目的 探讨运脾祛湿泄浊方治疗非酒精性脂肪性肝病(NAFLD)的临床疗效.方法 选取杭州市中医院2018年6月至2020年10月收治的非酒精性脂肪性肝病患者61例,按照随机数字表法分为观察组(32例)与对照组(29例).对照组予多烯磷脂酰胆碱胶囊(易善复)胶囊治疗,观察组在对照组基础上加用运脾祛湿泄浊方治疗.比较两组治疗前后症状改善积分、肝功能指标,以及临床疗效、不良反应情况.结果 两组患者经治疗后的临床症状积分均较治疗前降低(P<0.01);治疗后与对照组相比,观察组的临床症状积分显著降低(P<0.01);治疗后观察组血清丙氨酸转氨酶(ALT)、直接胆红素(DBIL)、甘油三酯(TG)、低密度脂蛋白(LDL)指标较治疗前明显降低(P<0.05).结论 运脾祛湿泄浊方治疗非酒精性脂肪性肝病有较好疗效,改善肝功能、降血脂效果显著,且安全性良好.
目的:研究丹酚酸A对棕榈酸诱导的A ML12肝细胞脂毒性的保护作用并初步探究其分子机制.方法:采用棕榈酸诱导A ML12细胞建立脂毒性模型并给予丹酚酸A进行干预,采用乳酸脱氢酶(LDH)法检测细胞损伤,采用酶法检测细胞内甘油三酯含量,采用Bodipy染色法观察胞内的脂滴,采用甲基偶氮唑盐微量酶反应比色法(MTT)检测细胞存活率,采用荧光细胞通透性染料罗丹明123和荧光显微镜检测线粒体膜电位.采用2',7'-二氯荧光黄双乙酸盐(DCFH-DA)和荧光显微镜检测细胞内活性氧(ROS)水平.采用Western blot技术检测沉默信息调节因子相关酶Ⅰ(silent information regulator 1,SIRT1)及腺苷酸活化蛋白激酶(AMP-activated protein kinase,AMPK)蛋白表达.结果:棕榈酸作为肝细胞脂毒性诱导剂可显著降低A ML12细胞存活率,加重细胞内脂质沉积,而丹酚酸A干预可改善棕榈酸诱导的肝细胞脂毒性损伤(P<0.05).其次,丹酚酸A干预可改善棕榈酸诱导的细胞线粒体膜电位降低(P<0.01)及细胞内ROS水平升高(P<0.01).此外,Western blot结果显示棕榈酸可显著抑制AMPK及SIRT1蛋白表达(P<0.05),而经过丹酚酸A处理可显著升高棕榈酸抑制的SIRT1及AMPK磷酸化水平(P<0.05).结论:丹酚酸A可有效改善脂毒性诱导的肝细胞损伤,该保护作用可能与其激活AMPK及SIRT1蛋白有关.
目的:研究丹酚酸A对脂毒性诱导的H9C2心肌细胞损伤的改善作用并初步探究其分子机制.方法:采用棕榈酸体外诱导建立H9C2心肌细胞脂毒性模型并给予丹酚酸A进行干预,采用乳酸脱氢酶法检测细胞损伤,采用细胞增殖-毒性检测试剂盒检测细胞存活率,采用罗丹明123染色观察心肌细胞线粒体膜电位变化,采用蛋白免疫印迹技术研究丹酚酸A改善作用的分子机制.结果:浓度为400μmol/L的棕榈酸可显著导致H9C2心肌细胞脂毒性损伤(P<0.05).不同浓度(10、20、40、80μmol/L)丹酚酸A暴露对心肌细胞无毒性作用(P>0.05).丹酚酸A干预显著改善脂毒性诱导的心肌细胞损伤及细胞线粒体膜电位降低(P<0.05).激活Toll样受体4(Toll-like re-ceptors,TLR4)可显著增强脂毒性诱导的心肌细胞损伤(P<0.05),而抑制TLR4显著减轻棕榈酸诱导的细胞脂毒性(P<0.05).此外,丹酚酸A显著抑制棕榈酸诱导的TLR4及其下游c-Jun氨基末端激酶(c-Jun N-terminal kinase,JNK MAPK)(P<0.05).结论:丹酚酸A改善脂毒性诱导的心肌细胞损伤,该保护作用可能与其抑制TLR4/JNK MA P K信号通路有关.
目的 探讨丹酚酸A(Sal A)通过诱导白色脂肪组织褐变增加动物体内促进能量消耗从而改善肥胖的作用研究.方法 腹腔注射Sal A对高脂喂饲的C57BL/6小鼠HFD模型;HE染色检测小鼠附睾脂肪的病理变化;Real-time PCR、Western Blot等分析脂肪组织中棕色脂肪细胞特异性基因/蛋白表达水平.结果 Sal A减轻HFD诱导的小鼠体重增加和附睾脂肪中的脂质积累,并显著上调附睾脂肪组织中UCP1和PGC-1α的基因及蛋白表达.结论 Sal A通过诱导脂肪组织褐变改善小鼠肥胖,对Sal A治疗肥胖相关疾病具有潜在的意义.
在世界范围内,胃癌是发生率和癌症相关死亡的第三常见原因的第四常见癌症[1].Chen等[2]报道,2015年中国胃癌发病率为679/100,000,病死率为498/100000.目前随着生活环境和营养状况的改善,以及我国及世界范围内医疗水平逐年上升,胃癌的发病率逐年下降.然而印戒细胞癌(SRCC)在胃癌中比例却正在逐年增加[3].本资料对现有研究文献进行荟萃分析以评估SRCC患者的预后以及淋巴结转移(LNM)情况,将有助于临床医生更清晰地了解SRCC患者,并为其选择更有利的治疗方案.
Objective: To compare the effects of generic (Aisuping()) and original intravenous esomeprazole (Nexiumumumumum()) on gasctric acid secretion. Methods: Ninety-three SD rats were randomly divided into six groups, including Aisuping() Low-dose group ( 10 mg·kg-1·d-1), Aisuping high-dose() Group ( 50 mg·kg-1·d-1), Nexium() Low-dose group ( 10 mg·kg-1·d-1), Nexium() High-dose group ( 50 mg·kg-1·d-1), saline group and blank group.Then, the secretion levels of gastric acid, pepsin and gastrin were detected on day 5 and day 14 after the multiple dose administration. Results: There was no significant difference in the secretion levels of gastric acid, pepsin and gastrin between Aisuping() And Nexium() Groups on day 5 and day 14. Both Aisuping() And Nexium() Could effectively inhibit gastric acid secretion in rats. Conclusion: Generic esomeprazole for injection has long and persistent gastric acid inhibitory effect in rats, which is equivalent to original esomeprazole.
Objective] The effect of Danshen Injection on the inhibition of 4-Hydroxynonenal in a mouse model of alcoholic liver disease(ALD). [Methods] Mice were fed ethanol-containing Lieber-DeCarli diet and injected Danshen.The levels of ALT and TG were measured by HE and relative kits. The expression of 4-hydroxynonenal(4-HNE), peroxisome proliferator activated receptors(PPARα), NF-κB in the liver was determined by Western Blot, respectively. [Resultes] ①Mice ALD model was built successfully. ②Danshen injection significantly reduced the level of ALT,TG(P<0.01) and inhibited hepatic lipid accumulation.③Compared with the Control Group, the expression of 4-HNE level and protein of the liver was significantly increased in ALD Group(P<0.01). Danshen Injection inhibited 4-HNE and activated PPARαand NF-κB (p65). [Conclusion] Supplementation of Danshen Injection, conferred a protective effect on ALD in mice, was associated with active hepatic PPARα, and decreased hepatic 4-HNE accumulation, and improved hepatic NF-κB activity.
To investigate the molecular characteristics and variation of newly isolated infectious bronchitis virus(IBV), the gene2 to gene6 genomic sequences of IBV WF2011 isolated from ovarian tissues of laying hens in Weifang of Shandong were sequenced. The sequence analysis showed that the C terminal sequence of WF2011 had the typical 5'-S-3a-3b-E-M-5a-5b-N-3' gene order and the typical avian coronavirus cleavage site of540RRSRR544in spike protein, as American strains of Gray, Holte, JMK, Conn46 and South Korean SNU8067 strain. Homology analysis and phylogenetic analysis showed that WF2011 had low identities with IBV reference strains. The identities of WF2011 S1 gene with those of other reference strains range from 61.8% to 86.0%. There is relatively far genetic distance between WF2011 and LX4, SAIBK, Mass-like strains. Therefore, WF2011 might be a new variant IBV isolate.
