Huntington's disease is an autosomal dominant neurodegenerative disorder caused by trinucleotide repeats expansion mutation in the first exon of HTT gene. Neurodegenerative disorders are difficult to study due to limited material availability and late onset of the manifestation. In vitro cell models provide a good opportunity to study molecular mechanisms of the pathology. New genome editing approaches allow us to generate isogenic cell lines that can be useful for drug screening and studying mechanisms of molecular and cellular events triggered by certain mutation on an equal genetic background. The aim of our work was establishing a method to introduce expanded CAG repeat tract into HTT gene by homology directed repair. We used a genome editing tool based on CRISPR/Cas9 system to facilitate the process. A donor construct bearing 215 CAG trinucleotide repeats flanked with long homology arms was generated. The method was approved on the HEK293Phoenix cell line. Mutant clones were obtained. It was shown that mutant HTT protein is expressed in the HEK293 mutant clones and affect their viability and proliferation rate.

The expression of genes Sox2, Klf4, Myc, Sall4, Gata6, Foxa2, Hnf4a, Cdx2, Esrrb, Hand1 in cell lines, embryos and organs of adult voles Microtus rossiaemeridionalis was studied. High resemblance of the expression patterns of these genes in the organs of adult voles, mice and humans was demonstrated. It was established that genes Gata6, Foxa2 and Hnf4a were specifically expressed in vole extraembryonic endoderm cells, while Cdx2 and Hand1 genes, in trophoblast stem cells. This shows that these genes can be used as markers of corresponding vole cell lines. Indirect confirmation pointing to the fact that Oct4 gene is a marker gene for epiblast cells both in the vole and mouse was obtained.

Изучена экспрессия генов Sox2, Klf4, Myc, Sall4, Gata6, Foxa2, Hnf4a, Cdx2, Esrrb, Hand1 в культивируемых клетках, эмбрионах и органах взрослых животных полевки Microtus rossiaemeridionalis. Показано высокое сходство паттерна экспрессии этих генов в органах взрослых особей у полевок, мыши и человека. Установлено, что гены Gata6, Foxa2 и Hnf4a специфически экспрессируются в клетках экстраэмбриональной энтодермы, а гены Cdx2 и Hand1 – в трофобластных стволовых клетках полевки, что дает основание в дальнейшем использовать эти гены как маркерные для соответствующих клеточных линий полевки. Получено косвенное подтверждение того, что у полевки, как и у мыши, ген Oct4 является маркерным для клеток эпибласта.

A pool of sudden death cases was analyzed, 68 cases with dysplasia of connecting tissue and pathology of cardiac valvular apparatus were revealed. Types valvular apparatus pathology were determined which could lead to the development of acute left ventricular failure. The assumption concerning ways of development of acute heart failure in this pool is done. Defects and pathologic forms of valves were established to have essential value for genesis of sudden death in persons of young age.
