Болезнь, вызванная вирусом Эбола (БВВЭ) — одно из самых высоколетальных вирусных заболеваний, поражающих человека и приматов. Возбудителем БВВЭ является вирус Эбола. В настоящее время известно шесть видов этого вируса, три из них патогенны для человека — это виды Заир (ZEBOV), Судан (SUDV) и Бундибугио (BDBV), вызывающие острые вирусные высоконтагиозные лихорадки у людей и приматов с летальностью до 90%. В большинстве случаев БВВЭ вызвана видом ZEBOV. Разработка вакцин против БВВЭ началась сразу после идентификации возбудителя в 1976 г. На сегодняшний день в мире зарегистрировано четыре вакцинных препарата для профилактики БВВЭ. Все они основаны на протективном антигене — гликопротеине (GP) вируса Эбола вида ZEBOV. В силу того, что виды SUDV и BDBV также могут быть причиной вспышек и эпидемий БВВЭ, очевидна необходимость разработки вакцин, способных обеспечить защиту от всех известных патогенных для человека видов вируса Эбола. В статье систематизированы данные относительно структуры, иммуногенных и протективных свойств GP вируса Эбола, проведен анализ иммунодоминантных эпитопов гликопротеина вирусов ZEBOV, SUDV и BDBV, необходимых для формирования протективного иммунитета, а также предложен рациональный, на наш взгляд, подход создания возможных вариантов вакцин против БВВЭ, вызванной разными видами вируса Эбола, состоящий в использовании векторных конструкций, экспрессирующих как минимум два варианта гликопротеина — GP вируса Эбола вида ZEBOV и вида SUDV.
Болезнь, вызванная вирусом Эбола (БВВЭ), - одно из опаснейших инфекционных заболеваний человека и животных. Впервые вспышка этого заболевания произошла в 1976 году в Судане и Заире. С тех пор было зафиксировано более 20 вспышек, самая крупная из которых в 2014-2016 годах переросла в эпидемию на территории Западной Африки и унесла жизни более 11000 человек. Хотя наиболее эффективным средством борьбы с эпидемиями является вакцинопрофилактика, к началу эпидемии БВВЭ в мире не было ни одной зарегистрированной и разрешенной к применению вакцины. Разработка первых вакцин против БВВЭ началась еще в 1980 году и прошла огромный технологический путь от инактивированных до генноинженерных вакцин на основе рекомбинантных вирусных векторов. Данный обзор посвящен векторным вакцинам против БВВЭ, которые показали наибольшую эффективность в доклинических исследованиях и в настоящее время находятся на разных этапах клинических исследований. Особое внимание уделено механизмам развития иммунного ответа, важным для защиты организма от БВВЭ, а также ключевым параметрам вакцин, необходимых для индукции длительного протективного иммунитета против БВВЭ.
Modern medicine now encounters with problem of the absence of effective antibacterial drugs, which are able to render therapeutic effect on chronic form of infectious process. Thus, the actual objective is to develop essentially new generation of drugs, on the basis of which should lie identification of new bacterial targets playing key role in process of chronization of infection as well as selection of new physiologically active substances, which are able to render highly specific inhibitory effect on selected target. Solving of this objective is possible during realization of new approaches for search and design of new drugs and, first of all, during usage of bioinformatics methods, which enable to identify new biotargets, select most effective chemical compounds-inhibitors and optimize their pharmacological and pharmacokinetic properties. The most promising bacterial target is secretion systems of pathogenic microorganisms participating in realization of their virulent characteristics and playing major role in transition of infectious process in chronic phase. We performed synthesis of and screening for 80 compounds, which allowed to select a range of inhibitors rendering specific target-directed effect on type 3 secretion system of Chlamydia. Obtained data allow to further assess of biological and therapeutic activity of these compounds on developed models of infectious process in vivo.
The wide use of antibiotics in livestock raising has contributed to the selection and accumulation of representatives of commensal microflora, as well as pathogenic bacteria, colonizing livestock and poultry. For this reason the problem of the possible transfer of antibiotic-resistance genes along the chain from bacteria, autochthonous for agricultural animals, to bacteria used for the production of foodstuffs, which are incorporated into normal microflora and may thus participate in the exchange of these genes with bacteria, enteropathogenic for humans, is a highly important task of medical microbiology. The article deals with the review of experimental data, indicative the possibility of the appearance of antibiotic-resistant pathogenic bacteria due to the transfer of antibiotic-resistance genes via alimentary chains.
In a study on the impact of chlamydial infection on host cell apoptosis, C. trachomatis were shown to protect host cell against staurosporin-induced apoptosis only at the middle stage of infection development (at 20 hours post infection), C. pneumoniae--at different stages of its growth cycle (from 2 to 7 day post infection). We found, that C. trachomatis elementary bodies fail to inhibit staurosporin-induced apoptotic stimuli. The clear antiapoptotic effect of cell lysate filtrate, infected with C. trachomatis, was demonstrated by cytometric analysis and luminescent microscopy. Our findings make it possible to use biochemical approach to identification of chlamydial antiapoptotic factors in future. Investigations directed at chlamydial antiapoptotic activities may aim to create the therapies of chronic chlamydial infection.
