LH)及孕酮(progesterone,P)相比差异无统计学意义(P>0.05).6)临床妊娠率观察组高于对照组(P<0.05),与空白组比较差异无统计学意义(P>0.05),对照组低于空白组(P<0.05).7)3组患者HCG日子宫内膜中Hox 10 mRNA均有表达,空白组表达最强,与观察组比较差异无统计学意义(P>0.05);对照组表达最弱,与观察组及空白组比较差异有统计学意义(P<0.05).结论:滋肾填精法可有效提高子宫内膜容受性,提升不孕患者临床妊娠率,其机理可能与增强子宫内膜中Hox 10 mRNA表达有关.
目的 观察瑞舒伐他汀对ApoE-/-小鼠动脉粥样硬化斑块凋亡相关蛋白Bcl-2和Caspase-3表达的影响. 方法 10只C57小鼠作为对照组,给予普通饮食,同品系22只ApoE-/-小鼠随机分为模型组(n=11)、干预组(n=11),高脂喂养8 w后,干预组给予瑞舒伐他汀悬液灌胃,模型组给予等量生理盐水. 继续喂养12 w后所有老鼠处死,全自动生化仪检测血清血脂,光学显微镜观察各组小鼠主动脉斑块形态及面积,免疫组化法检测动脉粥样斑块Bcl-2和Caspase-3蛋白的表达. 结果 与对照组相比较,模型组总胆固醇( TC)、甘油三酯( TG)、低密度脂蛋白胆固醇( LDL-C)明显升高(P<0.05),高密度脂蛋白胆固醇(HDL-C)明显降低(P<0.05);主动脉可见明显粥样斑块;Bcl-2 表达明显减少,Caspase-3 表达明显增多(P<0.05). 与模型组比较,TC、TG、LDL-C明显降低(P<0.05);干预组小鼠动脉粥样斑块面积减小(P<0.05), Bcl-2 表达明显增多,Caspase-3 表达明显减少(P<0.05). 结论 高脂饮食可引起ApoE-/-小鼠主动脉凋亡相关蛋白Bcl-2和Caspase-3的变化,瑞舒伐他汀可以通过上调Bcl-2、下调 Caspase-3的表达,抑制ApoE-/-小鼠动脉粥样硬化斑块的进展.
Objective Serine protease inhibitor member 3 of clade A (SERPINA3) gene on chromosome 14o32. 1 has been showed to be associated with intracranial aneurysms in a Polish population. In this study, the purpose was to test and analyze the polymorphism of the SERPINA3 in intracranial aneurysms in Chinese Hah nationality. Method To compare the genotype frequency and A allele frequency of rs4934polymorphism of the SERPINA3 in 275 patients with intracranial aneurysms and 361 controls by polymerase chain reaction and Taqman MGB probe analysis. Results The GA + AA genotype frequency of rs4934 polymorphism of SERPINA3 of aneurysm group (85. 1%)was no much higher than that of control group (81.44%) (χ2 = 1.475,P = 0.224). The A allele frequency ofaneurysm group (58.55%) was also no higher than the control group (59. 14%)(χ2 = 1.445, P = 0. 229). Adjustment for age, sex using logistic regression did not change the results (χ2 = 1. 499, P = 0. 221 ). Conclusions There is no significant difference of the rs4934 polymorphism of SERPINA3 between intracranial aneurysms and controls among individuals of Chinese Han nationality.
Purpose To determinate the vasoactive intestinal peptide in intestine of chicken and to compare the extraction technologies.Methods Duodenum,jejunum,and ileum segment of fresh intestine of chicken were first defatted,and then cleaned with water.ELISA method was used to detect the content of vasoactive intestinal peptides respectively after they were extracted from the processed intestinal segments(20.0 g respectively) by different methods.Results The content of vasoactive intestinal peptide in duodenum,jejunum,and ileum segment was 25.53,25.01 and 23.27 ng/g respectively.The content is higher by the extraction in hot water and vasoactive intestinal peptide is destroyed in acidic and hot environment.Conclusion Vasoactive intestinal peptide is rich in intestine of chicken.
Objective To test and analyze the polymorphism of the Type Ⅲ Collagen alpha1 gene in intracranial aneurysms. Methods To compare the genotype frequency and A allele frequency of the COL3A1 + 2209A/G polymorphism in 243 patients with intracranial aneurysms and 260 controls by polymerase chain reaction-restriction fragment length polymorphism (PCR-RFLP) method. Results The GA + AA genotype frequency of ancurysm group (41.56%) was much higher than that of the control group (28.08%) (χ2=10.15, P=0.001). The A allele frequency of aneurysm group (23.66%) was also higher than that of the control group (18.46 %) (χ2=9.13, P=0.003). Adjustment for age, sex, hypertension, smoking and alcohol consumption using logistic regression did not change the conclusions(χ2 = 6.76, P = 0. 009). Conclusions There is significant of difference of COL3A1 + 2209A/G polymorphism between intracranial aneurysms and controls.
Objective To study the possible role of IgG in th e mechanism of resorption or disappearance of the herniated interverteb ral disc.Methods Twenty-five adult dogs were used.The L 2~3 autologous nucleus pulposus were au tografted into the epidural space at L 5 S 1 .The dogs were divided into 5groups,and were sacrificed at 2,4,6,8and 12week s after surgery,respectively.The s pecimens were observed and analysed immunohistochemically fo r the presence of IgG.As control,the normal nucleus pulposus were harvested from the same dog and same immunohistochemical detections we re performed.Results(1)At different weeks after surgery,th e various disappearance and contraction of the nucleus pulposus were observed i n each group.(2)IgG were presented ,which tended to i ncrease as time progressed.(3)The trivial IgG were observed in the control specime ns.Conclusion Herniated nucleus pulposus can induce the infiltration of IgG immunization,which suggests that IgG play an important r ole in the early phase of the resorption of the herniated nucleus pulposus.And it may enhance the re-sorption process of the herniated di sc.This finding may provide histolo gic evidences in the conservative tr eatment on some type of LDH in the patients.
