Nicotiana tabacum leaves are mainly used in the production of cigarettes and cigars all over the world, and a large amount of waste materials will be produced in the process. High-value scopoletin can be extracted and separated from waste materials, but there are some problems, such as low purity of products and inability of large-scale production. In this study, we established an efficient and stable method for producing high-purity scopoletin from waste tobacco by containing acetone reflux extracting with dynamic axial compression-high performance liquid chromatography (DAC-HPLC) separating. In particular, the extraction system makes the scopoletin fully extracted through continuous reflux, the DAC-HPLC system makes the high-purity scopoletin repeatedly obtained, and the DAC-HPLC system was first used for scopoletin. 目的:考察家蚕蛹虫草(海宁株)子实体提取分离获得的麦角甾醇和虫草素联合抗肿瘤作用.方法:以家蚕蛹虫草(海宁株)子实体提取分离获得的麦角甾醇和虫草素为原料,开展联合给药(麦角甾醇:虫草素=3:2)的小鼠急性毒性试验,进一步构建lewis肺癌和H22肝癌荷瘤小鼠模型,以环磷酰胺作为阳性对照,分别建立模型组、试验组、空白组,考察联合给药对小鼠肺癌和肝癌的药效.结果:根据Bliss方法计算急性毒性试验数据,获得联合给药的LD50为3.98 g/kg.此外,药效试验结果表明,联合给药剂量在67~600 mg/kg内时,抑瘤率随给药剂量的升高而增强;当给药剂量为600 mg/kg时,与模型组相比,对小鼠肺癌和肝癌的抑制率最高,分别为50%和46%,其对小鼠Lewis肺癌模型的疗效优于环磷酰胺,然而,对小鼠H22肝癌肿瘤模型的效果与环磷酰胺无明显差异.结论:初步试验结果表明,在安全剂量范围内,家蚕蛹虫草(海宁株)子实体中麦角甾醇与虫草素联合给药具有抗肺癌肿瘤及肝癌肿瘤生长的作用,为肺癌和肝癌药物的开发及应用建立良好的基础.
